Very high cell densities and optimal vascularization characterize among others organs and tissues in vivo. In order to study organ-specific functions in vitro or to make use of them in medical devices/treatments in the future, this natural architecture should be rebuilt. An important aspect in this context is the appropriate ratio of medium to cell volume being so far not optimally reestablished in most of the currently available in vitro systems. To improve such culture conditions, we constructed a microstructure to culture hepatocytes and (without any addition of extracellular matrix material) characterized liver tissue in the form of evenly sized aggregates. The liver-specific differentiation status of such aggregates was monitored by their ability to perform CYP450 dependent xenobiotic metabolism along with the measurement of albumin secretion. Freshly isolated adult rat hepatocytes show an initial loss of total CYP450 content and of associated activities (mixed function oxidases). However, in the aggregate system, this level did not decrease further but remained stable or even increased throughout the culture period of 10-13 days. The CYP450 dependent metabolism of the hepatocytes is able to respond to classic inducing agents. The described culture efficiently supports liver-specific functions of adult rat hepatocytes and seems to be suited not only for use in an extracorporeal liver device but also for the formation of evenly sized small aggregates to be of use in transplantation of differentiated liver tissue. Moreover, after design variations, the microstructure can be applied for functional analysis of metabolically active hepatocytes as well as for toxicological and pharmacological validation.

Microstructured scaffolds for liver tissue cultures of high cell density: morphological and biochemical characterization of tissue aggregates.

Densit&#xE9;s cellulaires tr&#xE8;s &#xE9;lev&#xE9;es et la vascularisation optimale entre les organes de caract&#xE9;riser d&#x26;#39;autres tissus et in vivo. Pour &#xE9;tudier l&#x26;#39;orgue fonctions sp&#xE9;cifiques in vitro ou de faire usage d&#x26;#39;entre eux dans les dispositifs m&#xE9;dicaux ou des traitements &#xE0; l&#x26;#39;avenir, cette architecture naturelle devrait &#xEA;tre reconstruit. Un aspect important dans ce contexte est le ratio appropri&#xE9; de la moyenne &#xE0; volume de la cellule &#xE9;tant pour l&#x26;#39;instant pas de mani&#xE8;re optimale r&#xE9;tabli dans la plupart des syst&#xE8;mes actuellement disponibles dans in vitro. Pour am&#xE9;liorer les conditions de culture telles, nous avons construit une microstructure pour les h&#xE9;patocytes de la culture et (sans aucun ajout de mat&#xE9;riau de la matrice extracellulaire) caract&#xE9;ris&#xE9; les tissus du foie sous la forme d&#x26;#39;agr&#xE9;gats de taille &#xE9;gale. Le statut de diff&#xE9;renciation sp&#xE9;cifique du foie de ces agr&#xE9;gats a &#xE9;t&#xE9; suivie par leur capacit&#xE9; &#xE0; effectuer des CYP450 m&#xE9;tabolisme des x&#xE9;nobiotiques d&#xE9;pendante avec la mesure de la s&#xE9;cr&#xE9;tion de l&#x26;#39;albumine. Fra&#xEE;chement isol&#xE9;es des h&#xE9;patocytes de rats adultes montrent une perte initiale de contenu CYP450 totale et des activit&#xE9;s associ&#xE9;es (oxydases &#xE0; fonction mixte). Toutefois, dans le syst&#xE8;me global, ce niveau n&#x26;#39;a pas diminu&#xE9; davantage, mais est rest&#xE9; stable ou a m&#xEA;me augment&#xE9; tout au long de la p&#xE9;riode de culture de 10-13 jours. Le m&#xE9;tabolisme du CYP450 d&#xE9;pend des h&#xE9;patocytes est capable de r&#xE9;pondre &#xE0; des agents inducteurs classiques. La culture d&#xE9;crit soutient efficacement le foie des fonctions sp&#xE9;cifiques des h&#xE9;patocytes de rats adultes et semble &#xEA;tre adapt&#xE9; non seulement pour une utilisation dans un dispositif du foie extracorporelle, mais aussi pour la formation de petits agr&#xE9;gats de taille &#xE9;gale pour &#xEA;tre d&#x26;#39;utilisation dans la transplantation de tissu h&#xE9;patique diff&#xE9;renci&#xE9;e. En outre, apr&#xE8;s les variations de conception, de la microstructure peut &#xEA;tre appliqu&#xE9;e pour l&#x26;#39;analyse fonctionnelle des h&#xE9;patocytes m&#xE9;taboliquement actifs, ainsi que pour la validation toxicologiques et pharmacologiques.

Cella molto alta densit&agrave; e vascolarizzazione ottimale caratterizzano tra gli altri organi e tessuti in vivo. Al fine di studiare le funzioni di organo-specifiche in vitro o a fare uso di loro in dispositivi/trattamenti medici in futuro, questa architettura naturale dovrebbe essere ricostruita. Un aspetto importante in questo contesto &egrave; il rapporto appropriato di medio volume cella finora essere ristabilita in modo non ottimale nella maggior parte dei sistemi in vitro attualmente disponibili. Per migliorare tali condizioni di cultura, abbiamo costruito un microstruttura di epatociti di cultura e (senza alcuna aggiunta di materiale extracellulare) caratterizzato tessuto epatico sotto forma di aggregati in modo uniforme dimensioni. Dalla loro capacit&agrave; di eseguire CYP450 dipendente dal metabolismo xenobiotico insieme con la misurazione della secrezione di albumina &egrave; stato monitorato lo stato specifico del fegato differenziazione di tali aggregati. Epatociti di ratto adulto appena isolato mostrano una perdita iniziale del contenuto totale di CYP450 e delle attivit&agrave; connesse (miscelati ossidasi a funzione). Tuttavia, nell&#39;aggregato sistema, questo livello non ha fatto diminuire ulteriormente ma &egrave; rimasto stabile o addirittura aumentato per tutto il periodo della cultura di 10-13 giorni. Il CYP450 dipendente dal metabolismo degli epatociti &egrave; in grado di rispondere agli agenti di induzione classici. La cultura descritta in modo efficiente supporta le funzioni specifiche del fegato di epatociti di ratto adulto e sembra di essere adatto non solo per l&#39;uso in un dispositivo epatico extracorporeo, ma anche per la formazione di uniformemente dimensioni piccole aggregazioni per essere di uso nel trapianto del tessuto epatico differenziato. Inoltre, dopo variazioni di design, la microstruttura pu&ograve; essere applicata per l&#39;analisi funzionale di epatociti metabolicamente attivi anche per quanto riguarda la convalida tossicologica e farmacologica.

It has been reported that magnetic fields with flux densities ranging from microT to mT are able to induce heat shock factor, HSP72 mRNA or heat shock proteins in various cells. In this study we investigated changes in the HSP72 mRNA transcription level in three cell lines (HL-60, H9c2, and Girardi heart cells) and in the intracellular HSP72 protein content in two cell lines (HL-60 and Girardi heart cells) after treatment schemes using electromagnetic fields with a flux density of 2 microT to 4 mT, a frequency of 50 Hz and exposure times from 15 to 30 min. None of the treatments or modalities showed any significant effect on the HSP72 protein level, although HSP72 mRNA could be induced, at least to some extent, with one of the parameter combinations in all cell lines tested. Obviously, HSP72 mRNA transcription and translation are not necessarily coupled in certain cells. This leads to the conclusion that electromagnetic field effects on HSP72 mRNA levels are not indicative for downstream effects unless increased mRNA levels can be correlated with increased HSP72 protein levels as well.

We describe a multi-purpose platform for the three-dimensional cultivation of tissues. The device is composed of polymer chips featuring a microstructured area of 1-2 cm(2). The chip is constructed either as a grid of micro-containers measuring 120-300 x 300 x 300 microm (h x l x w), or as an array of round recesses (300 microm diameter, 300 microm deep). The micro-containers may be separately equipped with addressable 3D-micro-electrodes, which allow for electrical stimulation of excitable cells and on-site measurements of electrochemically accessible parameters. The system is applicable for the cultivation of high cell densities of up to 8 x 10(6) cells and, because of the rectangular grid layout, allows the automated microscopical analysis of cultivated cells. More than 1000 micro-containers enable the parallel analysis of different parameters under superfusion/perfusion conditions. Using different polymer chips in combination with various types of bioreactors we demonstrated the principal suitability of the chip-based bioreactor for tissue culture applications. Primary and established cell lines have been successfully cultivated and analysed for functional properties. When cells were cultured in non-perfused chips, over time a considerable degree of apoptosis could be observed indicating the need for an active perfusion. The system presented here has also been applied for the differentiation analysis of pluripotent embryonic stem cells and may be suitable for the analysis of the stem cell niche.

A chip-based platform for the in vitro generation of tissues in three-dimensional organization.

On d&#xE9;crit une plateforme &#xE0; usages multiples pour la culture tridimensionnelle des tissus. Le dispositif est compos&#xE9; de copeaux de polym&#xE8;res pr&#xE9;sentant une zone microstructur&#xE9;e de 1-2 cm (2). La puce est r&#xE9;alis&#xE9;e soit par une grille de micro-r&#xE9;cipients de mesure de 120 &#xE0; 300 x 300 x 300 microm&#xE8;tre (HxLxP), ou en tant que tableau d&#x26;#39;&#xE9;videments ronds (300 diam&#xE8;tre microm&#xE8;tre, 300 microm&#xE8;tre profonde). Les micro-conteneurs peuvent &#xEA;tre s&#xE9;par&#xE9;ment &#xE9;quip&#xE9; adressables 3D-micro-&#xE9;lectrodes, qui permettent pour la stimulation &#xE9;lectrique des cellules excitables et sur place de mesures des param&#xE8;tres accessibles par voie &#xE9;lectrochimique. Wir beschreiben eine Mehrzweck-Plattform f&uuml;r den dreidimensionalen Anbau von Geweben. Das Ger&auml;t besteht aus Polymer-Chips mit eine mikrostrukturierte Gegend von 1-2 cm(2). Der Chip wird erstellt, entweder als ein Raster von Mikro-Beh&auml;lter 120-300 X 300 X 300 Microm Messen (h x l x b), oder als Array von Runde Aussparungen (300 Microm Durchmesser, Tiefe 300 Microm). Mikro-Beh&auml;lter k&ouml;nnen separat mit adressierbaren 3D-Mikro-Elektroden, die elektrische Stimulation von excitable Zellen und vor-Ort-Messung elektrochemisch zug&auml;ngliche Parameter erm&ouml;glichen, ausger&uuml;stet sein. Das System gilt f&uuml;r den Anbau von hohen Zelldichten von bis zu 8 X 10 6 Zellen und aufgrund des rechteckigen Raster-Layouts, erm&ouml;glicht die automatisierte mikroskopische Analyse der kultivierten Zellen. Mehr als 1000 Mikro-Beh&auml;lter erm&ouml;glichen die parallele Analyse verschiedener Parameter unter Superfusion/Perfusion Bedingungen. Mit verschiedenen Polymer-Chips in Kombination mit verschiedenen Arten von Bioreaktoren zeigten wir die wichtigste Eignung der Chip-basierten Bioreaktor f&uuml;r Gewebekultur-Anwendungen. Prim&auml;r- und etablierte Zelllinien wurden erfolgreich kultiviert und funktionellen Eigenschaften untersucht. Wenn Zellen in nicht-perfused Chips kultiviert wurden, konnte im Laufe der Zeit ein betr&auml;chtliches Ma&szlig; an Apoptose beobachtet werden, die Notwendigkeit einer aktiven Perfusion angibt. Das hier vorgestellte System ist auch f&uuml;r die Analyse der Differenzierung von pluripotenten embryonalen Stammzellen angewendet wurde und geeignet f&uuml;r die Analyse der Stammzelle Nische.

Descriviamo una piattaforma polivalente per la coltura tridimensionale dei tessuti. Il dispositivo &egrave; composto da polimeri chip dotato di un&#39;area di 1-2 cm(2) microstrutturata. Il chip &egrave; costruito come una griglia di micro-contenitori di misura 120-300 x 300 x 300 microm (h x l x w), o come una matrice di rientranze rotonde (300 microm di diametro, profondo 300 microm). I micro-contenitori possono essere separatamente dotati indirizzabili 3D-micro-elettrodi, che consentono la stimolazione elettrica delle cellule eccitabili e misurazioni in loco dei parametri elettrochimicamente accessibile. Il sistema &egrave; applicabile per la coltivazione delle densit&agrave; alta cella di fino a 8 cellule di 10 x e, a causa del layout di griglia rettangolare, permette l&#39;analisi microscopica automatizzato delle cellule coltivate. Quando le cellule sono state coltivate in fiches non perfuso, nel corso del tempo un notevole grado di apoptosi potrebbe essere osservato che indica la necessit&agrave; di un&#39;attiva perfusione. Il sistema qui presentato &egrave; stato applicato anche per l&#39;analisi di differenziazione delle cellule staminali embrionali pluripotenti e pu&ograve; essere adatto per l&#39;analisi della nicchia delle cellule staminali.

Se describe una plataforma multi-prop&#xF3;sito para el cultivo tridimensional de los tejidos. El dispositivo se compone de chips de pol&#xED;mero que ofrecen un &#xE1;rea microestructurado de 1-2 cm (2). El chip se construye ya sea como una red de micro-contenedores de medici&#xF3;n 120-300 x 300 x 300 micras (hxlxw), o como una matriz de rebajes circulares (300 micras de di&#xE1;metro, 300 micras de profundidad). Los contenedores de micro-puede ser separado equipado con direccionables 3D micro-electrodos, que permiten la estimulaci&#xF3;n el&#xE9;ctrica de las c&#xE9;lulas excitables y mediciones in situ de par&#xE1;metros accesibles por v&#xED;a electroqu&#xED;mica. L&#xED;neas celulares primarias y constantes han sido cultivado con &#xE9;xito y se analizaron las propiedades funcionales. Cuando las c&#xE9;lulas fueron cultivadas en no-perfundidos virutas, con el tiempo un grado considerable de la apoptosis se pudo observar que indica la necesidad de una perfusi&#xF3;n activo. El sistema que aqu&#xED; se presenta se ha aplicado tambi&#xE9;n para el an&#xE1;lisis de la diferenciaci&#xF3;n de c&#xE9;lulas madre pluripotentes embrionarias y puede ser adecuado para el an&#xE1;lisis del nicho de c&#xE9;lulas madre.

While conventional rotation culture-based retinal spheroids are most useful to study basic processes of retinogenesis and tissue regeneration, they are less appropriate for an easy and inexpensive mass production of histotypic 3-dimensional tissue spheroids, which will be of utmost importance for future bioengineering, e.g. for replacement of animal experimentation. Here we compared conventionally reaggregated spheroids derived from dissociated retinal cells from neonatal gerbils (Meriones unguiculatus) with spheroids cultured on a novel microscaffold cell chip (called cf-chip) in a motion-free bioreactor. Reaggregation and developmental processes leading to tissue formation, e.g. proliferation, apoptosis and differentiation were observed during the first 10 days in vitro (div). Remarkably, in each cf-chip micro-chamber, only one spheroid developed. In both culture systems, sphere sizes and proliferation rates were almost identical. However, apoptosis was only comparably high up to 5 div, but then became negligible in the cf-chip, while it up-rose again in the conventional culture. In both systems, immunohistochemical characterisation revealed the presence of MÃ¼ller glia cells, of ganglion, amacrine, bipolar and horizontal cells at a highly comparable arrangement. In both systems, photoreceptors were detected only in spheroids from P3 retinae. Benefits of the chip-based 3D cell culture were a reliable sphere production at enhanced viability, the feasibility of single sphere observation during cultivation time, a high reproducibility and easy control of culture conditions. Further development of this approach should allow high-throughput systems not only for retinal but also other types of histotypic spheroids, to become suitable for environmental monitoring and biomedical diagnostics.

Bien que classiques de rotation de la culture &#xE0; base de sph&#xE9;ro&#xEF;des la r&#xE9;tine sont les plus utiles pour &#xE9;tudier les processus de base de retinogenesis et la r&#xE9;g&#xE9;n&#xE9;ration des tissus, ils sont moins appropri&#xE9; pour une production de masse facile et peu co&#xFB;teux de histotypic sph&#xE9;ro&#xEF;des tissus en 3 dimensions, qui seront d&#x26;#39;une importance capitale pour la bio-ing&#xE9;nierie avenir, par exemple pour le remplacement de l&#x26;#39;exp&#xE9;rimentation animale. Ici nous avons compar&#xE9; sph&#xE9;ro&#xEF;des classiquement agr&#xE9;g&#xE9;s de nouveau d&#xE9;riv&#xE9;s de cellules r&#xE9;tiniennes dissoci&#xE9;es de gerbilles (Meriones unguiculatus n&#xE9;onatales) avec sph&#xE9;ro&#xEF;des cultiv&#xE9;s sur une puce microscaffold cellule roman (cf appel&#xE9; puce) dans un bior&#xE9;acteur de mouvement libre. R&#xE9;agr&#xE9;gation et les processus de d&#xE9;veloppement menant &#xE0; la formation des tissus, par exemple, la prolif&#xE9;ration, l&#x26;#39;apoptose et la diff&#xE9;renciation ont &#xE9;t&#xE9; observ&#xE9;es pendant les 10 premiers jours in vitro (DIV). Dans les deux syst&#xE8;mes, les photor&#xE9;cepteurs ont &#xE9;t&#xE9; d&#xE9;tect&#xE9;s seulement dans les sph&#xE9;ro&#xEF;des de r&#xE9;tines P3. Avantages de la culture de cellules sur puce 3D &#xE9;taient une production fiable &#xE0; la sph&#xE8;re viabilit&#xE9; accrue, la faisabilit&#xE9; de l&#x26;#39;observation seule sph&#xE8;re pendant le temps de la culture, une grande reproductibilit&#xE9; et un contr&#xF4;le ais&#xE9; des conditions de culture. Poursuite de l&#x26;#39;&#xE9;laboration de cette approche devrait permettre &#xE0; haut d&#xE9;bit des syst&#xE8;mes non seulement pour les types de la r&#xE9;tine, mais aussi d&#x26;#39;autres de sph&#xE9;ro&#xEF;des histotypic, pour devenir adapt&#xE9;s &#xE0; la surveillance de l&#x26;#39;environnement et diagnostics biom&#xE9;dicaux.

W&auml;hrend herk&ouml;mmliche Drehung kulturbasierte retinale Sph&auml;roide n&uuml;tzlichsten grundlegenden Prozesse des Retinogenese und Geweberegeneration zu studieren sind, sind sie weniger geeignet f&uuml;r eine einfache und kosteng&uuml;nstige Massenproduktion von Histotypic 3-dimensionale Gewebe Sph&auml;roide, die f&uuml;r k&uuml;nftige Biotechnik, z.B. f&uuml;r Ersatz von Tierversuchen von gr&ouml;&szlig;ter Bedeutung sein wird. Hier verglichen wir konventionell Kriegsverbrechertribunals Sph&auml;roide von getrennten retinalen Zellen von Neugeborenen Rennm&auml;use (Meriones Unguiculatus) abgeleitet, mit Sph&auml;roide kultiviert auf eine neuartige Microscaffold-Zelle-Chip (Cf-Chip genannt) in einem Bewegungs-frei-Bioreaktor. Reaggregation und Entwicklungsprozesse zu Bildung, z.B. Proliferation, Apoptose und Differenzierung wurden w&auml;hrend der ersten 10 Tage in-vitro (Div) beobachtet. Bemerkenswert ist, in jeder Cf-Chip Mikro-Kammer entwickelt nur ein Sph&auml;roid. In beiden Systemen Kultur waren Sph&auml;re Gr&ouml;&szlig;en und Verbreitung Preise fast identisch. Apoptose wurde nur bis 5 Div vergleichsweise hoch, jedoch wurde dann in der Cf-Chip, vernachl&auml;ssigbar, w&auml;hrend es oben-wieder in der herk&ouml;mmlichen Kultur Rose. In beiden Systemen offenbart immunhistochemische Charakterisierung die Anwesenheit von M&uuml;ller-Glia-Zellen, Ganglion, Amacrine, bipolar und horizontalen Zellen bei der h&ouml;chst vergleichbare Anordnung. En ambos sistemas, los fotorreceptores se detectaron s&#xF3;lo en esferoides de retina P3. Los beneficios del cultivo de c&#xE9;lulas basada en el chip 3D eran una producci&#xF3;n fiable en la esfera mayor viabilidad, la factibilidad de la observaci&#xF3;n sola esfera durante el tiempo de cultivo, una alta reproducibilidad y f&#xE1;cil control de las condiciones de cultivo. Un mayor desarrollo de este enfoque deber&#xED;a permitir que los sistemas de alto rendimiento no s&#xF3;lo para los tipos de la retina, sino tambi&#xE9;n de otro tipo de esferoides histotypic, para convertirse en adecuado para la vigilancia ambiental y los diagn&#xF3;sticos biom&#xE9;dicos.
