The in vivo behavior of shell cross-linked knedel-like (SCK) nanoparticles is shown to be tunable via a straightforward and versatile process that advances SCKs as attractive nanoscale carriers in the field of nanomedicine. Tuning of the pharmacokinetics was accomplished by grafting varied numbers of methoxy-terminated poly(ethylene glycol) (mPEG) chains to the amphiphilic block copolymer precursors, together with chelators for the radioactive tracer and therapeutic agent (64)Cu, followed by self-assembly into block copolymer micelles and chemical cross-linking throughout the shell regions. (64)Cu-radiolabeling was then performed to evaluate the SCKs in vivo by means of biodistribution experiments and positron emission tomography (PET). It was found that the blood retention of PEGylated SCKs could be tuned, depending on the mPEG grafting density and the nanoparticle surface properties. A semiquantitative model of the density of mPEG surface coverage as a function of in vivo behavior was applied to enhance the understanding of this system.

The gadolinium species present in a rat kidney following intravenous administration of a gadolinium-based magnetic resonance contrast agent (Optimarkâ„¢, Gadoversetamide injection) to a rat was examined in the present study. The major gadolinium species in the supernatant of the rat kidney tissue extracts was determined by reversed-phase liquid chromatography with online inductively coupled plasma optical emission spectrometry (HPLC-ICP-OES). The identity of the compound was established by liquid chromatography electrospray ionization mass spectrometry (LC-ESI-MS) detection. The principal gadolinium(III) complex in a rat kidney tissue extract was identified as Gd-DTPA-BMEA 24 Hrs and 7 days after a single intravenous injection of Optimarkâ„¢ (gadoversetamide; Gd-DTPA-BMEA) at a dose of 5 mmol Gd/kg body weight. The study demonstrated for the first time the feasibility of the use of two complementary techniques, HPLC-ICP-OES and HPLC-ESI-MS to study the in vivo behavior of gadolinium-based magnetic resonance contrast media.

Las especies presentes en el gadolinio un ri&#xF1;&#xF3;n de rata tras la administraci&#xF3;n intravenosa de un agente basado en gadolinio magn&#xE9;tica de contraste de resonancia (Optimark &#x2122;, la inyecci&#xF3;n de gadoversetamida) a una rata fue examinado en el presente estudio. Las especies principales de gadolinio en el sobrenadante de los extractos de tejido de ri&#xF1;&#xF3;n de rata se determin&#xF3; por cromatograf&#xED;a en fase inversa con l&#xED;quido en l&#xED;nea con plasma acoplado inductivamente espectrometr&#xED;a de emisi&#xF3;n &#xF3;ptica (HPLC-ICP-OES). La identidad del compuesto fue establecido por cromatograf&#xED;a l&#xED;quida de masas electrospray espectrometr&#xED;a de ionizaci&#xF3;n (LC-ESI-MS) de detecci&#xF3;n. El director de gadolinio (III) en un extracto de tejido del ri&#xF1;&#xF3;n de rata fue identificado como Gd-DTPA-BMEA 24 horas y 7 d&#xED;as despu&#xE9;s de una inyecci&#xF3;n intravenosa &#xFA;nica de Optimark &#x2122; (gadoversetamida, Gd-DTPA-BMEA) a una dosis de 5 mmol de Gd / kg de peso corporal. El estudio demostr&#xF3; por primera vez la viabilidad de la utilizaci&#xF3;n de dos t&#xE9;cnicas complementarias, HPLC-ICP-OES y HPLC-ESI-MS para estudiar el comportamiento in vivo de gadolinio medios de resonancia magn&#xE9;tica de contraste.
