Sleep Restriction

Sleep restriction is the deliberate reduction of time spent in bed or allowed for sleep, a condition used to study how insufficient sleep affects physiology and behavior. By limiting the opportunity for sleep, it increases homeostatic sleep pressure and can alter circadian regulation, alertness, cognition, mood, and performance. In pharmacology, sleep restriction provides a controlled model for evaluating how sedative-hypnotic drugs, stimulants, and other compounds influence sleepiness, wakefulness, attention, and treatment-related adverse effects. These studies help distinguish drug actions from the effects of sleep loss and support safer, more effective therapies for sleep and neurobehavioral disorders.

Restriction Enzyme Digests

Restriction enzymes or endonucleases recognize and cut DNA at a specific sequence. These enzymes occur naturally in bacteria as a defense against bacteriophages - viruses that infect bacteria. Bacterial restriction enzymes cut the invading bacteriophage DNA while leaving the bacterial genomic DNA unharmed due to addition of methyl groups. This video explains the basic principles of restriction enzymes including: how restriction enzymes are named and the types of recognition sites and...

Quantitative Measurement of the Immune Response and Sleep in Drosophila

To understand a link between the immune response and behavior, we describe a method to measure locomotor behavior in Drosophila during bacterial infection as well as the ability of flies to mount an immune response by monitoring survival, bacterial load, and real-time activity of a key regulator of innate immunity, NFκB.

DNA Isolation and Restriction Enzyme Analysis - Student Protocol

DNA Isolation ExpandNOTE: In this experiment you will perform DNA isolation under two experimental conditions: one using a buffer containing the detergent SDS and one without detergent. Hypotheses: The alternate hypothesis for this experiment might be that the sample prepared without SDS, a strong anionic detergent that breaks apart cell membranes, will yield less DNA than the sample prepared with SDS. The null hypothesis for this experiment might be that both samples will yield an equal amount...
