Vesicle Fusion Analysis

Vesicle fusion analysis is the study of how membrane-bound vesicles merge with a target membrane, a process essential for regulated secretion and, in neurons, neurotransmitter release. In synaptic transmission, an action potential opens voltage-gated calcium channels; calcium activates fusion machinery, driving synaptic vesicles to dock, prime, and fuse with the presynaptic membrane so their contents enter the synaptic cleft. Experimental analyses measure fusion timing, frequency, and molecular regulation through fluorescence imaging, electrophysiology, and related approaches. These measurements clarify how synapses communicate and reveal mechanisms underlying neural circuit function, synaptic plasticity, and neurological disease.

Fusion of Secretory Vesicles with the Plasma Membrane

2023

Proteins and neurotransmitters in secretory vesicles can be released from a cell upon vesicle docking, priming, and fusion with the plasma membrane. Vesicles are docked and primed in preparation for the quick exocytosis of their contents in response to a stimulus. The fusion process is mainly carried out by a SNAP Receptor or SNARE complex, consisting of synaptobrevin, syntaxin-1, and SNAP-25. In 1993, Jim Rothman proposed that the antiparallel pairing of vesicular and transmembrane SNAREs, or...

Many different methods exist for the measurement of extracellular vesicles (EVs) using flow cytometry (FCM). Several aspects should be considered when determining the most appropriate method to use. Two protocols for measuring EVs are presented, using either individual detection or a bead-based approach.
