Hippocampal Architecture Preservation

Hippocampal architecture preservation is the maintenance of the hippocampus’s layered organization, cellular arrangement, and neural connections during tissue collection, processing, and analysis. It depends on carefully controlled procedures such as rapid fixation, appropriate sectioning, and conditions that minimize tissue distortion, degradation, and loss of spatial relationships. Preserving this structure allows researchers to examine regions including the dentate gyrus and cornu ammonis, relate neuronal morphology to circuit organization, and interpret changes associated with learning, memory, development, and neurological disease. Reliable architectural preservation therefore strengthens the anatomical basis for microscopy, histology, and experimental neuroscience.

This paper presents a complete methodology to prepare and preserve in vitro acute hippocampal slices from adult mice. This protocol allows the recording of very stable long-lasting long-term potentiation (LTP) for more than 8 hours with a success rate of 95%.

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.
