免疫系统的疾病会导致自身免疫性和炎症性疾病10 和癌症11。所有这些病理条件理想地要求迅速诊断为有效的治疗。在自身免疫性疾病的情况下, 血清学存在的自身抗体在大多数情况下是一个预测的诊断自身免疫 10,12。这些抗体与细胞表面和细胞外自身抗原发生反应, 它们往往存在多年之前的自身免疫性疾病10,12。免疫缺陷和癌症也被诊断为血液检测, 要么测量抗体等免疫元素的水平, 要么测量其功能活动 11

确定循环抗体的曲目及其血清学水平是最重要的, 以建立一个预后, 并评估所有上述病理条件的进展。我们之前已经证明了 pga 技术在分析不同动物物种中的循环抗体的潜力13-16, 最大限度地减少了大量血清学样本的使用, 避免了这一问题与抗体交叉反应17相关, 并允许对大量抗体进行高通量分析 15.

1. 糖片生产

  1. 微阵列制备
    1. 用非接触式机器人排列器 (落模量 ~ 900 pl), 在6个复制的 n-羟基琥珀衍生玻璃滑梯上, 以 300 mm 磷酸盐缓冲盐水 (pL, ph 8.5) 将糖类 (50 mm) 和多糖 (10μg/ml) 打印成铝。每张幻灯片包含4个不同的子数组块 (图 1a, 颜色) 重复6次。每个子数组由1B 不同的糖原点组成, 包括控件 (8 行 x14 列) (图 1b)。

  2. 糖芯片质量控制
    1. 使用复杂免疫球蛋白制剂的1mg/ml 溶液分析每个批次中的两个微阵列 (cip, 含有 igg、igm 和 iga), 将生物素化山羊抗人免疫球蛋白10μgml 溶液作为二级抗体 (igm + igg + iga), 其次是1μgml相应荧光链霉素共轭的溶液 (通过下面描述的协议, 请参见步骤 2)。
    2. 扫描和分析糖类芯片 (参见步骤 3, 糖基阵列的分析)。
    3. 使用芯片内和片间相关性高于0.9 的微阵列批次。

2. 糖原阵列技术

  1. 准备以下水溶液 (超纯水), 并将其存放在室温 (25°c):
    • pacfer-1: 1% (w/v) pbs 中的牛血清白蛋白 (bsa), 小20和 0.01% (w/v)
    • 缓冲区-2: 1% (w/v) bsa 在 pbs, 0.1% (v/v) tween-20 和 0.01% (w/v) nan3
    • 缓冲区 3:0.1% (v/v) tween-20 在 pbs 中。
    • 缓冲区-4: 0.001% (v/v) 在 pbs 中的 tween-20。
  2. 糖芯片和样品制备
    1. 将装有幻灯片的储物箱放在桌子上, 直到它们达到室温 (25°c)。
      注: 使用无粉末乳胶手套。糖片必须由条形码所在的玻璃滑梯底部操作。条形码将帮助您识别右侧, 避免与打印糖类的表面接触。
    2. 打开盒子, 取出糖片, 放在孵化器 (25°c), 已经用湿滤纸调节, 以保持室内湿度不变。
    3. 同时, 用 buffer-1 (1:20) 在 1.5 ml 管中稀释小鼠血清。用涡流混合器对血清溶液 (5秒) 进行同质化。

    4. 均匀化后, 在水浴中孵育37°c 稀释后的血清 10分钟, 以避免免疫球蛋白聚集。在 10, 000 x g 和25°c 下离心管 3分钟, 收集上清液并丢弃任何沉淀材料。
    5. 小心地将糖屑放入孵化器中。用1毫升的 buffer-3 在25°c 下将其培养 15分钟, 以消除糖片表面的任何残留材料。
    6. 将糖片固定在垂直位置, 然后用塑料巴斯德移液器用几滴 bufer-3 将其重新清洗。使用滤纸小心地从糖屑表面取出缓冲液。
  3. 反应: 抗体结合
    1. 将糖屑放入培养室。使用微型移液器将稀释后的血清样本涂在糖片表面。在37°c 条件下用眼眶搅拌 (30 转/分) 孵化1.5 小时, 确保使用移液器的尖端稀释后的血清样本覆盖糖片表面的所有干燥区域。
    2. 取出多余的样品, 在25°c 的 bufer-3 中浸泡5分钟的糖屑。然后, 将糖化芯片移动到具有 buffer-4 (5分钟) 的容器中, 最后用超纯水清洗 (5分钟) 的糖片。在 175 x g 和25°c 下离心糖芯片 1分钟, 以去除液体。
  4. 检测: 二级抗体
    1. 将糖屑放入培养室。在糖片表面上传播山羊抗鼠 (igg + igm) 的溶液 (5μg/ml), 该溶液与 bafer-2 中的生物素结合。在37°c 下进行轨道搅拌 (30 转/分) 孵化1小时。
    2. 取出未绑定的分数, 然后重复清洗步骤。
    3. 离心后, 在25°c 的黑暗中, 用相应的荧光标记链霉素溶液 (在 buffer-2 中) 的 2μg/ml 孵育糖芯片 45分钟 (30 转/分)。

3. 糖原阵列分析

  1. 扫描阵列

  2. 阵列量化
