Anti-inflammatory Drugs

Anti-inflammatory drugs are medicines that reduce inflammation, an immune response that can cause pain, swelling, heat, and tissue damage. They act through different mechanisms: nonsteroidal anti-inflammatory drugs inhibit cyclooxygenase enzymes and lower prostaglandin production, while corticosteroids alter gene transcription to suppress multiple inflammatory pathways. In pharmacology, these drugs are used to manage conditions such as arthritis, allergies, asthma, and other inflammatory disorders. Understanding their targets, therapeutic effects, and adverse reactions supports appropriate drug selection, improves symptom control, and guides the development of treatments with greater effectiveness and safety.

Take cultures of human monocyte-derived microglia-like cells.Introduce nanoparticles containing an anti-inflammatory drug into the experimental culture and the free drug into the control. The nanoparticles feature a hydrophilic shell and a hydrophobic core encapsulating the lipophilic drug. The cells endocytose the nanoparticles, which release the drug intracellularly, enhancing its delivery. The free drug enters by passive diffusion, a process constrained by the concentration gradient, which...

Here, we established a method for drug efficacy testing with surgical specimens of brain tumors, termed “tumor explant method”. With this method, we can evaluate drug efficacy without breaking the microenvironment of solid tumors. To validate reliability of this method, we describe representative data with our glioma specimen treated with the current first-line chemotherapeutic agent, temozolomide.

Antiasthma Drugs: Mast Cell Stabilizers and Anti-IgE Drugs

2024

Asthma is a chronic respiratory condition for which new therapeutic avenues, including anti-inflammatory drugs like mast cell stabilizers and anti-IgE treatments, continue to be developed. Mast cell stabilizers, such as cromolyn (also known as sodium cromoglycate) and nedocromil (Tilade), are effective drugs in asthma management. These stabilizers hinder histamine release by skillfully obstructing the activation of mast cells and other cellular entities. Notably, they navigate this task without...

We present an in vitro model which allows the study and analysis of coagulation in whole, non-anticoagulated blood. Anticoagulation in the system depends on the natural anticoagulation effect of healthy endothelial cells and endothelial cell activation will result in clotting.
