Giant neurones in the caudal pontine reticular nucleus (PnC) play a crucial role in mediating the mammalian startle response. They receive input from cochlear, trigeminal and vestibular nuclei and project directly to motoneurones. Furthermore, they integrate modulatory input from different brain regions either enhancing or inhibiting startle responses. One prominent startle modulation is prepulse inhibition where a non-startling stimulus presented prior to the startle stimulus inhibits a subsequent startle response. Several behavioural studies have indicated that this inhibition is mediated by muscarinic receptors at the level of the PnC. Here, we performed whole-cell patch-clamp recordings from PnC giant neurones in acute rat brain slices in order to examine muscarinic inhibition. We stimulated afferent trigeminal and auditory fibres and applied muscarinic agonists and antagonists in order to investigate their effect on excitatory postsynaptic current amplitudes, paired-pulse ratio and passive membrane properties of PnC giant neurones. The cholinergic agonist carbachol and the muscarinic agonist oxotremorine significantly reduced excitatory postsynaptic current amplitudes and increased the paired-pulse ratio. Carbachol additionally reduced the membrane resistance of postsynaptic PnC giant neurones. The subtype-specific antagonists AF-DX116 (M2 preferring) and tropicamide (M4 preferring) antagonized the oxotremorine effect indicating that M4 and possibly M2 receptor subtypes are involved in this inhibition. The G-protein-activated inward rectifying potassium channel blocker tertiapin-Q had no effect on oxotremorine-induced inhibition of giant neurones. Our results show a mainly presynaptically mediated strong inhibition of PnC giant neurones by activation of M4 and possibly M2 receptors that presumably contribute to prepulse inhibition.

Activation of muscarinic cholinergic receptors inhibits giant neurones in the caudal pontine reticular nucleus.

Neurones g&#xE9;ants dans la partie caudale du noyau r&#xE9;ticulaire pontique (PNC) jouent un r&#xF4;le crucial dans la m&#xE9;diation de la r&#xE9;ponse de sursaut chez les mammif&#xE8;res. Ils re&#xE7;oivent d&#x26;#39;entr&#xE9;e de la cochl&#xE9;e, les noyaux du trijumeau et vestibulaire et du projet directement &#xE0; motoneurones. En outre, ils int&#xE8;grent l&#x26;#39;entr&#xE9;e modulateur de diff&#xE9;rentes r&#xE9;gions du cerveau, soit am&#xE9;liorer ou inhiber les r&#xE9;ponses surprennent. Ici, nous avons effectu&#xE9; l&#x26;#39;ensemble des cellules de patch-clamp enregistrements de neurones g&#xE9;ants de la PNC dans aigu&#xEB;s tranches de cerveau de rat afin d&#x26;#39;examiner l&#x26;#39;inhibition muscarinique. Nous avons stimul&#xE9; fibres aff&#xE9;rentes du trijumeau et auditif et appliqu&#xE9;e agonistes muscariniques et antagonistes afin d&#x26;#39;enqu&#xEA;ter sur leur effet sur excitateurs postsynaptiques amplitudes de courant, les paires d&#x26;#39;impulsions de rapport et les propri&#xE9;t&#xE9;s des membranes passives de neurones g&#xE9;ants de la PNC. Le carbachol agoniste cholinergique et la oxotr&#xE9;morine agoniste muscarinique consid&#xE9;rablement r&#xE9;duit excitateurs postsynaptiques amplitudes de courant et augmente le rapport double choc. Carbachol en outre r&#xE9;duit la r&#xE9;sistance de la membrane des neurones post-synaptiques g&#xE9;ants de la PNC. Les antagonistes sp&#xE9;cifiques du sous-type AF-DX116 (M2 pr&#xE9;f&#xE9;rant) et le tropicamide (M4 pr&#xE9;f&#xE9;rant) antagonise l&#x26;#39;effet oxotr&#xE9;morine indiquant que types de r&#xE9;cepteurs M4 et, &#xE9;ventuellement, M2 sont impliqu&#xE9;es dans cette inhibition. Le G-protein-activ&#xE9; l&#x26;#39;int&#xE9;rieur bloqueur des canaux potassiques rectifiant tertiapin-Q n&#x26;#39;a eu aucun effet sur l&#x26;#39;oxotr&#xE9;morine induite par l&#x26;#39;inhibition de neurones g&#xE9;ants. Nos r&#xE9;sultats montrent une inhibition pr&#xE9;synaptique principalement m&#xE9;di&#xE9;e par des neurones g&#xE9;ants forte de la PNC par l&#x26;#39;activation des r&#xE9;cepteurs M4 et &#xE9;ventuellement M2 qui, vraisemblablement, de contribuer &#xE0; prepulse inhibition.

Riesige Nervenzellen im kaudalen Pontinische Retikul&auml;re Kern (PnC) spielen eine entscheidende Rolle bei der Vermittlung der S&auml;ugetier-Startle-Reaktion. Eingabe erhalten sie von Cochlea, trigeminus und vestibul&auml;re Kerne und Projekt direkt an Motoneurones. Dar&uuml;ber hinaus integrieren sie modulierende Eingaben von verschiedenen Hirnregionen entweder verbessern oder Hemmung Startle Antworten. Wir stimuliert afferente Fasern von trigeminal und auditive und Muskarinischer Agonisten und Antagonisten angewendet, um ihre Wirkung auf exzitatorischen postsynaptisches aktuelle Amplituden, gepaart-Pulse-Verh&auml;ltnis und passive Membran Eigenschaften der PnC riesige Nervenzellen zu untersuchen. Die cholinerge Agonisten Carbachol und Muskarinischer Agonist-Oxotremorine deutlich reduziert exzitatorischen postsynaptisches aktuelle Amplituden und das gepaart-Pulse-Verh&auml;ltnis erh&ouml;ht. Carbachol reduziert zus&auml;tzlich den Membran-Widerstand der postsynaptisches PnC riesige Nervenzellen. Der Subtyp-spezifische Antagonisten AF-DX116 (lieber M2) und Tropicamid (M4 lieber) gegen sich aufgebracht, die Oxotremorine Wirkung angibt, die, dass diese Hemmung M4 und m&ouml;glicherweise M2-Rezeptor-Subtypen beteiligt sind. Die G-Protein-aktivierte nach innen Gleichrichterdiode Kalium Kanal Blocker Tertiapin-Q hatte keinen Einfluss auf die Oxotremorine-induzierte Hemmung der riesigen Neuronen. Unsere Ergebnisse zeigen eine haupts&auml;chlich presynaptically vermittelte starke Hemmung der PnC Riesen Neuronen durch Aktivierung der M4 und eventuell M2-Rezeptoren, die vermutlich zur Hemmung der prepulse beitragen.

Gigante neuroni del nucleo reticolare Pontina caudale (PnC) svolgono un ruolo cruciale nella mediazione della risposta di startle nei mammiferi. Essi ricevere input dal progetto e i nuclei cocleari, del trigemino e vestibolari direttamente a motoneurones. Il sottotipo specifici antagonisti AF-DX116 (preferendo M2) e tropicamide (M4 preferendo) antagonizzata l&#39;effetto Oxotremorina indicando che M4 e possibilmente sottotipi del recettore M2 sono coinvolti in questa inibizione. La G-proteina-attivato inward rettificando potassio canale blocker tertiapin-Q ha avuto alcun effetto sull&#39;inibizione Oxotremorina-indotta dei neuroni giganti. I nostri risultati mostrano un&#39;inibizione mediata principalmente presynaptically forte dei neuroni giganti PnC mediante l&#39;attivazione della M4 e possibilmente recettori M2 che presumibilmente contribuiscono alla inibizione prepulse.

Las neuronas gigantes en el n&#xFA;cleo caudal pontina reticular (PnC) juegan un papel crucial en la mediaci&#xF3;n de la respuesta de sobresalto mam&#xED;feros. Ellos reciben el aporte de los n&#xFA;cleos cocleares, vestibulares y del trig&#xE9;mino y del proyecto directamente a las motoneuronas. Por otra parte, se integran de entrada de modulador de diferentes regiones del cerebro, ya sea las respuestas de sobresalto que propician o inhiben. Un destacado la modulaci&#xF3;n de sobresalto es la inhibici&#xF3;n prepulso en un est&#xED;mulo no sorprendente de presentar antes de que el est&#xED;mulo de sobresalto inhibe una respuesta de sobresalto posterior. Varios estudios de comportamiento indican que esta inhibici&#xF3;n est&#xE1; mediada por los receptores muscar&#xED;nicos en el &#xE1;mbito de la PNC. Aqu&#xED;, se realiz&#xF3; un conjunto de c&#xE9;lulas patch-clamp grabaciones de PNC neuronas gigantes en agudas rodajas de cerebro de rata con el fin de examinar la inhibici&#xF3;n de los receptores muscar&#xED;nicos. El agonista colin&#xE9;rgico carbacol y la oxotremorina agonista muscar&#xED;nico redujo significativamente excitatorios postsin&#xE1;pticos amplitudes de corriente y el aumento de la relaci&#xF3;n de dos a dos pulsos. Carbacol adicionalmente reduce la resistencia de la membrana de las neuronas postsin&#xE1;pticas PnC gigantes. Los antagonistas del subtipo espec&#xED;fico de AF-DX116 (M2 prefiriendo) y tropicamida (M4 prefiriendo) antagoniza el efecto oxotremorina lo que indica que los subtipos de receptores M2 y M4 y posiblemente est&#xE1;n involucrados en esta inhibici&#xF3;n. El G-prote&#xED;na-activada hacia adentro bloqueador del canal de potasio rectificador tertiapin-Q no tuvo efecto sobre oxotremorina-inducida por la inhibici&#xF3;n de las neuronas gigantes. Nuestros resultados muestran una fuerte inhibici&#xF3;n presin&#xE1;ptica mediada por sobre todo de las neuronas PnC gigante por la activaci&#xF3;n de los receptores M2 y M4, posiblemente, que supuestamente contribuyen a la inhibici&#xF3;n prepulso.

Saccharomyces cerevisiae shows a marked preference for glucose and fructose, revealed by the repression of genes whose products are involved in processing other carbon sources. This response seems to be driven by sugar phosphorylation in the first steps of glycolysis rather than by the external sugar concentration. To gain a further insight into the role of the internal sugar signalling mechanisms, were measured the levels of upper intracellular glycolytic metabolites and adenine nucleotides in three mutant strains, HXT1, HXT7 and TM6*, with progressively reduced uptake capacities in comparison with the wild type. Reducing the rate of sugar consumption caused an accumulation of hexose phosphates upstream of the phosphofructokinase (PFK) and a reduction of fructose-1,6-bisphosphate levels. Mathematical modelling showed that these effects may be explained by changes in the kinetics of PFK and phosphoglucose isomerase. Moreover, the model indicated a modified sensitivity of the pyruvate dehydrogenase and the trichloroacetic acid cycle enzymes towards the NAD/NADH in the TM6* strain. The activation of the SNF1 sugar signalling pathway, previously observed in the TM6* strain, does not correlate with a reduction of the ATP : AMP ratio as reported in mammals. The mechanisms that may control the glycolytic rate at reduced sugar transport rates are discussed.

Reduzierung des Zuckerverbrauch verursacht eine Ansammlung von Hexose Phosphate stromaufw&auml;rts von der Phosphofructokinase (PFK1) und eine Verringerung der Fruktose-1, 6-bisphosphat-Ebenen. Mathematische Modellierung zeigten, dass diese Effekte durch &Auml;nderungen in der Kinetik der PFK und Phosphoglucose-Isomerase erkl&auml;rt werden k&ouml;nnen. The acoustic startle reflex is strongly inhibited by a moderate-intensity acoustic stimulus that precedes the startling stimulus by roughly 10-1000 ms (prepulse inhibition, PPI). At long interstimulus intervals (ISIs) of 100-1000 ms, PPI in rats is reduced by the muscarinic receptor antagonist scopolamine. Here, we studied the role of GABA receptors in PPI at full ISI ranges in both mice and rats. In B6 mice, PPI begins and ends at shorter ISIs (4 and 1000 ms, respectively) than in Wistar rats (8 and 5000 ms). The GABA(A) antagonist bicuculline (1 mg/kg i.p.) reduced PPI at ISIs near the peak of PPI in both rats and mice. The GABA(B) antagonist phaclofen (10 or 30 mg/kg i.p. in rats or mice, respectively) reduced PPI only at long ISIs, similar to the effects of the muscarinic antagonist scopolamine (1 mg/kg i.p.). The effects of phaclofen and scopolamine were additive in rats, suggesting independent effects of GABA(B) and muscarinic receptors. Patch-clamp recordings of startle-mediating PnC (nucleus reticularis pontis caudalis) giant neurons in rat slices show that EPSCs evoked by either trigeminal or auditory fiber stimulation were inhibited by the GABA(A/C) agonist muscimol or the GABA(B) agonist baclofen via postsynaptic mechanisms. Hyperpolarization of PnC neurons by muscimol was reversed with bicuculline, indicating that postsynaptic GABA(A) receptors strongly inhibit PnC giant neurons needed for startle. Therefore, GABA receptors on PnC giant neurons mediate a substantial part of PPI, with GABA(A) receptors contributing at the peak of PPI, and GABA(B) receptors adding to muscarinic effects on PPI at long ISIs.

Inorganic polyphosphate (poly-P) consists of just a chain of phosphate groups linked by high energy bonds. It is found in every organism and is implicated in a wide variety of cellular processes (e.g. phosphate storage, blood coagulation, and pathogenicity). Its metabolism has been studied mainly in bacteria while remaining largely uncharacterized in eukaryotes. It has recently been suggested that poly-P metabolism is connected to that of highly phosphorylated inositol species (inositol pyrophosphates). Inositol pyrophosphates are molecules in which phosphate groups outnumber carbon atoms. Like poly-P they contain high energy bonds and play important roles in cell signaling. Here, we show that budding yeast mutants unable to produce inositol pyrophosphates have undetectable levels of poly-P. Our results suggest a prominent metabolic parallel between these two highly phosphorylated molecules. More importantly, we demonstrate that DDP1, encoding diadenosine and diphosphoinositol phosphohydrolase, possesses a robust poly-P endopolyphosphohydrolase activity. In addition, we prove that this is an evolutionarily conserved feature because mammalian Nudix hydrolase family members, the three Ddp1 homologues in human cells (DIPP1, DIPP2, and DIPP3), are also capable of degrading poly-P.

Polifosfato inorg&#xE1;nico (poli-P) consta de s&#xF3;lo una cadena de grupos fosfato unidos por enlaces de alta energ&#xED;a. Se encuentra en todos los organismos y est&#xE1; implicado en una amplia variedad de procesos celulares (por ejemplo fosfato de almacenamiento, coagulaci&#xF3;n de la sangre, y la patogenicidad). Su metabolismo se ha estudiado principalmente en las bacterias sin dejar de ser en gran medida no caracterizados en las c&#xE9;lulas eucariotas. Recientemente se ha sugerido que la poli-P metabolismo est&#xE1; conectado a la de las especies altamente inositol fosforilado (pirofosfatos inositol). La identificaci&#xF3;n de un familiar conservado evolutivamente de Endopolyphosphatases polifosfato inorg&#xE1;nico

Inositol phosphates are key signaling molecules affecting a large variety of cellular processes. Inositol-polyphosphate multikinase (IPMK) is a central component of the inositol phosphate biosynthetic routes, playing essential roles during development. IPMK phosphorylates inositol 1,4,5-trisphosphate to inositol tetrakisphosphate and subsequently to inositol pentakisphosphate and has also been described to function as a lipid kinase. Recently, a catalytically inactive mammalian IPMK was reported to be involved in nutrient signaling by way of mammalian target of rapamycin and AMP-activated protein kinase. In yeast, the IPMK homologue, Arg82, is the sole inositol-trisphosphate kinase. Arg82 has been extensively studied as part of the transcriptional complex regulating nitrogen sensing, in particular arginine metabolism. Whether this role requires Arg82 catalytic activity has long been a matter of contention. In this study, we developed a novel method for the real time study of promoter strength in vivo and used it to demonstrate that catalytically inactive Arg82 fully restored the arginine-dependent transcriptional response. We also showed that expression in yeast of catalytically active, but structurally very different, mammalian or plant IPMK homologue failed to restore arginine regulation. Our work indicates that inositol phosphates do not regulate arginine-dependent gene expression.

Many lines of evidence suggest that a reciprocally interconnected network comprising the amygdala, ventral hippocampus (vHC), and medial prefrontal cortex (mPFC) participates in different aspects of the acquisition and extinction of conditioned fear responses and fear behavior. This could at least in part be mediated by direct connections from mPFC or vHC to amygdala to control amygdala activity and output. However, currently the interactions between mPFC and vHC afferents and their specific targets in the amygdala are still poorly understood. Here, we use an ex-vivo optogenetic approach to dissect synaptic properties of inputs from mPFC and vHC to defined neuronal populations in the basal amygdala (BA), the area that we identify as a major target of these projections. We find that BA principal neurons (PNs) and local BA interneurons (INs) receive monosynaptic excitatory inputs from mPFC and vHC. In addition, both these inputs also recruit GABAergic feedforward inhibition in a substantial fraction of PNs, in some neurons this also comprises a slow GABAB-component. Amongst the innervated PNs we identify neurons that project back to subregions of the mPFC, indicating a loop between neurons in mPFC and BA, and a pathway from vHC to mPFC via BA. Interestingly, mPFC inputs also recruit feedforward inhibition in a fraction of INs, suggesting that these inputs can activate dis-inhibitory circuits in the BA. A general feature of both mPFC and vHC inputs to local INs is that excitatory inputs display faster rise and decay kinetics than in PNs, which would enable temporally precise signaling. However, mPFC and vHC inputs to both PNs and INs differ in their presynaptic release properties, in that vHC inputs are more depressing. In summary, our data describe novel wiring, and features of synaptic connections from mPFC and vHC to amygdala that could help to interpret functions of these interconnected brain areas at the network level.

Ex vivo dissection of optogenetically activated mPFC and hippocampal inputs to neurons in the basolateral amygdala: implications for fear and emotional memory.

Prepulse inhibition (PPI) of startle occurs when intensity stimuli precede stronger startle-inducing stimuli by 10-1000 ms. PPI deficits are found in individuals with schizophrenia and other psychiatric disorders, and they correlate with other cognitive impairments. Animal research and clinical studies have demonstrated that both PPI and cognitive function can be enhanced by nicotine. PPI has been shown to be mediated, at least in part, by mesopontine cholinergic neurons that project to pontine startle neurons and activate muscarinic and potentially nicotine receptors (nAChRs). The subtypes and anatomical location of nAChRs involved in mediating and modulating PPI remain unresolved. We tested the hypothesis that nAChRs that are expressed by pontine startle neurons contribute to PPI. We also explored whether or not these pontine receptors are responsible for the nicotine enhancement of PPI. While systemic administration of nAChR antagonists had limited effects on PPI, PnC microinfusions of the non-α7nAChR preferring antagonist TMPH, but not of the α7nAChR antagonist MLA, into the PnC significantly reduced PPI. Electrophysiological recordings from startle-mediating PnC neurons confirmed that nicotine affects excitability of PnC neurons, which could be antagonized by TMPH, but not by MLA, indicating the expression of non-α7nAChR. In contrast, systemic nicotine enhancement of PPI was only reversed by systemic MLA and not by TMPH or local microinfusions of MLA into the PnC. In summary, our data indicate that non-α7nAChRs in the PnC contribute to PPI at stimulus intervals of 100 ms or less, whereas activation of α7nAChRs in other brain areas is responsible for the systemic nicotine enhancement of PPI. This is important knowledge for the correct interpretation of behavioral, preclinical, and clinical data as well as for developing drugs for the amelioration of PPI deficits and the enhancement of cognitive function.

Nicotine receptors mediating sensorimotor gating and its enhancement by systemic nicotine.

Increasing evidence suggests that parallel plastic processes in the amygdala involve inhibitory elements to control fear and extinction memory. GABAergic medial paracapsular intercalated cells (mpITCs) are thought to relay activity from basolateral nucleus (BLA) and prefrontal cortex to inhibit central amygdala output during suppression of fear. Recently, projection diversity and differential behavioral activation of mpITCs in distinct fear states suggest additional functions. Here, we show that mpITCs receive convergent sensory thalamic and cortical inputs that undergo fear learning-related changes and are dynamically modulated via presynaptic GABAB receptors recruited by GABA released from the mpITC network. Among mpITCs, we identify cells that inhibit but are also mutually activated by BLA principal neurons. Thus, mpITCs take part in fear learning-modulated feedforward and feedback inhibitory circuits to simultaneously control amygdala input and output nuclei. Our findings place mpITCs in a unique position to gate acquired amygdala-dependent behaviors via their direct sensory inputs.

Sensory inputs to intercalated cells provide fear-learning modulated inhibition to the basolateral amygdala.

Throughout life, fear learning is indispensable for survival and neural plasticity in the lateral amygdala underlies this learning and storage of fear memories. During development, properties of fear learning continue to change into adulthood, but currently little is known about changes in amygdala circuits that enable these behavioural transitions. In recordings from neurons in lateral amygdala brain slices from infant up to adult mice, we show that spontaneous and evoked excitatory and inhibitory synaptic transmissions mature into adolescence. At this time, increased inhibitory activity and signalling has the ability to restrict the function of excitation by presynaptic modulation, and may thus enable precise stimulus associations to limit fear generalization from adolescence onward. Our results provide a basis for addressing plasticity mechanisms that underlie altered fear behaviour in young animals.
