Caenorhabditis elegans is an important model organism for modern biologic research. An essential aspect of C. elegans research is the production of transgenic animals for study. These are often generated via microinjection, but biolistic bombardment has become increasingly popular. However, many of the plasmids previously generated for use in microinjection are not readily used for bombardment due to the lack of a convenient marker. The unc-119 gene is often used as a marker since unc-119 rescue can be observed at low magnification, allowing rescued animals to be easily distinguished from the larger number of non-rescued animals. Here we report the use of homologous recombination in Escherichia coli as a method to insert a cassette containing the unc-119 gene into commonly used plasmids at the site of the ampicillin resistance gene which is simpler than other methods like subcloning. These cassettes are flanked by regions homologous to the 5' and 3' ends of the ampicillin resistance gene and contain either the unc-119 gene and the kanamycin resistance gene or a unc-119:mCherry fusion gene and the kanamycin resistance gene. The resulting plasmids may be used for biolistic bombardment to yield animals that display unc-119 rescue, and also express the recipient plasmid transgene.

Allerdings sind viele der Plasmide, die zuvor f&uuml;r den Einsatz in Mikroinjektion generiert nicht leicht f&uuml;r Bombardement aufgrund des Mangels an eine bequeme Markierung verwendet. Das Unc-119-Gen wird h&auml;ufig als Marker verwendet, da Unc-119-Rettung bei kleiner Vergr&ouml;&szlig;erung, so dass gerettete Tiere leicht unterschieden aus der gr&ouml;&szlig;eren Anzahl von Tieren nicht gerettet werden beobachtet werden kann. Maintenance of a stable, properly folded, and catalytically active proteome is a major challenge to organisms in the face of multiple internal and external stresses which damage proteins and lead to protein misfolding. Here we show that internal metabolic stress produced by reactive intermediates resulting from tyrosine degradation triggers the expression of the aip-1 gene, which is critical in responses to the environmental toxin arsenic and the clearance of unstable polyglutamine and Abeta proteins. aip-1 acts via binding to the proteosome and enhancing proteosomal function. We find that full induction of aip-1 depends on the oxidative-stress-responsive skn-1 transcription factor but significant induction still occurs without skn-1. Importantly, activation of skn-1 with wdr-23(RNAi), which dramatically induces the expression of other skn-1 target genes, produces a minimal increase in aip-1 expression. This suggests that the previously demonstrated specificity in aip-1/AIRAP induction could reflect the actions of multiple synergistic activators, such as the heat shock factor homolog hsf-1, which we also find is required for full induction. These may be triggered by proteosome dysfunction, as we find that this event links the multiple inducers of aip-1. Together, our results show that cell stress triggers aip-1 expression by both skn-1-dependent and -independent pathways.

El mantenimiento de un proteoma estable, plegada correctamente, y activa catal&#xED;ticamente es un gran desaf&#xED;o para los organismos de cara a m&#xFA;ltiples presiones internas y externas que da&#xF1;an las prote&#xED;nas y dar lugar a mal plegamiento de prote&#xED;nas. Aqu&#xED; nos muestran que la tensi&#xF3;n interna metab&#xF3;lica producida por intermediarios reactivos que resultan de la degradaci&#xF3;n de la tirosina activa la expresi&#xF3;n de la AIP-1 gen, que es fundamental en las respuestas al ars&#xE9;nico, la toxina del medio ambiente y la liquidaci&#xF3;n de poliglutaminas inestable y prote&#xED;nas Abeta. AIP-1 act&#xFA;a a trav&#xE9;s de la uni&#xF3;n a la funci&#xF3;n proteosomal proteosoma y mejorar. Encontramos que la inducci&#xF3;n completa de la AIP-1 depende del estr&#xE9;s oxidativo que responde a SKN-1 factor de transcripci&#xF3;n, sino una inducci&#xF3;n significativa persiste sin SKN-1. Es importante destacar que, la activaci&#xF3;n de SKN-1 con WDR-23 (RNAi), que dr&#xE1;sticamente induce la expresi&#xF3;n de otros genes diana 1-SKN, produce un aumento m&#xED;nimo en la AIP-1 de expresi&#xF3;n. Esto sugiere que la especificidad demostrado previamente en la inducci&#xF3;n aip-1/AIRAP podr&#xED;an reflejar las acciones de m&#xFA;ltiples activadores sin&#xE9;rgicos, tales como el choque t&#xE9;rmico hom&#xF3;logo factor de HSF-1, que tambi&#xE9;n encontramos es necesaria para la inducci&#xF3;n completa. Esto puede ser provocado por una disfunci&#xF3;n del proteosoma, como nos encontramos con que este evento une a los m&#xFA;ltiples inductores de AIP-1. En conjunto, nuestros resultados muestran que el estr&#xE9;s celular desencadena AIP-1 de expresi&#xF3;n por ambas v&#xED;as de SKN-1-dependientes e independientes.
