Amine Depletion

Amine depletion is the reduction of stored biogenic amines, including dopamine, norepinephrine, and serotonin, in cells or tissues, a process that helps explain how some pharmacological agents alter neurotransmission. It commonly occurs when vesicular storage or release is disrupted, such as when vesicular monoamine transporter function is inhibited, leaving amines exposed to cytoplasmic metabolism and reducing their availability for synaptic signaling. In pharmacology, amine depletion is used to investigate the roles of monoamines in behavior, cardiovascular regulation, and drug responses, while also helping researchers interpret therapeutic effects, adverse reactions, and adaptive changes following prolonged treatment.

Amines to Amides: Acylation of Amines

2025

Various carboxylic acid derivatives (such as acid chlorides, esters, and anhydrides) can be used for the acylation of amines to yield amides. The reaction requires two equivalents of amines. The first amine molecule functions as a nucleophile and attacks the carbonyl carbon to produce a tetrahedral intermediate. This is followed by the loss of the leaving group and restoration of the C=O bond. Next, the second equivalent of amine serves as a Brønsted base and deprotonates the quaternary amide...

Depletion and Reconstitution of Macrophages in Mice

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.

Preparation of Amines: Alkylation of Ammonia and Amines

2025

Alkylation is one of the methods used to prepare amines. Direct alkylation of ammonia or a primary amine with an alkyl halide gives polyalkylated amines along with a quaternary ammonium salt through successive SN2 reactions. This process of making the quaternary salt through the direct alkylation method is called exhaustive alkylation. Each alkylation step makes the nitrogen center more nucleophilic, which triggers successive alkylations until a quaternary ammonium salt is formed. Considering...
