Pulmonary Artery Smooth Muscle Cells

Pulmonary artery smooth muscle cells are contractile cells in the vessel wall that regulate pulmonary vascular tone and blood flow between the heart and lungs. Their contraction is controlled largely by intracellular calcium: vasoconstrictor signals increase calcium entry or release, activating myosin and narrowing the artery, while relaxing signals reduce calcium-dependent force. In pharmacology, these cells provide a model for studying pulmonary hypertension, vascular remodeling, and drug responses. Investigations of calcium channels, cyclic nucleotide pathways, endothelin signaling, and nitric oxide-mediated relaxation help explain how therapies alter pulmonary vascular resistance and support the development of targeted treatments.

We have developed a novel and reproducible technique to isolate primary cultures of pulmonary artery smooth muscle cells (PASMC) from mice as young as P7, thereby allowing better study of the signaling pathways involved in neonatal smooth muscle cell contraction and relaxation.
