Drosophila Alzheimer Model

Drosophila Alzheimer models are genetically engineered fruit flies used to reproduce key molecular and cellular features of Alzheimer’s disease, including amyloid-β toxicity, tau-related pathology, neuronal dysfunction, and memory loss. In common designs, researchers express human disease-associated genes in selected fly neurons, where altered protein production can disrupt synaptic signaling, promote neuronal degeneration, and impair learning or lifespan. These models enable rapid genetic screening and evaluation of candidate therapeutic compounds under controlled laboratory conditions. Because flies develop quickly and share conserved cellular pathways with humans, they help connect disease-associated proteins to neural mechanisms while complementing studies in mammalian systems.

One of the pathological characteristics of AD is the formation of Amyloid β protein positive neuritic plaques. In this protocol we describe two methods to detect neuritic plaques in transgenic AD model mice: immunohistochemical detection using the ABC and DAB method and fluorescent detection using thioflavin S staining method.

Here, we describe a new approach to inflict closed-head traumatic brain injury in Drosophila melanogaster. Our method has the advantage of directly delivering repetitive impacts with adjustable strength to the head alone. Further exploration of the invertebrate system will help to illuminate the pathogenesis of chronic traumatic encephalopathy.
