Vascular Compartment Isolation

Vascular compartment isolation is a laboratory method for separating blood vessels and vascular-associated cells from surrounding brain tissue so their structure, molecular profile, and interactions can be studied independently. The procedure typically uses tissue homogenization or enzymatic dissociation, followed by filtration, centrifugation, or density-based separation to enrich microvessels while reducing neural and glial contamination. Isolated compartments support analyses of the blood-brain barrier, endothelial and perivascular cells, vascular signaling, and neurovascular coupling. By linking vascular changes to neuronal function and disease, the method helps investigate stroke, neurodegeneration, inflammation, and therapeutic delivery across the brain vasculature.

We provide a novel strategy to isolate viral replication compartments (RC) from adenovirus (Ad)-infected human cells. This approach represents a cell-free system that can help to elucidate the molecular mechanisms regulating viral genome replication and expression as well as regulation of viral-host interactions established at the RC.
