In vertebrate embryos, most spinal commissural axons cross the ventral midline (VM) and project either alongside or significant distances away from the floor plate (FP). The upregulation of repulsive Robo1/2 receptors on postcrossing commissural axons, in mammals, presumably allows these axons to respond to the midline-associated repellents, Slit1-3, facilitating their expulsion from, and prohibiting their reentry into, the FP. Compelling data suggest that Robo3 represses Robo1/2 function on precrossing axons and that Robo1/2 inhibit attractive guidance receptors on postcrossing axons, thereby ensuring that decussated axons are selectively responsive to midline Slits. However, whether Robo1/2 expel decussated commissural axons from the VM and/or prevent their reentry into the FP has not been explicitly established in vivo. Furthermore, some commissural axons do not require Robo1/2 to elaborate appropriate contralateral projections in the mouse spinal cord. Here, we use unilateral in ovo electroporation together with Atoh1 and Neurog1 enhancer elements to visualize, and assess the consequences of manipulating Robo expression on, dl1 and dl2 chick commissural axons. In response to misexpressing a cytoplasmic truncation of Robo1 and/or Robo2, which should block all Robo-ligand interactions, postcrossing commissural axons extend alongside, but do not project away from or reenter the FP. In contrast, misexpression of full-length Robo2 prevents many commissural axons from crossing the VM. Together, these findings support key and selective in vivo roles for Robo receptors in presumably altering the responsiveness of decussated commissural axons and facilitating their expulsion from the VM within the chick spinal cord.

In the developing nervous system, pathfinding axons navigate through a series of intermediate targets in order to form synaptic connections. Vertebrate spinal commissural axons extend toward and across the floor plate (FP), a key intermediate target located at the ventral midline (VM). Subsequently, post-crossing commissural axons grow either alongside or significant distances away from the floor plate (FP), but never re-cross the VM. Consistent with this behavior, post-crossing commissural axons lose responsiveness to the FP-associated chemoattractants, Netrin-1 and SHH, and gain responsiveness to Slits, which are potent midline repellents, in vitro. In addition, the results of several in vivo studies suggest that the upregulation of Slit-binding repulsive Robo receptors, Robo1/2, alters the responsiveness of decussated commissural axons to midline guidance cues. Nevertheless, in vertebrates, it is unclear whether Robo1/2 are the sole or major repellent receptors responsible for driving these commissural axons away from the VM and preventing their re-entry into the FP. We recently re-visited these issues in the chick spinal cord by assessing the consequences of manipulating Robo expression on commissural axons in ovo. Our findings suggest that, at least in chick embryos, the upregulation of repulsive Robos on post-crossing axons alters the responsiveness of these axons to midline repellents and facilitates their expulsion from, but is not likely to have a significant role in preventing their re-entry into the VM.

Leaving the midline: how Robo receptors regulate the guidance of post-crossing spinal commissural axons.

Despite the general uniformity in cellular composition of the adult cerebellum (Cb), the expression of proteins such as ZebrinII/AldolaseC and the small heat shock protein HSP25 reveal striking patterns of parasagittal Purkinje cell (PC) stripes. Based on differences in the stripe configuration within subsets of lobules, the Cb can be further divided into four anterior-posterior transverse zones: anterior zone (AZ) = lobules I-V, central zone (CZ) = lobules VI-VII, posterior zone (PZ) = lobules VIII and anterior IX, and the nodular zone (NZ) = lobules posterior IX-X. Here we used whole-mount and tissue section immunohistochemistry to show that neurofilament heavy chain (NFH) expression alone divides all lobules of the mouse Cb into a complex series of parasagittal stripes of PCs. We revealed that the striped pattern of NFH in the vermis of the AZ and PZ was complementary to ZebrinII and phospholipase C ÃŸ3 (PLCÃŸ3), and corresponded to phospholipase C ÃŸ4 (PLCÃŸ4). In the CZ and NZ the stripe pattern of NFH was complementary to HSP25 and corresponded to PLCÃŸ3. The boundaries of the NFH stripes were not always sharply delineated. Instead, a gradual decrease in NFH expression was observed toward the edges of particular stripes, resulting in domains comprised of overlapping expression patterns. Furthermore, the terminal field distributions of mossy and climbing fibers had a complex but consistent topographical alignment with NFH stripes. In summary, NFH expression reveals an exquisite level of Cb stripe complexity that respects the transverse zone divisions and delineates an intricately patterned target field for Cb afferents.

Dans le CZ et NZ la rayure de l&#x26;#39;HNF &#xE9;tait compl&#xE9;mentaire &#xE0; Hsp25 et correspondait &#xE0; PLC&#xDF;3. Les limites de la rayures NFH ne sont pas toujours nettement d&#xE9;limit&#xE9;es. Dar&uuml;ber hinaus mussten die terminal-Feld-Distributionen moosig und Klettern Fasern eine komplexe, aber konsistente topographische Ausrichtung mit NFH Streifen. Zusammenfassend zeigt NFH Ausdruck ein exquisites Niveau der Cb-Streifen-Komplexit&auml;t, die respektiert die transversale Trennfl&auml;chen und schildert ein aufwendig gemusterten Zielfeld f&uuml;r Cb-afferenten.

Nonostante la generale uniformit&agrave; nella composizione cellulare del cervelletto adulti (Cb), l&#39;espressione di proteine quali ZebrinII/AldolaseC e la proteina shock termico piccolo HSP25 rivelano sorprendenti modelli di strisce di cellule di Purkinje (PC) parasagittal. Sulla base delle differenze nella configurazione della striscia all&#39;interno di sottoinsiemi di lobuli, il Cb pu&ograve; essere ulteriormente suddiviso in quattro zone trasversali antero-posteriore: zona anteriore (AZ) = lobuli I-V, zona centrale (CZ) = lobuli VI-VII, zona posteriore (PZ) = lobuli VIII e IX anteriore e la zona nodulare (NZ) = lobuli posteriore IX-X. Qui abbiamo usato intero-monta e tessuto sezione immunoistochimica per mostrare che l&#39;espressione neurofilamento pesante catena (NFH) solo divide tutti i lobuli del mouse Cb in una complessa serie di strisce parasagittal del PC. Abbiamo rivelato che il pattern a righe di NFH nel verme della AZ e PZ era complementare a ZebrinII e fosfolipasi C &szlig;3 (PLC&szlig;3) e corrispondeva alla fosfolipasi C &szlig;4 (PLC&szlig;4). In CZ e NZ il motivo a strisce di NFH era complementare al HSP25 e corrispondeva a PLC&szlig;3. I confini delle strisce NFH non erano sempre nettamente delineati. Invece, &egrave; stata osservata una graduale diminuzione nell&#39;espressione di NFH verso i bordi delle strisce particolare, risultando in domini composto da sovrapposizione pattern di espressione. Inoltre, le distribuzioni di campo terminale delle fibre muschiose e arrampicate ha avuto un allineamento topografico complesso ma coerenza con strisce NFH. In sintesi, espressione di NFH rivela un squisita livello di complessit&agrave; di banda Cb che rispetta le divisioni trasversali zona e delinea un campo di destinazione riccamente modellato per Cb afferenze.

A pesar de la uniformidad general de la composici&#xF3;n celular del cerebelo adulto (Cb), la expresi&#xF3;n de prote&#xED;nas tales como ZebrinII / AldolaseC y la prote&#xED;na de choque t&#xE9;rmico Hsp25 peque&#xF1;a revelan patrones en huelga de parasagital de c&#xE9;lulas de Purkinje (PC) rayas. Sobre la base de las diferencias en la configuraci&#xF3;n de banda dentro de los subgrupos de los l&#xF3;bulos, el Banco Central puede ser dividida en cuatro zonas anterior-posterior transversales: la zona anterior (AZ) = l&#xF3;bulos IV, zona central (CZ) = l&#xF3;bulos VI-VII, zona posterior (PZ ) = l&#xF3;bulos VIII y IX anterior, y la zona nodular (NZ) = l&#xF3;bulos posterior IX-X. Aqu&#xED; hemos utilizado todo el montaje y la inmunohistoqu&#xED;mica secci&#xF3;n de tejido para demostrar que la cadena pesada de los neurofilamentos (HNF) expresi&#xF3;n por s&#xED; sola divide a todos los l&#xF3;bulos de la Cb rat&#xF3;n en una compleja serie de rayas parasagitales de PC. Hemos puesto de manifiesto que el patr&#xF3;n rayado de HNF en el vermis del AZ y PZ era complementaria ZebrinII y la fosfolipasa C SS3 (PLC&#xDF;3), y correspondi&#xF3; a la fosfolipasa C SS4 (PLC&#xDF;4). En la Rep&#xFA;blica Checa y Nueva Zelanda el patr&#xF3;n de rayas de la HNF es complementaria a la Hsp25 y correspondi&#xF3; a PLC&#xDF;3. Los l&#xED;mites de las rayas de HNF no siempre claramente delineada. En su lugar, una disminuci&#xF3;n gradual en la expresi&#xF3;n de HNF se observ&#xF3; hacia los bordes de rayas particulares, dando lugar a dominios compuestos de la superposici&#xF3;n de los patrones de expresi&#xF3;n. Por otra parte, las distribuciones de campo terminales de musgo y las fibras trepadoras ten&#xED;a una alineaci&#xF3;n topogr&#xE1;fico complejo, pero coherente con las rayas NFH. En resumen, la expresi&#xF3;n de HNF revela un nivel de complejidad exquisita banda Cb que respete las divisiones de zonas transversales y delimita un campo de destino intrincado patr&#xF3;n de aferentes Cb.

Neural circuits are organized into complex topographic maps. Although several neuroanatomical and genetic tools are available for studying circuit architecture, a limited number of methods exist for reliably revealing the global patterning of multiple topographic projections. Here we used wheat germ agglutinin (WGA) conjugated to Alexa 555 and 488 for dual color fluorescent mapping of parasagittal spinocerebellar topography in three dimensions. Using tissue section and wholemount imaging we show that WGA-Alexa tracers have three main characteristics that make them ideal tools for analyses of neural projection topography. First, the intense brightness of Alexa fluorophores allows multi-color imaging of patterned afferent projections in wholemount preparations. Second, WGA-Alexa tracers robustly label the entire trajectory of developing and adult projections. Third, long tracts such as the adult spinocerebellar tract can be traced in less than 6Â h. Moreover, using WGA-Alexa tracers we resolved a level of complexity in the compartmentalized topography of the spinocerebellar projection map that has never before been appreciated. In summary, we introduce versatile tracers for rapidly labeling multiple topographic projections in three dimensions and uncover wiring complexities in the spinocerebellar map.

The cerebellum (Cb) of mammals and birds consists of an evolutionarily conserved map defined by Purkinje cell (PC) protein expression. In mice, ZebrinII/aldolaseC is expressed in a striking array of stripes in lobules I-V (anterior zone; AZ) and VIII-anterior IX (posterior zone; PZ), whereas the small heat shock protein 25 (HSP25) is expressed in stripes in lobules VI-VII (central zone, CZ) and posterior IX-X (nodular zone, NZ). Little is known about whether molecularly defined afferent subsets terminate within specific PC stripes or whether their topography is conserved across species. Using immunohistochemistry, we demonstrate in adult mice and rats that cocaine- and amphetamine-regulated transcript (CART) expression can be used to partition sensory-motor projections into complex topographic maps. We found that in mice CART was expressed in climbing fiber bands that generally corresponded to the pattern of HSP25-expressing PCs in the CZ/NZ. In contrast, CART was expressed in climbing fiber bands in all four transverse zones of the rat Cb. Within the rat AZ/PZ, climbing fibers terminated selectively within the dendrites of ZebrinII-immunoreactive PCs. In additional experiments, we observed CART expression in loose clusters of spinocerebellar mossy fibers in the mouse AZ/PZ, whereas in rat CART immunoreactive mossy fibers terminated predominantly in the CZ/NZ. We conclude that, although the overall topography of CART-expressing afferents is restricted within a conserved map of PC stripes and transverse zones, their termination patterns also reflect species-specific compartmental features.

Le cervelet (Cb) de mammif&#xE8;res et d&#x26;#39;oiseaux comprend une carte conserv&#xE9;e d&#xE9;fini par l&#x26;#39;&#xE9;volution, cellules de Purkinje (PC) expression de la prot&#xE9;ine. Chez la souris, ZebrinII / aldolaseC est exprim&#xE9; dans un tableau frappant de rayures dans les lobules IV (ant&#xE9;rieure zone; AZ) et VIII-IX ant&#xE9;rieure (zone post&#xE9;rieure; PZ), tandis que la prot&#xE9;ine de choc thermique petite 25 (Hsp25) est exprim&#xE9;e en bandes dans les lobules VI-VII (zone centrale, CZ) et post&#xE9;rieure IX-X (nodulaire zone, NZ). Nous avons constat&#xE9; que chez les souris CART a &#xE9;t&#xE9; exprim&#xE9; dans l&#x26;#39;escalade des bandes de fibres qui correspondait g&#xE9;n&#xE9;ralement &#xE0; la structure de Hsp25-exprimant PC dans le CZ / NZ. En revanche, CART a &#xE9;t&#xE9; exprim&#xE9; dans l&#x26;#39;escalade des bandes de fibres dans les quatre zones transversales de la Cb rat. Dans le rat AZ / PZ, fibres grimpantes fin s&#xE9;lectivement dans les dendrites de ZebrinII-immunor&#xE9;actives PC. Dans d&#x26;#39;autres exp&#xE9;riences, nous avons observ&#xE9; l&#x26;#39;expression CART en grappes l&#xE2;ches de spinoc&#xE9;r&#xE9;belleuses fibres moussues dans le AZ souris / PZ, alors que dans CART rat immunor&#xE9;actifs fibres moussues fin principalement dans la CZ / NZ. Nous concluons que, bien que la topographie g&#xE9;n&#xE9;rale du CART exprimant aff&#xE9;rences est restreinte dans une carte conserv&#xE9;e de rayures PC et les zones transversales, leurs modes de terminaison refl&#xE8;tent &#xE9;galement sp&#xE9;cifiques aux esp&#xE8;ces caract&#xE9;ristiques parcellaires.

Das Kleinhirn (Cb) von S&auml;ugetieren und V&ouml;geln besteht aus eine evolution&auml;r konservierte Karte von Purkinje Zelle (PC)-Protein-Ausdruck definiert. ZebrinII/AldolaseC in den M&auml;usen dr&uuml;ckt sich in einem markanten Array aus Streifen in L&auml;ppchen I-V (anterior Zone; AZ) und VIII-anterior IX (posterior Zone; PZ), w&auml;hrend das kleine Hitze-Schock-Protein 25 (HSP25) in Streifen in L&auml;ppchen, VI-VII (zentrale Zone, CZ) und posterior IX-X (nodul&auml;re Zone, NZ) ausgedr&uuml;ckt wird. Wenig ist bekannt &uuml;ber ob molekular definierte afferente Teilmengen innerhalb bestimmter PC Streifen beendet oder ob ihre Topographie Arten konserviert wird. Mit Immunohistochemistry, zeigen wir in Erwachsenen M&auml;usen und Ratten, die Kokain und Amphetamin-regulierte Abschrift (Einkaufswagen) Ausdruck zu Partition sensorisch-motorischen Projektionen in komplexen Topographische Karten verwendet werden kann. Wir fanden, dass bei M&auml;usen CART in Klettern Faser-Bands, die im allgemeinen entsprach dem Muster der HSP25-Ausdruck von PCs in der CZ/NZ ausgedr&uuml;ckt wurde. Im Gegensatz dazu wurde CART ausgedr&uuml;ckt Klettern Faser B&auml;nder in allen vier quer Zonen der Ratte Cb. Innerhalb der Ratte AZ/PZ Klettern Fasern selektiv innerhalb beendet die Dendriten von ZebrinII-immunreaktivem PCs. In weiteren Experimenten beobachteten wir CART Ausdruck in lockeren B&uuml;scheln spinozerebell&auml;re moosige Fasern in der Maus AZ/PZ, w&auml;hrend bei Ratte CART immunreaktivem moosige Fasern vorwiegend in der CZ/NZ beendet. Wir schlie&szlig;en, dass, obwohl die allgemeine Topographie der afferenten CART-Ausdruck innerhalb einer konservierten Karte von PC Streifen und quer Zonen eingeschr&auml;nkt ist, ihre K&uuml;ndigung-Muster auch artspezifische Bicompartmental Funktionen wider.

Nei topi, ZebrinII/aldolaseC &egrave; espresso in una gamma sorprendente di strisce in lobuli I-V (zona anteriore; AZ) e IX VIII-anteriore (zona posteriore; PZ), mentre la proteina shock termico piccolo 25 (HSP25) &egrave; espressa in strisce in lobuli VI-VII (zona centrale, CZ) e posteriore IX-X (zona nodulare, NZ). Abbiamo scoperto che nei topi carrello &egrave; stato espresso in arrampicata su fasce di fibra che in genere corrispondevano al modello di esprimere HSP25 pz in CZ/NZ. Al contrario, carrello &egrave; stato espresso in arrampicata bande di fibra in tutte le quattro zone trasversale del ratto Cb. All&#39;interno il ratto AZ/PZ, arrampicata fibre terminato selettivamente all&#39;interno i dendriti dei PC ZebrinII-immunoreattive. In ulteriori esperimenti, abbiamo osservato espressione di carrello in loose ammassi di fibre muschiose spinocerebellare nel topo AZ/PZ, mentre nel ratto immunoreattive fibre muschiose carrello terminato prevalentemente in CZ/NZ. Possiamo concludere che, sebbene la topografia generale delle afferenze esprimendo CART &egrave; limitata all&#39;interno conservata in una mappa di zone trasversali e strisce di PC, anche sui modelli di terminazione riflettono caratteristiche specie-compartimentali.

En ratones, ZebrinII / aldolaseC se expresa en una matriz sorprendente de rayas en l&#xF3;bulos IV (zona anterior; AZ) y VIII-IX anterior (posterior zona; PZ), mientras que la prote&#xED;na de choque t&#xE9;rmico peque&#xF1;o 25 (Hsp25) se expresa en rayas en lobulillos VI-VII (zona central, CZ) y posterior IX-X (nodular zona, Nueva Zelanda). The cerebellum receives sensory signals from spinocerebellar (lower limbs) and dorsal column nuclei (upper limbs) mossy fibers. In the cerebellum, mossy fibers terminate in bands that are topographically aligned with stripes of Purkinje cells. While much is known about the molecular heterogeneity of Purkinje cell stripes, little is known about whether mossy fiber compartments have distinct molecular profiles. Here, we show that the vesicular glutamate transporters VGLUT1 and VGLUT2, which mediate glutamate uptake into synaptic vesicles of excitatory neurons, are expressed in complementary bands of mossy fibers in the adult mouse cerebellum. Using a combination of immunohistochemistry and anterograde tracing, we found heavy VGLUT2 and weak VGLUT1 expression in bands of spinocerebellar mossy fibers. The adjacent bands, which are in part comprised of dorsal column nuclei mossy fibers, strongly express VGLUT1 and weakly express VGLUT2. Simultaneous injections of fluorescent tracers into the dorsal column nuclei and lower thoracic-upper lumbar spinal cord revealed that upper and lower limb sensory pathways innervate adjacent VGLUT1/VGLUT2 parasagittal bands. In summary, we demonstrate that VGLUT1 and VGLUT2 are differentially expressed by dorsal column nuclei and spinocerebellar mossy fibers, which project to complementary cerebellar bands and respect common compartmental boundaries in the adult mouse cerebellum.

Hier zeigen wir, dass die vesikul&auml;re Glutamat-Transporter VGLUT1 und VGLUT2, die Aufnahme von Glutamat in synaptische Vesikel von exzitatorischen Neuronen zu vermitteln, erg&auml;nzende Bands moosige Fasern im adulten Maus Kleinhirn ausgedr&uuml;ckt werden. Mit einer Kombination von Immunhistochemie und Anterograde Tracing, fanden wir schwere VGLUT2 und schwachen VGLUT1 Ausdruck in den Bands von spino moosige Fasern. Die angrenzenden Bands, die teilweise aus dorsalen Spalte Kerne moosige Fasern bestehen, schnelles stark VGLUT1 und schwach ausdr&uuml;ckliche VGLUT2. Gleichzeitige Injektionen von Fluoreszenz Tracer in der dorsalen Spalte Kerne und unteren thorakalen-obere lumbales R&uuml;ckenmark ergab, dass die oberen und unteren Gliedma&szlig;en sensorische Bahnen neben VGLUT1/VGLUT2 parasagittale B&auml;nder anregen. Zusammenfassend l&auml;sst sich sagen beweisen wir, dass VGLUT1 und VGLUT2 differenziell ausgedr&uuml;ckt werden von der dorsalen Spalte Kerne und spinozerebell&auml;re moosigen Fasern, das Projekt zu erg&auml;nzenden zerebell&auml;re Bands und respektieren gemeinsame Bicompartmental Grenzen im adulten Maus Kleinhirn.

Il cervelletto riceve segnali sensoriali da spino-cerebellare (arti inferiori) e fibre muschiose di colonna dorsale nuclei (arti superiori). Nel cervelletto, muschiosi fibre terminano in fasce che sono topograficamente allineati con strisce di cellule di Purkinje. Mentre molto &egrave; conosciuto circa l&#39;eterogeneit&agrave; molecolare delle cellule di Purkinje strisce, poco si sa circa se compartimenti muschioso fibra sono distinti profili molecolari. Qui, ci mostra che i trasportatori di glutammato vescicolare, VGLUT1 e VGLUT2, che mediare assorbimento glutammato nelle vescicole sinaptiche dei neuroni eccitatori, sono espresse nelle bande complementari di fibre muschiose nel cervelletto di topo adulto. Utilizzando una combinazione di immunoistochimica e tracciatura anterograda, abbiamo trovato VGLUT2 pesante e debole espressione VGLUT1 in bande di fibre muschiose spino-cerebellare. Le bande adiacenti, che sono in parte costituite da fibre muschiose colonna dorsale nuclei, esprimono fortemente VGLUT1 e debolmente VGLUT2 express. Iniezioni simultanee di traccianti fluorescenti in nuclei della colonna dorsale e lombare toracico superiore del midollo spinale inferiore ha rivelato che percorsi sensoriali degli arti superiori e inferiori innervano VGLUT1/VGLUT2 parasagittal bande adiacenti. In sintesi, ci dimostrano che VGLUT1 e VGLUT2 sono differenzialmente espressi dalla colonna dorsale nuclei e fibre muschiose spino-cerebellare, che proiettano complementare bande cerebellare e rispettano i confini compartimentali comuni nel cervelletto di topo adulto.

El cerebelo recibe las se&#xF1;ales sensoriales de espinocerebelosa (miembros inferiores) y n&#xFA;cleos de la columna dorsal (miembros superiores) fibras musgosas. En el cerebelo, las fibras musgosas terminan en bandas que est&#xE1;n topogr&#xE1;ficamente alineados con las rayas de las c&#xE9;lulas de Purkinje. Aunque se sabe mucho sobre la heterogeneidad molecular de rayas c&#xE9;lulas de Purkinje, poco se sabe acerca de si los compartimientos cubiertos de musgo de fibra tienen diferentes perfiles moleculares. Aqu&#xED;, mostramos que los transportadores de glutamato vesicular VGLUT1 y VGLUT2, que median la captaci&#xF3;n de glutamato en ves&#xED;culas sin&#xE1;pticas de las neuronas excitadoras, se expresan en las bandas complementarias de las fibras musgosas del cerebelo de rat&#xF3;n adulto.
