Interstitial Branching

Interstitial branching is the formation of new neuronal processes from points along an existing axon or dendrite rather than exclusively at its terminal, enabling neurons to expand and refine their connections. In axons, local remodeling of the cytoskeleton produces a protrusion from the shaft that develops into a collateral branch, guided by extracellular cues and neuronal activity toward appropriate targets. This process contributes to neural circuit formation, target innervation, and connectivity during development, while regulated branch growth can also support circuit remodeling and recovery after injury. Studying interstitial branching helps explain how neurons establish complex networks in the nervous system.

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure. Comparing the absorbance maxima of the uncharacterized sample with that of polysaccharides with known branched structures indicates the presence of branching in the uncharacterized sample.

Interstitial fluid flow is elevated in solid tumors and can modulate tumor cell invasion. Here we describe a technique to apply interstitial fluid flow to cells embedded in a matrix and then measure its effects on cell invasion. This technique can be easily adapted to study other systems.

This protocol describes a method to collect cardiac interstitial fluid from the isolated, perfused rat heart. To physically separate interstitial transudate from coronary venous effluent perfusate, the Langendorff perfused heart is inverted, and the transudate (interstitial fluid) formed on the cardiac surface is collected using a soft latex cap.
