Organelle Fraction Purification

Organelle fraction purification is a biochemical technique that separates subcellular structures from disrupted cells, enabling researchers to analyze organelle-specific composition and function. The process typically combines controlled homogenization with differential centrifugation or density-gradient centrifugation, in which particles separate according to size, mass, and buoyant density while preserving target fractions under suitable buffer and temperature conditions. In neuroscience, purified synaptic vesicles, mitochondria, nuclei, or microsomal membranes can support studies of neurotransmitter storage, energy metabolism, gene regulation, and membrane protein activity. This approach helps connect organelle dysfunction with neuronal signaling, neurodegeneration, and other brain disorders, although fraction purity must be assessed carefully.

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.

We describe a rapid and effective method for purification of mitochondria from the yeast Saccharomyces cerevisiae. This method enables the high-yield isolation of pure mitochondria that are essentially free of contamination by other organelles and retain their structural and functional integrity after their purification.
