Photoconvertible Fluorophores

Photoconvertible fluorophores are fluorescent molecules whose emission properties can be permanently altered by exposure to specific wavelengths of light, making them useful for resolving where labeled molecules are and how they move over time. In their initial state, these probes emit one color; focused illumination triggers a photochemical rearrangement that shifts their excitation or emission spectrum, allowing selected cells, organelles, or proteins to be marked within a larger population. In neuroscience, photoconversion supports live imaging of neuronal architecture, axonal transport, synaptic remodeling, and cell lineage, while complementing techniques for mapping circuits and tracking activity-related structural changes.

Here, we present a method for the photoactivated switch of photoconvertible fluorescent proteins (PCFPs) in the living zebrafish embryo and further tracking of photoconverted protein at specific time points during development. This methodology allows monitoring of cell biological events underlying different developmental processes in a live vertebrate organism.
