使用纳米结构脂质载体包封抗癌治疗药物达卡巴嗪

摘要

目前最常用的纳米结构脂质载体(NLC)制备方法包括油包水乳化、均质化及固化过程。本研究在固化后引入高剪切分散处理,对这一方法进行了改进,以获得粒径适宜、药物包封率和载药效率更高的NLC,作为达卡巴嗪递送的潜在载体。

摘要

目前临床使用的达卡巴嗪(Dac)唯一剂型为静脉输注,由于该药物在水溶液中分散性差,导致治疗效果不佳。为克服这一问题,本研究开发了一种由甘油棕榈硬脂酸酯和肉豆蔻酸异丙酯组成的纳米结构脂质载体(NLC),用于包载Dac。通过将油包水乳液固化后采用高剪切分散(HSD)方法,制备得到粒径可控的NLC。优化了表面活性剂浓度、HSD转速和时间等合成参数,最终获得粒径为155 ± 10 nm、多分散指数为0.2 ± 0.01、zeta电位为-43.4 ± 2 mV的最小NLC。这些最优参数也被用于载药NLC的制备。所得载Dac的NLC粒径为190 ± 10 nm,多分散指数为0.2 ± 0.01,zeta电位为-43.5 ± 1.2 mV。药物包封率和载药量分别达到98%和14%。这是首次报道使用NLC包载Dac的研究,表明NLC有望成为一种新型药物载体,以改善Dac的治疗特性。

引言

达卡巴嗪(Dac)是一种烷化剂,通过甲基化核酸或直接损伤DNA发挥抗肿瘤作用,从而导致细胞周期阻滞和细胞死亡1

作为一线化疗药物,Dac 已被单独使用或与其他化疗药物联合用于治疗多种癌症2-6。迄今为止,它是治疗皮肤性和转移性黑色素瘤最有效的药物,而黑色素瘤是侵袭性最强的皮肤癌类型3,7,8。然而,其应答率最高仅为20%,且治疗效果常伴随严重的全身性副作用。

在天然状态下,达卡巴嗪(Dac)具有亲水性,但由于其光敏性而稳定性较差9。目前临床上唯一可用的剂型是用于静脉输注的无菌粉末混悬液7,8。该药物疗效响应率低且全身毒性高,主要原因在于其水溶性差,导致靶部位药物可及性低,同时在非靶部位分布广泛,从而限制了药物的最大使用剂量10。静脉给药后药物迅速降解和代谢,加之耐药性的产生,进一步限制了其临床应用和治疗效果11。因此,迫切需要开发用于治疗恶性黑色素瘤的达卡巴嗪新型制剂。

如 Marilene et al.12 所综述,含有脂质体、胶束或纳米结构颗粒的胶体系统因其在药物递送中的应用而受到广泛研究。在过去十年中,纳米结构颗粒作为潜在的药物载体受到越来越多的关注,因其能够提高药物载药效率、控制药物释放、改善药物的药代动力学和生物分布,从而降低药物的全身毒性13。然而,迄今为止仅有少数纳米制剂被用于达卡巴嗪(Dac)递送的研究,这些研究显示药物可免受光降解、药物溶解度提高以及治疗效果增强10,14,15。但这些制剂存在包封效率低的问题,且部分制剂使用了成本较高的合成聚合物纳米颗粒。

由固态脂质和液态脂质混合物制成的纳米结构脂质载体(NLC)已被开发用于药物递送16,17。待包封的药物通常在液态脂质和固态脂质两相中均可溶解18,从而实现高载药量和可控释放19。本研究旨在开发一种基于NLC包封技术的新达克(Dac)制剂,采用甘油棕榈硬脂酸酯和肉豆蔻酸异丙酯作为脂质材料。制备过程包括油包水乳化、蒸发、固化和均质化。对所制备的NLC进行了粒径、形貌、超微结构和分散性的表征,并测定了药物包封效率和载药量20

方案

1. 水包油乳液的制备

  1. 称取甘油棕榈硬脂酸酯(120 mg)、肉豆蔻酸异丙酯(60 mg)、d-α-生育酚聚乙二醇琥珀酸酯(30 mg)和大豆卵磷脂(30 mg),加入12.5 ml有机溶剂(6.25 ml丙酮和6.25 ml乙醇)中。在70 °C(高于固体脂质熔点5 °C)的水浴中迅速溶解混合物。
  2. 将125、250或375 mg泊洛沙姆188加入12.5 ml去离子水(ddH2O)中,配制成1–3%(分别对应)的泊洛沙姆188水溶液,并在与上述相同的温度下加热。
  3. 将步骤1.2中的水相溶液逐滴加入步骤1.1中的油相溶液中,在400 rpm磁力搅拌下形成乳剂。继续在400 rpm下搅拌乳剂4小时,以使有机溶剂完全挥发。

2. 固化与均质化

  1. 将乳液置于低温环境(4 °C)中静置 2 小时,使其固化/结晶。
  2. 为获得纳米结构脂质载体(NLC),使用均质机对乳液进行高剪切分散(HSD),转速为 10,000–15,000 rpm,处理时间为 10–40 分钟。

3. 纳米脂质载体(NLC)制备工艺的优化

  1. 取步骤2.2中含1%、2%和3%表面活性剂浓度的样品,分别在10,000、15,000和20,000 rpm的转速下进行高剪切分散(HSD),并分别在10、20、30和40分钟的时间点取样。
  2. 检测样品的粒径(PS)、多分散指数(PDI)、形态及超微结构20
    注意:产生最小粒径(155 nm)和最低PDI值(0.2)的参数被确定为最优条件。

4. 达卡巴嗪负载的NLC(NLC-Dac)的制备

  1. 按照步骤1.1所述方法制备油相溶液,在70 °C水浴中溶解前加入Dac(70 mg)。
  2. 按照步骤1.2所述方法制备含1%表面活性剂的水相溶液,并将该溶液逐滴加入步骤4.1中制备的油相溶液中,在400 rpm磁力搅拌下形成乳液。继续搅拌乳液4小时,以蒸发有机溶剂。
  3. 将乳液置于4 °C冷藏室中2小时,使其按照步骤2所述固化/结晶,最后根据步骤3确定的最佳参数,对乳液进行高剪切分散(HSD)处理。

结果

采用不同参数,以甘油棕榈硬脂酸酯和肉豆蔻酸异丙酯制备的NLC及NLC-Dac,对其粒径(PS)、多分散系数(PDI)、形态学和超微结构进行了表征20。NLC的粒径和多分散系数受表面活性剂浓度、高剪切分散(HSD)速度及时间的影响。根据NLC的粒径和PDI判断,当表面活性剂浓度为1%,剪切分散速度为15,000 rpm、持续30 min时效果最佳(图1ABC),因此本研究选择该条件作为NLC制备的最优参数。

基于脂质的纳米结构颗粒已被用于为疏水性药物的递送提供高度亲脂的载体。纳米结构脂质载体(NLC)是第二代固态纳米结构脂质载体,在室温和体温下均呈固态。在NLC中将固态脂质与液态脂质结合,可导致结晶结构更不完善,从而提高药物包封效率,并减少储存过程中包封药物的析出。

对于NLC的合成,最常用的方法包括油包水乳化法、均质化以及固化/结晶过程21,22。均质化可使NLC在水相中充分分散,而低温固化则促使内部油相结晶。已有报道采用多种均质化方法,包括磁力搅拌、超声处理和高压均质(HSD),这些方法在固化前和/或固化过程中使用23,24
