银种子介导生长法制备金纳米星

我们采用银种子介导生长法合成了星形金纳米星。纳米星的直径范围为200至300 nm,尖端数量在7到10之间。这些纳米粒子具有宽泛的表面等离子体共振模式,其峰值位于近红外区。

摘要

纳米级胶体的物理、化学和光学性质取决于其材料组成、尺寸和形状1-5。目前人们对其在光热消融、药物递送及其他多种生物医学应用中的使用具有浓厚兴趣6。金因其低毒性而被特别广泛使用7-9。金属纳米胶体的一个特性是可表现出强烈的表面等离子体共振10。表面等离子体共振模式的峰值取决于金属纳米胶体的结构和组成。由于表面等离子体共振模式由光激发,因此需要其峰值吸收位于近红外区域,以匹配生物组织透光率最高的波长范围11, 12

我们介绍一种合成星形胶体金的方法,也称为星形纳米粒子13-15或纳米星16。该方法基于含有银种子的溶液,银种子作为胶体金各向异性生长的成核剂17-22。对所得胶体金进行扫描电子显微镜(SEM)分析显示,70%的纳米结构为纳米星,其余30%的颗粒为十面体和菱形体的无定形聚集体。纳米星的吸收峰位于近红外区域(840 nm)。因此,本方法制备的金纳米星适用于生物医学应用,特别是光热消融治疗。

方案

1. 银种子的制备

  1. 配制硝酸银(AgNO₃)储备液3) 取任意质量的样品与 10 mL 去离子水(DI 水)混合,计算该溶液的摩尔浓度。将溶液置于避光处,避免光照。
  2. 加入14.7 mg柠檬酸钠三水合物(Na3C6H5O7) 加入 10 mL 去离子水中,配制成 5 mM 溶液。振荡小瓶直至粉末完全溶解。
  3. 加入 15.1 mg 硼氢化钠(NaBH₄)4) 转移至另一含有10 mL去离子水的小瓶中,配制成40 mM溶液。立即盖紧小瓶。用手轻轻振荡溶液,然后将其放入盛有冰块的烧杯中。将烧杯置于冰箱中,并启动计时器(t1 = 0)。该新配制的溶液将在15分钟后使用,此时间足以使其充分冷却。
  4. 从硝酸银储备液(1.1)中配制10 mL浓度为0.25 mM的溶液。将搅拌子放入小瓶中,并开始搅拌。
  5. 向1.2)至1.4)中加入0.25 mL的柠檬酸钠三碱溶液
  6. 在 t1 = 15 分钟时,从冰箱中取出硼氢化钠溶液(1.3)。用移液器吸取 0.4 mL 该溶液,并加入至 1.5 中。注意:应一次性快速加入该溶液,溶液颜色将变为黄色。搅拌溶液 5 分钟。
  7. 在 t1 静置20分钟,停止搅拌,将磁铁从离心管中移开,并将离心管置于避光处。不要封闭离心管。
  8. 将溶液置于室温下避光保存至少2小时后再使用。建议在制备后一周内使用种子。

2. 生长溶液的配制

  1. 通过将 140 mg 抗坏血酸(C6H8O6)加入 10 mL 去离子水中,配制 80 mM 的抗坏血酸溶液。
  2. 配制 10 mL 浓度较高的氯金酸(HAuCl4)溶液。计算该溶液的摩尔浓度。将溶液避光保存。
  3. 通过将 364 mg 十六烷基三甲基溴化铵(CTAB - C19H42BrN)加入含有 20 mL 去离子水的样品瓶中,配制 20 mL 的 50 mM 溶液。立即将搅拌子放入样品瓶中,并在 30°C 的加热板上开始搅拌。待 CTAB 粉末完全溶解且溶液变为透明后,关闭加热板的加热功能,但继续搅拌直至完成步骤 2.7)。
  4. 将溶液 1.1)加入溶液 2.3)中,使最终摩尔浓度为 4.9×10-2 mM。启动计时器(t2 = 0)。
  5. 在 t2 = 1 min 时,将溶液 2.2)加入 2.4)中,使最终摩尔浓度为 0.25 mM。
  6. 在 t2 = 2 min 时,向 2.5)中加入 0.1 mL 的 2.1)溶液。溶液将变为无色。
  7. 在 t2 = 2 min 20 sec 时,向 2.6)中加入 0.05 mL 的 1.8)溶液(银种子)。持续搅拌悬浮液 15 分钟。悬浮液最初会变为蓝色,随后变为棕色。
  8. 在 t2 = 17 min 时停止搅拌,取出搅拌子,并将悬浮液在室温下静置 24 小时。

3. 分离金纳米星中的CTAB以用于成像、表征或实验

注意:十六烷基三甲基溴化铵(CTAB)在室温下可能结晶。为溶解晶体,可将金溶胶加热至30°C,或将小瓶浸入热水中直至晶体溶解。

  1. 将悬浮液超声处理 2 分钟。
  2. 将悬浮液在 730 rcf 条件下离心 5 分钟。纳米星将聚集在管壁上。
  3. 用移液器尽可能吸除上清液,注意不要吸走纳米星。
  4. 向管中加入去离子水,并超声处理 2 分钟。
  5. 将悬浮液在 460 rcf 条件下离心 3 分钟。由于悬浮液中 CTAB 含量较低,因此需要较低的离心力即可分离纳米星。
  6. 重复步骤 3.3)和 3.4)。
  7. 向悬浮液中加入去离子水,并在 380 rcf 条件下离心 3 分钟。
  8. 重复步骤 3.3)和 3.4)。纳米星现已可用于成像、光谱分析或实验。

4. 代表性结果:

图1显示了使用JEOL 2010-F透射电子显微镜(TEM)拍摄的银种子的透射电子显微镜图像。这些种子呈球形,平均尺寸为15 nm。金纳米星在扫描电子显微镜(SEM)模式下使用Hitachi S-5500进行成像。图2展示了采用本方法合成的纳米星在逐步放大下的图像。星形颗粒约占胶体中所有颗粒的70%。未形成完整星形的颗粒表现为十面体和菱形体的无定形聚集体(未显示)。图3显示了多个单个金纳米星。纳米星的尺寸范围为200 nm至300 nm,尖端数量在7到10个之间。若采用本方法合成的金纳米颗粒在CTAB中保存,其形貌在合成后至少1个月内保持稳定。

我们使用 Olis Cary-14 分光光度计测量了银种子和纳米星的吸收光谱。种子的吸收峰位于 400 nm,而纳米星的吸收峰位于 800 nm 至 850 nm 之间(图 4)。

讨论

在本研究中,我们介绍了一种利用银种子合成金纳米星的方法。我们发现,使用银种子可使纳米星的产率达到70%。这些纳米星具有近红外吸收峰,对应于其表面等离子体共振模式,吸收峰中心位于800 nm至850 nm之间7, 23。这些特性使得我们的金纳米星可用于生物医学领域24-26,例如光热消融治疗。

此处所述方法与其他方法的一个主要区别在于使用银种子而非金种子。采用银种子可制备出具有更长尖端和更小核心的金纳米星。由于存在多种纳米胶体合成方法,不同制备方案之间的产率难以直接比较。然而,与采用类似种子介导合成的方法27(产率可达40%–50%28)相比,本方法所获得的目标形貌金胶体产率更高,达到70%。此外,我们的悬浮液稳定性超过一个月。尽管我们的纳米星尺寸较大16,但其表面等离子体共振模式已红移至近红外区,因而更适用于生物应用。
