In Vivo Neurotransmission

In vivo neurotransmission is the communication between neurons and their target cells within an intact living nervous system, where signaling shapes sensation, movement, cognition, and behavior. When an action potential reaches a presynaptic terminal, calcium influx triggers neurotransmitter release into the synaptic cleft; the molecules then bind postsynaptic receptors and are cleared through reuptake, enzymatic degradation, or diffusion. Studying this process in living animals connects molecular signaling to neural circuits and behavior, while approaches such as electrophysiology, microdialysis, and genetically encoded sensors reveal changes in neurotransmitter dynamics. These findings support research into brain function, neurological disorders, and potential therapeutic strategies.

Adrenergic Neurons: Neurotransmission

2023

Postganglionic sympathetic fibers (except those supplying the sweat glands) releasing noradrenaline or norepinephrine are called noradrenergic or adrenergic neurons. Noradrenaline, dopamine, adrenaline, or epinephrine are collectively called "catecholamines" as they contain a catechol moiety and an amine side chain. The five stages of neurotransmitter release involve their synthesis, storage, release, reuptake and metabolism. Synthesis: Catecholamine synthesis requires tyrosine, which is taken...

Fast-scan cyclic voltammetry can monitor in vivo dopamine neurotransmission in the context of drugs, disease, and other experimental manipulations. This work describes the implementation of QNsim1.0, a software to model electrically stimulated dopamine responses according to the quantitative neurobiological model to quantify estimates of dopamine release and reuptake dynamics.

Cholinergic Neurons: Neurotransmission

Cholinergic neurotransmission involves the synthesis and the release of acetylcholine (ACh) in order to transmit nerve impulses across the synapse. The process begins with the synthesis of acetyl CoA, a precursor for ACh, from ATP, acetate, and coenzyme A in the mitochondria. Choline, another vital precursor, is transported inside the neuron through choline transporters, including high-affinity choline transporter CHT1, low-affinity choline transporter CTL1, and lower-affinity choline...

Ex vivo ERG can be used to record electrical activity of retinal cells directly from isolated intact retinas of animals or humans. We demonstrate here how common in vivo ERG systems can be adapted for ex vivo ERG recordings in order to dissect the electrical activity of retinal cells.
