制备肾皮质细胞外基质来源水凝胶

本文介绍了一种利用肾脏皮质细胞外基质制备水凝胶的方案,以保留天然肾脏细胞外基质(ECM)的结构和生化组成。文中描述了该水凝胶的制备过程及其应用。最后,探讨了利用该水凝胶支持肾脏特异性细胞与组织再生及生物工程构建的前景。

摘要

细胞外基质(ECM)提供重要的生物物理和生化信号,以维持组织稳态。现有的合成水凝胶可为 体外 细胞培养环境,但缺乏诱导细胞产生生理行为所必需的蛋白质和配体组成。本文描述了一种源自肾脏皮质细胞外基质(ECM)的水凝胶制备方法,该水凝胶具有适当的机械强度和支持性的生化组成。该水凝胶通过机械匀浆和溶解脱细胞的人肾脏皮质ECM制备而成。该基质保留了天然肾脏皮质ECM的蛋白质比例,同时能够形成具有生理机械刚度的凝胶。该水凝胶可作为基底,用于在生理条件下维持源自肾脏皮质的细胞。此外,可通过调控水凝胶的组成以模拟疾病环境,从而为未来研究肾脏疾病提供可能。

引言

细胞外基质(ECM)提供重要的生物物理和生化信号,以维持组织稳态。其复杂的分子组成调控着组织的结构与功能特性。结构蛋白为细胞提供空间感知能力,并支持细胞黏附与迁移。1结合的配体与细胞表面受体相互作用以调控细胞行为2肾脏细胞外基质含有大量分子,其组成和结构因解剖位置、发育阶段和疾病状态而异3,4重现细胞外基质(ECM)的复杂性是研究肾源性细胞的关键方面 体外.

先前尝试模拟细胞外基质(ECM)微环境主要集中在对完整组织进行脱细胞处理,以制备可重新接种细胞的支架。脱细胞处理通常使用化学去垢剂,如十二烷基硫酸钠(SDS)或非离子型去垢剂,并采用器官整体灌注或浸没振荡的方法进行。5,6,7,8,9,10,11,12,13此处提供的支架可保留天然组织细胞外基质中的结构与生化信号;此外,利用供体特异性细胞进行再细胞化在重建外科手术中具有临床意义14,15,16,17,18,19然而,这些支架缺乏结构灵活性,因此无法与当前使用的许多设备兼容 体外 研究。为了克服这一限制,许多研究团队已将脱细胞ECM进一步加工成水凝胶20,21,22,23,24这些水凝胶可与注射成型和生物墨水兼容,并克服了脱细胞支架对细胞造成的微米级空间限制。此外,天然细胞外基质中的分子组成和比例得以保留。3,25在此,我们展示了一种制备源自肾脏皮质细胞外基质(kECM)的水凝胶的方法。

本实验方案的目的是制备一种可模拟肾脏皮质区域微环境的水凝胶。通过在1% SDS溶液中持续振荡处理,对肾脏皮质组织进行脱细胞以去除细胞成分。由于SDS能够快速清除具有免疫原性的细胞物质,因此常被用于组织脱细胞过程6,7,9,26。随后,对所得的肾源性细胞外基质(kECM)进行机械均质化和冷冻干燥处理5,6,9,11,26。使用强酸联合胃蛋白酶进行溶解,最终获得水凝胶储备溶液20,27。该方法可保留对结构支撑和信号转导具有重要作用的天然kECM蛋白3,25。所制备的水凝胶刚度可控制在与天然人肾皮质相近一个数量级的范围内28,29,30。该基质可提供一种生理性的微环境,已被用于维持肾脏特异性细胞的静息状态,相较于其他基质蛋白来源的水凝胶更具优势。此外,可通过调控基质成分(例如添加I型胶原蛋白)来模拟疾病环境,用于研究肾纤维化及其他肾脏疾病31,32。

方案

人类肾脏由 LifeCenter Northwest 按照器官获取组织协会制定的伦理准则分离获得。本方案遵循华盛顿大学规定的动物护理和细胞培养指南。

1. 人肾组织的制备

  1. 脱细胞溶液的制备
    1. 对一个5000 mL烧杯和一根70 x 10 mm搅拌子进行灭菌处理。
    2. 在烧杯中将十二烷基硫酸钠(SDS)按1:1000(重量:体积)的比例溶于经高压灭菌的去离子水中。将溶液置于搅拌台上,以约200 rpm的速度搅拌24小时,或直至SDS完全溶解。
      注意:通常,2500 mL的1% SDS溶液足以对一个完整的人类肾脏进行脱细胞处理。
    3. 将溶液转移至500 mL无菌真空滤器中,过滤后收集到已灭菌的可密封容器中。
  2. 肾脏组织的处理
    1. 清洗并高压灭菌一对镊子、两个止血钳、一对通用剪刀、两个手术刀片手柄、一个用铝箔覆盖的1000 mL烧杯,以及一根36 x 9 mm搅拌子。
    2. 在组织培养超净台内铺上吸水垫。将烧杯、一个无菌组织培养皿(150 x 25 mm)和完整的肾脏器官放入超净台中。向烧杯中加入500 mL的1% SDS溶液。
      注意:人类肾脏由LifeCenter NorthWest提供,运输过程中保持冰上保存。
    3. 将肾脏置于无菌组织培养皿中(图1A)。使用手术刀轻轻刮除肾包膜周围的全部肾周脂肪(图1B)。
    4. 用手术刀在肾脏上端做一条浅表的8-10 cm切口,深度以刚好切开肾包膜而不损伤下方皮质组织为宜。使用两个止血钳将肾包膜从皮质组织上剥离(图1C)。
    5. 沿肾脏外侧冠状面使用手术刀将肾脏纵向切为两半(图1D)。用手术刀剔除两半肾脏中的髓质区域,分离出皮质组织(图1E),并将皮质组织切成0.5 cm3大小的组织块(图1F)。去除所有肉眼可见的大血管。
  3. 细胞外基质的分离
    1. 在组织培养超净台中,向一个1000 mL烧杯中加入500 mL的1% SDS溶液。将切碎的皮质组织和搅拌子放入含SDS溶液的烧杯中。用高压灭菌的铝箔覆盖烧杯,并将其置于组织培养超净台外的搅拌台上,以约400 rpm的速度搅拌。
    2. 皮质组织在搅拌台上处理24小时后,将烧杯移入组织培养超净台中,并放置一个由尼龙网制成的40 µm无菌细胞滤筛。另取一个1000 mL烧杯加入200 mL次氯酸钠溶液(漂白剂),并将其置于超净台内。
    3. 使用移液器将SDS溶液通过细胞滤筛转移至含漂白剂的烧杯中。将所有SDS溶液移除,直至烧杯中仅剩脱细胞组织和细胞滤筛。
      注意:细胞滤筛应能防止组织在溶液抽吸过程中被带出。
    4. 将细胞滤筛保留在烧杯中,加入500 mL新鲜SDS溶液。用同一片铝箔覆盖烧杯,并以相同转速继续置于搅拌台上。
    5. 每24小时重复步骤1.3.1–1.3.3,共使用新鲜SDS溶液处理五天。
    6. 按照步骤1.3.1–1.3.3所述方法,使用高压灭菌的去离子水对脱细胞组织进行清洗,每24小时更换一次,共清洗三天。
    7. 按照步骤1.3.1–1.3.3所述方法,使用细胞培养级水对脱细胞组织进行清洗,每24小时更换一次,共清洗两天。
    8. 重复步骤1.3.1–1.3.2。将脱细胞组织(此后称为kECM)转移至一个30 mL自立式锥形管中,并加入细胞培养级水直至完全浸没所有组织。

2. 水凝胶储备溶液的制备

  1. 脱细胞组织的机械处理
    1. 在组织培养罩内,使用组织匀浆器在锥形管中对kECM进行机械匀浆,持续2分钟。
      注意:匀浆后的kECM应呈现为不透明溶液,无可见ECM碎片。
    2. 将含有kECM的锥形管浸入液氮中,直至管周沸腾现象完全消失。将kECM在-4 °C下保存过夜。
  2. 冷冻脱细胞组织的冻干处理
    1. 略微松开锥形管盖以允许气体交换,并将管子放入冻干机中。冻干kECM三天,或直至其呈现为细腻的白色粉末。储存于-4 °C。
  3. 凝胶的化学消化与溶解
    1. 对一个20 mL闪烁瓶及其瓶盖、一根15.9 × 7.9 mm搅拌子以及一对尖头镊子进行高压灭菌。
    2. 称取冻干kECM的质量,并根据以下公式计算溶解kECM至3%(30 mg/mL)溶液所需的HCl体积和胃蛋白酶质量,其中mpepsin为胃蛋白酶质量,mtissue为冻干组织质量,VHCl为0.01 N HCl的体积:

    3. 在组织培养罩内,向闪烁瓶中加入猪胃蛋白酶、0.01 N HCl和搅拌子,置于搅拌器上以约500 rpm转速搅拌,直至胃蛋白酶完全溶解。将冻干kECM转移至闪烁瓶中,继续在搅拌器上以约500 rpm转速搅拌三天。

3. 水凝胶凝胶化

  1. 肾脏ECM水凝胶的制备
    1. 通过将kECM水凝胶储备液与1 N NaOH、10x培养基添加剂(M199)和细胞培养基混合来形成凝胶。所有溶液均需保持在冰上。
      注意:细胞培养使用的终浓度为7.5 mg/mL。1 mL kECM凝胶足以满足所展示的细胞培养实验需求。
    2. 使用以下公式确定可使用的kECM凝胶产量及所需kECM储备液的体积,其中Vfinal为所制备凝胶的体积,Vstock kECM为所需kECM储备液的体积,Cstock kECM为kECM储备液的浓度,Cfinal为终凝胶的浓度:

    3. 使用以下公式计算所需中和试剂的体积,其中VNaOH为1 N NaOH的体积,V10X为M199 10X培养基添加剂的体积,V1X为细胞培养基的体积:

    4. 在组织培养超净台中,用移液器将中和试剂(NaOH、M199和细胞培养基)加入无菌的30 mL直立锥形管中。使用微型刮刀混合中和试剂溶液。
    5. 使用无菌1 mL注射器将适量的kECM储备液转移至中和试剂溶液中。用微型刮刀轻轻搅拌,直至获得颜色均匀的水凝胶溶液。
      注意:搅拌时应缓慢轻柔,避免引入气泡。
    6. 若需将细胞掺入kECM水凝胶中,则在步骤3.1.1.3的中和溶液体积计算中,从细胞培养基体积(V1X)中减去10 µL。
      1. 将细胞悬浮于10 µL细胞培养基中。使用以下公式确定所需细胞数量,其中#cells表示待悬浮的细胞数,Vfinal为所制备凝胶的体积:

      2. 在kECM储备液与中和试剂溶液混合完成后,用移液器将10 µL含细胞的悬浮液加入终kECM凝胶中。用微型刮刀搅拌,直至细胞均匀分布。
  2. 使用1 mL注射器将kECM水凝胶填充至所需的细胞培养装置中。
  3. 将凝胶在37 °C下静置1小时,之后再进行细胞转移或接种。

结果

kECM水凝胶提供了一种化学成分与天然肾脏微环境相似的基质,用于肾脏细胞培养。制备该水凝胶时,首先从完整肾脏器官中机械分离出肾皮质组织并进行切碎(图1)。使用化学去污剂对组织进行脱细胞处理(图2A.1-A.3),随后用清水冲洗以去除去污剂颗粒(图2A.4-A.6),从而获得分离的肾皮质ECM。组织学评估证实,典型的基底膜蛋白(如胶原蛋白-IV和层粘连蛋白)以及结构蛋白(如胶原蛋白-I)均得以保留,仅纤连蛋白有所缺失(图2B)。此外,ECM中的蛋白质组成(包括亚型的保留)与天然肾ECM中观察到的数值一致(图3)。将ECM(图4A)进行机械均质化和冷冻干燥...

讨论

基质提供了调控细胞行为的重要力学和化学信号。合成水凝胶能够支持复杂的三维图案化,但无法提供生理基质微环境中存在的多样化细胞外信号。来源于天然细胞外基质(ECM)的水凝胶是进行体内(in vivo)和体外(in vitro)研究的理想材料。以往的研究已利用去细胞化ECM水凝胶来包覆合成生物材料,以防止宿主产生免疫反应33,34,诱导干细胞分化35,36,37,38,作为二维培养和软光刻细胞培养的基底39,40,41,
