Dopamine Neurotransmitter

Dopamine is a neurotransmitter, a chemical messenger that enables communication between nerve cells and helps regulate behavior, cognition, movement, and physiological functions. Neurons synthesize dopamine from tyrosine, release it into the synaptic cleft in response to electrical activity, and transmit its signal by binding to dopamine receptors on neighboring cells; reuptake and enzymatic breakdown then limit the message. In psychology, dopamine signaling is closely associated with reward learning, motivation, attention, and reinforcement, although it does not simply produce pleasure. Studying dopamine pathways helps researchers understand addiction, Parkinson’s disease, schizophrenia, and how behavior is shaped by experience.

This video demonstrates the assessment of dopamine uptake in synaptosomes harvested from mouse brains. It outlines steps for preparing synaptosomes for dopamine uptake via dopamine transporters, treating them with varying concentrations of radiolabeled dopamine, isolating the synaptosomes, and measuring radioactivity to quantify dopamine uptake.

We present a novel PET imaging approach for capturing dopamine fluctuations induced by cigarette smoking. Subjects smoke in the PET scanner. Dynamic PET images are modeled voxel-by-voxel in time by lp-ntPET, which includes a time-varying dopamine term. The results are 'movies' of dopamine fluctuations in the striatum during smoking.
