Carboplatin (CPT), today the most important platinum(II) anticancer drug, manifests an extreme kinetic inertness, in vitro, at physiological pH; the actual mechanisms for its activation inside cells are still poorly understood. We show here that horse heart cytochrome c reacts with CPT, leading to the formation of stable platinum/protein adducts. The two major CPT-cytochrome c species resulting from the aforementioned reaction were characterised by electrospray ionisation mass spectrometry (ESI-MS). Notably, both these adducts have the ability to react with guanosine 5'-monophosphate (5'-GMP), giving rise to the respective cytochrome c-CPT-5'-GMP ternary complexes. Additional ESI-MS measurements on enzymatically cleaved cytochrome c adducts suggest that protein platination probably occurs at Met65. The mechanistic implications of these findings are discussed.

Nous montrons ici que le cheval coeur du cytochrome c r&eacute;agit avec CPT, conduisant &agrave; la formation de platine stable/prot&eacute;ines adduits. Les deux principales CPT-cytochrome c esp&egrave;ces r&eacute;sultant de la r&eacute;action ci-dessus ont &eacute;t&eacute; caract&eacute;ris&eacute;s par spectrom&eacute;trie de masse d&#39;ionisation &eacute;lectrospray (ESI-MS). Wir zeigen hier, dass Pferd Herz Cytochrom C reagiert mit CPT, f&uuml;hrt zur Bildung von stabilen Platin/Protein-Addukte. Die beiden wichtigsten CPT-Cytochrom C Arten infolge der oben genannten Reaktion waren gepr&auml;gt von Elektrospray-Ionisations-Massenspektrometrie (ESI-MS). Vor allem diese beiden Addukte mit 5&#39;-Guanosinmonophosphat (5&#39;-GMP), reagieren, die zu den jeweiligen Cytochrom c-CPT-5&#39;-GMP tern&auml;re komplexe haben. Zus&auml;tzliche ESI-MS-Messungen an enzymatisch gespalten Cytochrom c Addukte deuten darauf hin, dass Protein-Platination tritt wahrscheinlich zur Met65. Die mechanistische Folgerung aus diesen Erkenntnissen werden diskutiert.

Le due principali CPT-citocromo c specie derivanti dalla reazione di cui sopra sono state caratterizzate da spettrometria di massa di ionizzazione elettrospray (ESI-MS). The primary sequence of proteins usually dictates a single tertiary and quaternary structure. However, certain proteins undergo reversible backbone rearrangements. Such metamorphic proteins provide a means of facilitating the evolution of new folds and architectures. However, because natural folds emerged at the early stages of evolution, the potential role of metamorphic intermediates in mediating evolutionary transitions of structure remains largely unexplored. We evolved a set of new proteins based on approximately 100 amino acid fragments derived from tachylectin-2--a monomeric, 236 amino acids, five-bladed beta-propeller. Their structures reveal a unique pentameric assembly and novel beta-propeller structures. Although identical in sequence, the oligomeric subunits adopt two, or even three, different structures that together enable the pentameric assembly of two propellers connected via a small linker. Most of the subunits adopt a wild-type-like structure within individual five-bladed propellers. However, the bridging subunits exhibit domain swaps and asymmetric strand exchanges that allow them to complete the two propellers and connect them. Thus, the modular and metamorphic nature of these subunits enabled dramatic changes in tertiary and quaternary structure, while maintaining the lectin function. These oligomers therefore comprise putative intermediates via which beta-propellers can evolve from smaller elements. Our data also suggest that the ability of one sequence to equilibrate between different structures can be evolutionary optimized, thus facilitating the emergence of new structures.

La secuencia primaria de prote&#xED;nas generalmente dicta una sola estructura terciaria y cuaternaria. Sin embargo, ciertas prote&#xED;nas experimentar reordenamientos reversibles columna vertebral. Tales prote&#xED;nas metam&#xF3;rficas proporcionar un medio para facilitar la evoluci&#xF3;n de nuevos pliegues y arquitecturas. Sin embargo, debido a los pliegues naturales surgi&#xF3; en las primeras etapas de la evoluci&#xF3;n, el papel potencial de los productos intermedios metam&#xF3;rficas en la mediaci&#xF3;n de las transiciones evolutivas de la estructura se mantiene en gran parte inexplorado. Hemos desarrollado un conjunto de nuevas prote&#xED;nas sobre la base de aproximadamente 100 fragmentos de amino&#xE1;cidos derivados de tachylectin-2 - una monom&#xE9;ricas, 236 amino&#xE1;cidos, cinco palas de la h&#xE9;lice beta. Sus estructuras revelan una asamblea &#xFA;nica pentam&#xE9;rica y nuevas estructuras de h&#xE9;lice beta. Aunque id&#xE9;nticos en secuencia, las subunidades oligom&#xE9;ricas adoptar dos, o incluso tres estructuras, diferentes que juntas permiten el montaje de dos h&#xE9;lices pentam&#xE9;rica conectado a trav&#xE9;s de un enlazador peque&#xF1;o. La mayor&#xED;a de las subunidades adoptar una estructura de tipo salvaje-como dentro de cada cinco palas h&#xE9;lices. Sin embargo, las subunidades puente exhiben los swaps de dominio y los intercambios asim&#xE9;tricos un cord&#xF3;n que les permitan completar las dos h&#xE9;lices y conectarlos. Por lo tanto, la naturaleza modular y metam&#xF3;rficas de estas subunidades permitido cambios dr&#xE1;sticos en la estructura terciaria y cuaternaria, mientras que el mantenimiento de la funci&#xF3;n de la lectina.
