One of the most challenging issues in the characterization of magnetic materials is to obtain a quantitative analysis on the nanometer scale. Here we describe how electron magnetic circular dichroism (EMCD) measurements using the transmission electron microscope can be used for that purpose, utilizing reciprocal space maps. Applying the EMCD sum rules, an orbital to spin moment ratio of mL/mS=0.08+/-0.01 is obtained for Fe, which is consistent with the commonly accepted value. Hence, we establish EMCD as a quantitative element-specific technique for magnetic studies, using a widely available instrument with superior spatial resolution.

Quantitative magnetic information from reciprocal space maps in transmission electron microscopy.

Calcium phosphate ceramic coatings, applied on surfaces of metallic and polymeric biomaterials, can improve their performance in bone repair and regeneration. Spider silk is biocompatible, strong and elastic, and hence an attractive biomaterial for applications in connective tissue repair. Recently, artificial spider silk, with mechanical and structural characteristics similar to those of native spider silk, has been produced from recombinant minispidroins. In the present study, supersaturated simulated body fluid was used to deposit calcium phosphate coatings on recombinant spider silk fibres. The mineralization process was followed in time using scanning electron microscopy equipped with an energy dispersive x-ray (EDX) detector and Raman spectroscope. Focused ion beam technology was used to produce a cross section of a coated fibre, which was further analysed by EDX. Preliminary in vitro experiments using a culture of bone marrow-derived human mesenchymal stem cells (hMSCs) on coated fibres were also performed. This study showed that recombinant spider silk fibres were successfully coated with a homogeneous and thick crystalline calcium phosphate layer. In the course of the mineralization process from modified simulated body fluid, sodium chloride crystals were first deposited on the silk surface, followed by the deposition of a calcium phosphate layer. The coated silk fibres supported the attachment and growth of hMSCs.

Electron magnetic chiral dichroism (EMCD) is an emerging tool for quantitative measurements of magnetic properties using the transmission electron microscope (TEM), with the possibility of nanometer resolution. The geometrical conditions, data treatment and electron gun settings are found to influence the EMCD signal. In this article, particular care is taken to obtain a reliable quantitative measurement of the ratio of orbital to spin magnetic moment using energy filtered diffraction patterns. For this purpose, we describe a method for data treatment, normalization and selection of mirror axis. The experimental results are supported by theoretical simulations based on dynamical diffraction and density functional theory. Special settings of the electron gun, so called telefocus mode, enable a higher intensity of the electron beam, as well as a reduction of the influence from artifacts on the signal. Using these settings, we demonstrate the principle of acquiring real space maps of the EMCD signal. This enables advanced characterization of magnetic materials with superior spatial resolution.

In questo articolo, &egrave; prestare particolare attenzione per ottenere una misurazione quantitativa affidabile del rapporto tra orbitali di spin momento magnetico utilizzando modelli di diffrazione energia filtrata. Per questo scopo, si descrive un metodo per il trattamento dei dati, normalizzazione e selezione dell&#39;asse specchio. Las condiciones geom&eacute;tricas, configuraci&oacute;n de datos tratamiento y arma de electr&oacute;n se encuentra para influir en la se&ntilde;al EMCD. En este art&iacute;culo, se toma especial cuidado para obtener una medida cuantitativa fiable de la relaci&oacute;n de orbital al momento magn&eacute;tico, utilizando patrones de difracci&oacute;n de energ&iacute;a filtrada de la vuelta. Para ello, se describe un m&eacute;todo para el tratamiento de los datos, la normalizaci&oacute;n y la selecci&oacute;n del eje del espejo. Los resultados experimentales son apoyados por simulaciones te&oacute;ricas basadas en la difracci&oacute;n din&aacute;mico y la teor&iacute;a funcional de la densidad. Ajustes especiales de la arma de electr&oacute;n, denominados telefocus modo, permiten una mayor intensidad de los haces de electrones, as&iacute; como una reducci&oacute;n de la influencia de los artefactos en la se&ntilde;al. Estos par&aacute;metros, demostramos el principio de adquirir mapas de espacio real de la se&ntilde;al EMCD. The present work provides the first real-space analysis of nanobead-DNA coil interactions. Immobilization of oligonucleotide-functionalized magnetic nanobeads in rolling circle amplified DNA-coils was studied by complex magnetization measurements and transmission electron microscopy (TEM), and a statistical analysis of the number of beads hybridized to the DNA-coils was performed. The average number of beads per DNA-coil using the results from both methods was found to be around 6 and slightly above 2 for samples with 40 and 130 nm beads, respectively. The TEM analysis supported an earlier hypothesis that 40 nm beads are preferably immobilized in the interior of DNA-coils whereas 130 nm beads, to a larger extent, are immobilized closer to the exterior of the coils. The methodology demonstrated in the present work should open up new possibilities for characterization of interactions of a large variety of functionalized nanoparticles with macromolecules, useful for gaining more fundamental understanding of such interactions as well as for optimizing a number of biosensor applications.

The energy-filtered transmission electron microscopy (EFTEM) technique has been used to study ion-exchange processes in conductive polymer composite nanofibers. The elemental distributions of carbon, nitrogen, oxygen, chlorine, boron, phosphorus, molybdenum, and sulfur within polypyrrole-cellulose nanofibers, used as potential controlled electrochemical solid phase extraction media, have been studied by EFTEM. The distribution of ions within the polypyrrole-cellulose nanofibers and the penetration depth of ions into the material as a function of the size and charge of the latter were investigated. Further, the spatial distribution of single stranded DNA hexamers inside polypyrrole-cellulose nanofibers was mapped subsequent to the electrochemically controlled extraction of DNA from a borate buffer solution. The results show that the EFTEM mapping technique provides unpreceded possibilities for studies of the distribution of ions inside conductive polymer composites.

Die Verteilung der Ionen innerhalb der Polypyrrol-Cellulose-Nanofasern und die Eindringtiefe der Ionen in das Material als eine Funktion der Gr&ouml;&szlig;e und Ladung der letzteren wurden untersucht. Le distribuzioni elementali di carbonio, azoto, ossigeno, cloro, boro, fosforo, molibdeno e zolfo entro le nanofibre di polipirrolo-cellulosa, utilizzate come potenziale controllato media estrazione in fase solida elettrochimico, sono state studiate da EFTEM. Las distribuciones elementales de carbono, nitr&oacute;geno, ox&iacute;geno, cloro, boro, f&oacute;sforo, molibdeno y azufre dentro de polypyrrole-celulosa nanofibras, utilizado como potencial hab&iacute;a controlado por medios de la extracci&oacute;n de fase s&oacute;lida electroqu&iacute;mica, han sido estudiadas por EFTEM. La distribuci&oacute;n de iones dentro de las nanofibras polypyrrole-celulosa y la profundidad de la penetraci&oacute;n de iones en el material como una funci&oacute;n del tama&ntilde;o y la carga de estos &uacute;ltimos fueron investigados. Adem&aacute;s, la distribuci&oacute;n espacial de solo hexamers trenzado de ADN dentro de polypyrrole-celulosa nanofibras fue trazada con posterioridad a la extracci&oacute;n electroqu&iacute;micamente controlada de ADN de un tamp&oacute;n de borato. Disaccharides are, in general, the first choice as formulation compounds when freeze-drying microorganisms. Although polysaccharides and other biopolymers are considered too large to stabilise and interact with cell components in the same beneficial way as disaccharides, polymers have been reported to support cell survival. In the present study we compare the efficiency of sucrose and the polymers Ficoll, hydroxyethylcellulose, hydroxypropylmethylcellulose and polyvinylalcohol to support the survival of three bacterial strains during freeze drying. The initial osmotic conditions were adjusted to be similar for all formulations. Formulation characterisation was used to interpret the impact that different compound properties had on cell survival.

Obgleich Polysaccharide und andere Biopolymere zu gro&szlig; betrachtet sind, zu stabilisieren und die Interaktion mit dem Zellbestandteile vorteilhaft wie Disaccharide, wurde Polymere berichtet, Zelle &uuml;berleben zu unterst&uuml;tzen. In der vorliegenden Studie vergleichen wir die Effizienz der Saccharose und die Polymere Ficoll, Hydroxyethylcellulose, Hydroxypropylmethylcellulose und Polyvinylalcohol zu unterst&uuml;tzen, das &Uuml;berleben der drei Bakterienst&auml;mme w&auml;hrend der Trocknung Einfrieren. Sebbene polisaccaridi e altri biopolimeri sono considerati troppo grandi per stabilizzare e interagire con i componenti delle cellule allo stesso modo benefico come disaccaridi, polimeri sono stati segnalati per sostenere la sopravvivenza delle cellule. Nel presente studio abbiamo confrontare l&#39;efficienza di saccarosio e polimeri di Ficoll, hydroxyethylcellulose, idrossipropilmetil e polyvinylalcohol per sostenere la sopravvivenza di tre ceppi batterici durante liofilizzazione. Le condizioni iniziali osmotiche sono state adeguate per essere simile per tutte le formulazioni. Impacto de las propiedades de la matriz sobre la supervivencia de bacterias liofilizadas.

Gold nanoparticles (AuNPs) coated with stabilizing molecular monolayers are utilized in areas ranging from life sciences to nanoelectronics. Here we present a novel and facile one-pot single phase procedure for the preparation of stable AuNPs with good dispersity, which are coated with Î±,Ï‰-alkanedithiols whose outer Ï‰-thiol is protected by a triphenylmethyl group. Using dielectrophoresis we were able to trap these AuNPs, coated with Ï‰-thiol protecting groups, in a 20 nm gold electrode nanogap. The Ï‰-thiol group was then deprotected under acidic conditions in situ once the AuNPs were correctly positioned in the device. The subsequent deprotection resulted in an increase of conductance by up to 3 orders of magnitude, indicating that the isolated dithiol-coated AuNPs were fused into a covalently bonded network with AuNP-molecule-AuNP as well as electrode-molecule-AuNP linkages. Furthermore, complete characterization of the AuNP surface-bonded alkanedithiols was achieved using a series of one- and two-dimensional NMR spectroscopy techniques. Our spectra of the molecule-coated AuNPs show well-resolved signals, only slightly broader than for free molecules in solution, which is in contrast to many earlier reported NMR spectral data of molecules attached to AuNPs. Complementary diffusion NMR spectroscopic experiments were performed to prove that the observed alkanedithiols are definitely surface-bonded species and do not exist in free and unattached form.

Formation and NMR spectroscopy of Ï‰-thiol protected Î±,Ï‰-alkanedithiol-coated gold nanoparticles and their usage in molecular charge transport junctions.

Magnetic multilayered, onion-like, heterostructured nanoparticles are interesting model systems for studying magnetic exchange coupling phenomena. In this work, we synthesized heterostructured magnetic nanoparticles composed of two, three, or four components using iron oxide seeds for the subsequent deposition of manganese oxide. The MnO layer was allowed either to passivate fully in air to form an outer layer of Mn(3)O(4) or to oxidize partially to form MnO|Mn(3)O(4) double layers. Through control of the degree of passivation of the seeds, particles with up to four different magnetic layers can be obtained (i.e., FeO|Fe(3)O(4)|MnO|Mn(3)O(4)). Magnetic characterization of the samples confirmed the presence of the different magnetic layers.

Two-, three-, and four-component magnetic multilayer onion nanoparticles based on iron oxides and manganese oxides.

The use of single molecules as building blocks for practical electronic devices and sensors has high potential for novel applications due to the versatility of electronic properties of the molecules. Nano-sized molecules offer great potential for further miniaturization of electronic devices. We describe a method where such molecules are used to bridge a nanoparticles-nanoelectrode interface and thus determine the electrical properties of such a junction. We describe in detail the fabrication of the platform, its functionalization with molecules, and the basics of the electrical measurements. This platform has been shown to guide electrical current through a few molecules. The versatility of such nanoparticle-molecule-nanoelectrode heterojunctions makes this platform suitable for both basic molecular electronics measurements and also for molecular sensing devices in biological and medical applications.

Nanoparticle bridges for studying electrical properties of organic molecules.

The focused-ion-beam (FIB) is the method of choice for site-specific sample preparation for Transmission Electron Microscopy (TEM) in material sciences. A lamella can be physically lifted out from a specific region of a bulk specimen with submicrometer precision and thinned to electron transparency for high-resolution imaging in the TEM. The possibility to use this tool in life sciences applications has been limited by the lack of lift-out capabilities at the cryogenic temperatures often needed for biological samples. Conventional cryo-TEM sample preparation is mostly based on ultramicrotomy, a procedure that is not site-specific and known to produce artifacts. Here we demonstrate how a cooled nanomanipulator and a custom-built transfer station can be used to achieve cryo-preparation of TEM samples with the FIB, enabling high-resolution investigation of frozen-hydrated specimens in the TEM.
