Personality Fixation

Personality fixation is a psychodynamic concept describing the persistence of patterns, needs, or coping styles associated with an earlier stage of psychological development. It proposes that unresolved conflicts or unmet needs can reinforce defenses and repetitive behaviors, shaping how a person responds to relationships, stress, and changing circumstances. In psychology, the concept helps organize interpretations of rigid personality patterns and recurring interpersonal difficulties, although its Freudian origins and empirical support remain debated. Contemporary clinicians may consider fixation alongside attachment, learning history, social context, and current mental-health symptoms rather than treating it as a standalone diagnosis.

Fluorescence in-situ hybridization (FISH) is often required in combination with histopathology and molecular diagnostics for selection of therapy in personalized medicine. A novel non-cross-linking, formalin-free tissue fixative that allows high quality morphologic, molecular and FISH analyses from the same specimen by addition of a post-fixation step before FISH is presented.

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens. The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper.
