所有的研究和实验规程都得到了布法罗大学和纽约西部动物护理和使用委员会的批准, 并符合这些委员会的准则。

1. 实验设置和一般建议

请注意:本协议从23个月大开始, 在 c57bl/6j 背景下总共使用了24只雌性小鼠。小鼠携带了一个有条件的 sirtloxp-exon4-loxp突变29, 然而, 这并没有在这个实验中诱导。

  1. 确保小鼠收到永久标识符, 如耳标、rfid 芯片或尾巴纹身。
    请注意:建议在评估期间使用临时标记 (例如, 永久标记标记尾), 以便在评估期间快速识别。
  2. 在一天结束时使用0.25-0.5% 的漂白剂 (v/v) 或 70% (v/v) 乙醇清洁跑步机, 或在两次试验之间取出粪便或尿液。在启动新的试验之前完全使用干式解决方案。

2. 使老鼠适应跑步机设备

请注意:在基线实验前1个月开始小鼠的适应。

  1. 在手动模式下使用跑步机软件 (v3.4.7) 设置初始培训计划 (图 1和表 1)。
    1. 打开跑步机软件。然后, 单击该文件并打开实验。
    2. 在 "适应"下的"手动" 选项卡 (图 1) 上输入表1中所示的值。

  8. 在第二次耐力训练后的一至两天内, 提供第4节所述的两次跑步机上坡无氧能力评估, 当天进行, 训练间隔至少30分钟。

3. 平面连续跑步机评估

  1. 按照步骤2.1 中所述创建跑步机程序, 使用表 1 "评估" 下所示的值。在 "冲击检测"下, 将会话编号设置为 1, 活动通道数设置为1到 6, 具体取决于鼠标数、访问网格的次数为 10, 冲击次数设置为20。

5. 短时间高强度间隔训练

  1. 设置跑步机上坡 (25°), 并卸下塑料盖。
  2. 确定每个鼠标的强度。使用最后一个成功完成阶段的速度 (步骤 4.5), 并使用表 3查找强度组和相应的"基础""冲刺" 和"虚线" 速度。

    请注意:演习期间不进行电击。
  6. 激励老鼠使用化妆刷 (推荐) 或舌头抑制器运行, 以轻松激励落在冲击栅附近或冲击栅上的小鼠。如果鼠标在5秒内没有响应重试, 则为 10秒, 然后每30秒重试一次, 或在恢复间隔期间重试, 直到会话结束。

  7. 将无法在连续两次运动中完成前三个间隔的小鼠移动到强度较低的组 (表 3)。
  8. 将连续两次运动中不需要动机的小鼠转移到强度较高的组 (表 3)。
  9. 在跑步机运行时, 以笼子里的久坐控制老鼠为例。或者, 可以在不移动的跑步机的车道上放置久坐控制10分钟。
