Respiratory syncytial virus (RSV) causes bronchiolitis, the main cause of infantile hospitalization. Immunity against reinfection is poor, and there is great interest in boosting vaccine responses using live vectors expressing host cytokines. We therefore constructed a recombinant RSV expressing murine interleukin 18 (RSV/IL-18), a cytokine capable of inducing strong antiviral immune responses. In vitro RSV/IL-18 replicated at wild-type levels and produced soluble IL-18. In naÃ¯ve BALB/c mice, RSV/IL-18 infection significantly increased both IL-18 mRNA and protein and attenuated the peak viral load 3-fold. Despite a reduced viral load, RSV/IL-18 infection caused a biphasic weight loss at days 2 and 6 postinfection that was not seen in wild-type infection. Day 2 disease was associated with enhanced pulmonary natural killer (NK) cell numbers and activity and was prevented by NK cell depletion during infection; day 6 disease was correlated with CD8 T-cell recruitment and was enhanced by NK cell depletion. IL-18 expression during priming also enhanced RSV-specific antibody responses and T-cell responses on secondary RSV infection. Therefore, while IL-18 boosted antiviral immunity and reduced the viral load, its coexpression worsened disease. This is the first recombinant RSV with this property, and these are the first studies to demonstrate that NK cells can induce pathology during pulmonary viral infections.

Virus sincicial respiratorio (VSR) causa bronquiolitis, la principal causa de hospitalizaci&oacute;n infantil. Inmunidad contra la reinfecci&oacute;n es pobre, y hay gran inter&eacute;s en impulsar las respuestas de la vacuna usando vectores vivos expresando citoquinas de anfitri&oacute;n. Por lo tanto, construimos una RSV recombinante expresando murino interleukin 18 (RSV/IL-18), una citoquina capaces de inducir la respuesta inmune antiviral fuerte. RSV/IL-18 in vitro hab&iacute;a replicado en los niveles de tipo salvaje y producido soluble IL-18. En ratones BALB/c de ingenuo, infecci&oacute;n de RSV/IL-18 significativamente hab&iacute;a aumentado tanto IL-18 mRNA y prote&iacute;na y aten&uacute;a la carga viral del pico 3-fold. A pesar de una menor carga viral, infecci&oacute;n de RSV/IL-18 caus&oacute; una p&eacute;rdida de peso bif&aacute;sica en postinfection d&iacute;as 2 y 6 que no fue visto en salvaje-tipo infecci&oacute;n. D&iacute;a 2 enfermedad se asoci&oacute; con un mayor pulmonar natural killer (NK) n&uacute;mero de c&eacute;lulas y actividad y fue prevenida por el agotamiento de la c&eacute;lula NK durante la infecci&oacute;n; d&iacute;a 6 enfermedad se correlacion&oacute; con el reclutamiento de c&eacute;lulas T CD8 y fue realzada por el agotamiento de c&eacute;lulas NK. Expresi&oacute;n de IL-18 durante el cebado tambi&eacute;n hab&iacute;a mejorado las respuestas del anticuerpo espec&iacute;fico RSV y respuestas de c&eacute;lulas T en la infecci&oacute;n secundaria de RSV. Por lo tanto, si bien IL-18 impuls&oacute; la inmunidad antiviral y reduce la carga viral, su coexpresi&oacute;n empeorado la enfermedad. Se trata de la primera RSV recombinante con esta propiedad, y &eacute;stos son los primeros estudios para demostrar que las c&eacute;lulas NK pueden inducir patolog&iacute;a durante infecciones virales pulmonares.

Coexpresi&oacute;n de interleucina 18 durante la infecci&oacute;n por virus sincitial respiratorio da lugar a mayor enfermedad mediada por c&eacute;lulas asesinas naturales.
