In Vitro Skin Models

In vitro skin models are laboratory systems that reproduce key features of human skin outside the body, providing controlled platforms for studying tissue function, disease, and treatment responses. They are typically built from cultured skin cells, including keratinocytes and fibroblasts, arranged in layered cultures that can develop barrier properties and interact with other cell types. In neuroscience, models incorporating sensory neurons help examine neurocutaneous signaling, inflammation, itch, pain, and responses to chemical or physical stimuli. These systems can reduce reliance on animal studies, support drug and cosmetic testing, and clarify how skin communicates with the nervous system in health and disease.

Improving 2D and 3D Skin In Vitro Models Using Macromolecular Crowding

We present a protocol to obtain cell-derived matrices rich in extracellular matrix proteins, using macromolecular crowders (MMC). In addition, we present a protocol which incorporates MMC in 3D organotypic skin co-culture generation, which reduces culture time while maintaining maturity of construct.

This video describes the procedure to induce two-stage skin carcinogenesis to study tumor initiation by topical application of chemicals. In the first step, a mutagen, DMBA, is applied onto the skin of a mouse model which initiates tumor formation by causing mutations in the stem cells, followed by the application of a growth stimulator, TPA, which accelerates skin papilloma formation.
