Dopamine Uptake Assay

Dopamine uptake assays are experimental methods that measure how efficiently cells or synaptic preparations remove dopamine from the extracellular environment, providing a functional readout of dopamine transporter (DAT) activity. In a typical neuroscience assay, cells or membrane preparations are exposed to a known dopamine concentration for a defined interval; transporter-mediated uptake is then quantified by measuring retained dopamine, often with a labeled substrate, while temperature, timing, and transporter inhibitors help distinguish specific uptake from nonspecific accumulation. These assays support characterization of DAT kinetics, comparison of transporter function across models, and evaluation of drugs or neurotoxicants that alter dopaminergic signaling, with implications for research on reward, movement, and neurological disease.

Measuring Dopamine Uptake in Mouse Brain Synaptosomes Using Radiolabeled Dopamine

2025

This video demonstrates the assessment of dopamine uptake in synaptosomes harvested from mouse brains. It outlines steps for preparing synaptosomes for dopamine uptake via dopamine transporters, treating them with varying concentrations of radiolabeled dopamine, isolating the synaptosomes, and measuring radioactivity to quantify dopamine uptake.

Assessment of Dopaminergic Homeostasis in Mice by Use of High-performance Liquid Chromatography Analysis and Synaptosomal Dopamine Uptake

Synaptosomal dopamine uptake and high-performance liquid chromatography analysis represent experimental tools to investigate dopamine homeostasis in mice by assessing the function of the dopamine transporter and levels of dopamine in striatal tissue, respectively. Here we present protocols to measure dopamine tissue content and assess the functionality of the dopamine transporter.

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

2025

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.
