获取生物材料的细胞毒性和细胞反应

摘要

本方法旨在通过制备可溶性提取物,结合细胞活力检测和表型分析(包括流式细胞术、RT-PCR、免疫细胞化学以及其他细胞和分子生物学技术),评估生物材料的细胞毒性。

摘要

生物材料直接或间接与人体组织接触,因此评估其细胞毒性至关重要。此类评估可通过多种方法进行,但不同方法之间存在较大差异,影响了结果的可重复性及相互比较。本文提出一种利用可溶性浸提液评估生物材料细胞毒性的实验方案,我们将其应用于牙科生物材料。文中详细描述了浸提液的制备过程,包括颗粒制备及其在培养基中的浸提步骤。生物材料细胞毒性的评估基于四甲基偶氮唑盐(MTT)法检测细胞代谢活性、磺酰罗丹明B(SRB)法检测细胞活力、流式细胞术分析细胞死亡模式,以及May-Grünwald Giemsa染色观察细胞形态。此外,本文还提供了一套评估细胞功能的方案,通过免疫细胞化学和PCR检测特定标志物的表达水平。本方案采用浸提液法,为生物材料的细胞毒性及细胞效应评价提供了可重复且可靠的全面指导。

引言

生物相容性可定义为材料整合组织并诱导有利治疗反应的能力,且不引起局部或全身性损伤1,2,3。对于任何拟用于医学用途的材料,其生物相容性评价均至关重要。因此,本实验方案为致力于开发新型生物材料或探索现有生物材料新应用的研究人员提供了一种系统且全面的方法。

体外细胞毒性试验广泛用于生物相容性评价的第一阶段,通常采用原代细胞培养或细胞系。其结果是评估潜在临床应用价值的初步指标。除了在生物材料研发中具有关键作用外,此类检测也是满足美国和欧盟监管机构(FDA 和 CE 认证)市场准入现行法规要求的强制性环节4,5,6,7,8。此外,在生物医学研究中采用标准化检测方法,有助于对相似生物材料或医疗器械的不同研究结果进行重复验证和比较,具有重要意义9

国际标准化组织(ISO)指南被众多独立的商业、监管和学术实验室广泛采用,用于以准确且可重复的方式测试材料。ISO 10993-5 涉及体外细胞毒性评估,ISO 10993-12 则涉及样品制备10,11。在生物材料测试中 ,根据材料类型、接触组织及治疗目的,提供了三类测试方法以供选择:浸提液法、直接接触法和间接接触法8,11,12,13。浸提液法通过将生物材料加入细胞培养基中进行富集获得;直接接触法是将生物材料直接放置于细胞培养物上;而间接接触法则是通过屏障(如琼脂糖凝胶)将材料与细胞分隔后进行共孵育11。必须设置适当的对照组,并至少进行三次独立的实验5,8,10,11,14

模拟或放大临床条件对于确定细胞毒性潜力至关重要。在进行浸提物测试时,材料的表面积; 培养基体积;培养基和材料的pH值;材料的溶解度、渗透压和扩散比率;以及振荡、温度和时间等浸提条件均会影响培养基的富集程度5

该方法可对多种固态和液态药物制剂的细胞毒性进行定量和定性评估。可开展多种检测,如中性红摄取试验、集落形成试验、MTT 检测和 XTT 检测5,10,14

目前发表的大多数细胞毒性评估研究采用的是较为简单的检测方法,即 MTT 和 XTT 法,这些方法提供的信息有限。生物相容性评估不仅应包括对细胞毒性的评价,还应包括对测试材料生物活性的评估——本方案支持这一观点2。在有合理依据并加以记录的情况下,应使用额外的评估标准。因此,本方案旨在提供一份全面的指南,详细阐述一套用于生物材料细胞毒性评价的方法。此外,本文还描述了对不同细胞过程的评估,包括细胞死亡类型、细胞形态、细胞在特定蛋白质合成中的功能以及特定组织的生成。

方案

1. 制备菌体沉淀

  1. 通过在聚氯乙烯(PVC)板上打制已知尺寸的圆形孔来制备聚氯乙烯(PVC)模具。
    注:PVC模具可制成不同尺寸。使用公式 A = h(2πr) + 2πr2 计算PVC模具的接触表面积(r:圆柱半径;h:圆柱高度)。
  2. 根据制造商说明书,并尽可能在实验开始前制备待测生物材料。
    注:对于膏状/膏状配方的生物材料,需手动用调拌刀混合适量基质膏和催化剂。对于基于液体和粉末配方的其他材料,应根据制造商说明书或适用于新材料的方法,进行手动调拌或使用振动机械混合。对于液态材料,此步骤无需进行。从第2步开始实验方案。
  3. 用调拌刀将生物材料置于模具中,并让其在适当时间内固化。
    注:生物材料的固化时间及固化条件必须遵循制造商说明书或适用于新材料的规定。
  4. 固化后,将生物材料颗粒从PVC模具中取出,并放入容器中(可使用6孔板或培养皿)。
  5. 通过将颗粒置于紫外灯(UV)下每侧照射20分钟,对颗粒进行灭菌。

2. 获取生物材料的提取物

注意:所有操作均应在严格的无菌条件下进行。

  1. 根据1.1节所述公式计算颗粒表面积,确定所需的颗粒数量。
    注意:作为参考值,250 mm的接触表面积2/mL11,15 通过加入9粒(r 3 mm × h 1.5 mm)的颗粒实现 mL 培养基。
  2. 制备可溶性提取物(富含生物材料的提取物)。
    1. 将沉淀物置于50 mL离心管中,加入相应量的细胞培养基。将离心管放入培养箱中,在37°C下孵育24小时 37°,持续旋转。
      注意:使用适用于细胞培养的细胞培养基。
    2. 24小时后,从培养箱中取出试管。此时,提取物的浓度为1/1或100%。
    3. 通过向细胞培养基中连续等体积添加预处理培养基,对提取物进行系列稀释。
      注意:培养基无需调节pH。
      1. 加入1 mL培养基至1 mL 100%提取物中,得到50%提取物。再加入1 mL培养基至1 mL 50%提取物中,得到25%提取物,依此类推(图1).
        注意:使用每种化合物对应的相关浓度。

3. 细胞与生物材料浸提液的孵育

  1. 制备细胞悬液,并根据实验所需的细胞数量,将其接种至合适的细胞容器中,例如多孔板。
    1. 从处于对数生长期、融合度为80%至90%的细胞培养瓶开始。
    2. 弃去细胞培养液,用磷酸盐缓冲液(PBS)洗涤,并加入胰蛋白酶-EDTA溶液(75 cm2 培养瓶使用1至2 mL)消化贴壁细胞使其脱落。
    3. 加入细胞培养液(75 cm2 培养瓶使用2至4 mL),将细胞悬液转移至离心管中,在200 × g 条件下离心5分钟。
    4. 将沉淀重悬于已知体积的细胞培养液中。
      注意:本方案适用于贴壁细胞培养;但经过简单调整后也可用于悬浮细胞培养。
    5. 使用血细胞计数板对细胞进行计数,并计算细胞悬液的浓度。
    6. 将确定数量的细胞悬液用培养基稀释后接种至多孔培养板中。作为参考,接种密度建议为5 – 20 × 105 个细胞/cm2
      注意:应根据细胞类型及其特性(特别是细胞倍增时间)计算合适的接种细胞数量。
  2. 将细胞置于培养箱中孵育24小时,以促进细胞贴壁。
  3. 孵育结束后,向培养板中加入待测材料的可溶性提取液。
    1. 吸除原有细胞培养液。
    2. 根据预设的浓度梯度,向各孔中加入生物材料的提取液;对照孔中加入新鲜细胞培养液。
    3. 将培养板继续孵育24小时或更长时间。
      注意:每次实验均需设置阴性对照组,即不加处理因素、仅维持在正常培养液中的细胞。孵育时间可根据具体研究目的进行调整。

4. 代谢活性的评估

  1. 在细胞与生物材料提取物共孵育后,从培养板中吸去培养基,并用 PBS 清洗每个孔。
  2. 在每个孔中加入适量体积的 PBS(pH 7.4)配制的 0.5 mg/mL 3-(4,5-二甲基噻唑-2-基)-2,5-二苯基氯化四氮唑(MTT)溶液。
  3. 将培养板在 37 °C 条件下避光孵育 4 小时或过夜。
  4. 为溶解生成的甲臜结晶,向每个孔中加入适量体积的异丙醇中 0.04 M 盐酸溶液,并将培养板振荡 30 分钟。
    注意:根据孔的大小调整 MTT 和异丙醇的用量。
  5. 如有必要,通过上下吹打混匀各孔内容物,直至肉眼不可见结晶。
  6. 使用分光光度计,在 570 nm 波长下以 620 nm 作为参比波长测定吸光度。
  7. 为计算代谢活性,将处理组细胞的吸光度除以对照组培养物的吸光度;为获得百分比值,乘以 100。

5. 细胞死亡评估

注意:进行此评估时,每个实验条件至少应使用 106 个细胞。

  1. 根据所评估的条件,使用正确标识的离心管。
  2. 在细胞与生物材料提取物共孵育后,收集培养基至相应的离心管中。
  3. 消化并悬浮细胞,将细胞悬液加入相应的离心管中。
  4. 在120 × g 条件下离心5分钟,浓缩细胞悬液。
  5. 用PBS洗涤沉淀,在1,000 × g 条件下离心5分钟,弃去PBS。
  6. 加入1 mL PBS,将细胞沉淀转移至已标识的流式管中。
  7. 在1,000 × g 条件下离心5分钟,去除PBS。
  8. 加入100 µL结合缓冲液(0.01 M HEPES、0.14 mM NaCl和0.25 mM CaCl216,室温避光孵育约15分钟,使细胞膜恢复。
  9. 室温避光条件下,加入2.5 μL荧光标记的Annexin-V和1 μL碘化丙啶,孵育15分钟。
  10. 孵育结束后,加入400 μL PBS,立即上机进行流式分析。使用适当的软件进行数据的分析与定量。
  11. 以活细胞、早期凋亡、晚期凋亡/继发性坏死及坏死细胞的百分比形式呈现结果。

6. 形态学评估

  1. 选择合适尺寸的无菌玻璃盖玻片,使其能够放入多孔板内。
  2. 使用无菌镊子将每片盖玻片放入一个孔中。
  3. 将适当浓度的细胞悬液加入各孔中,于37 °C、含95%空气和5% CO2的湿润环境中孵育过夜。
  4. 将细胞培养物暴露于提取物中,操作方法如前所述。
  5. 吸除培养基,并用PBS洗涤。
  6. 让盖玻片在室温下干燥,然后加入足量的May-Grünwald染液以覆盖盖玻片;孵育3分钟。
  7. 去除染液,并用蒸馏水洗涤1分钟。
  8. 去除水液,加入足量的吉姆萨(Giemsa)染液以覆盖盖玻片;孵育15分钟。
  9. 用流水冲洗盖玻片。
  10. 将盖玻片转移至载玻片上。
  11. 在显微镜下观察。使用选定的放大倍数拍摄照片。

7. 通过逆转录聚合酶链式反应(RT-PCR)评估细胞功能

注意:进行此评估时,每个条件至少应使用 2×106 个细胞。例如,碱性磷酸酶可作为评估成牙本质细胞活性的兴趣基因。其他兴趣基因见表1

  1. 按照上述方法接种细胞。
    注意:接种的细胞浓度可能需要根据细胞类型以及所研究生物材料的细胞毒性进行相应调整。
  2. 按照上述方法用可溶性提取物孵育细胞。
  3. 按照前述方法将细胞解离,获得细胞悬液。
  4. 用 PBS 洗涤细胞两次;洗涤时在室温下以 200 × g 离心 5 分钟。
  5. 通过将细胞沉淀重悬于 1 mL RNA 纯化溶液(例如 NZYol)中,剧烈搅拌并反复吹打,以裂解细胞。
  6. 将样品在室温下孵育 5 分钟。
  7. 加入 200 µL 氯仿,用手剧烈振荡试管 15 秒。
  8. 在室温下孵育 3 分钟。
  9. 在 4 °C 下以 12,000 × g 离心 15 分钟。在此离心过程中,样品会形成两相,RNA 位于水相(上层)。
  10. 将水相转移至新管中,加入 500 µL 冷异丙醇以沉淀 RNA。
  11. 在室温下孵育样品 10 分钟,然后在 4 °C 下以 12,000 × g 离心 10 分钟。
  12. 弃去上清液,并在 4 °C 下以 7,500 × g 离心 5 分钟,用 1 mL 75% 乙醇洗涤沉淀。
  13. 在室温下干燥沉淀,直至乙醇完全挥发。
  14. 用无 RNase 的水重悬沉淀。
  15. 使用吸收光谱法在 260 nm 和 280 nm 波长下定量并测定样品纯度。确定 RNA 纯度,使用纯度比值(A260/280)约为 2.0 的样品。
  16. 将样品储存在 -80 °C 下。
  17. 按照试剂盒制造商的说明书进行 RT-PCR 实验。17
    注意:根据研究目的,选择需检测的特定标志物。

8. 通过蛋白质鉴定评估细胞功能

注意:根据研究目标,选择需要评估的特定蛋白质。例如,本研究以牙本质涎蛋白(DSP)作为评估成牙本质细胞活性的兴趣蛋白。其他感兴趣的蛋白质见表1

  1. 将细胞培养在盖玻片上,并按前述方法加入提取物处理。
  2. 用 PBS 洗涤细胞培养物。
  3. 在室温下用 3.7% 多聚甲醛固定 30 分钟。
  4. 用 PBS 洗涤两次。
  5. 用含 0.5% Triton 的 PBS 处理 15 分钟以进行透化。
  6. 用含 0.3% 过氧化氢的 PBS 封闭过氧化物酶,作用 5 分钟。
  7. 用 PBS 洗涤两次。
  8. 用含 0.5% 牛血清白蛋白(BSA)的溶液洗涤两次。
  9. 用 2% BSA 封闭细胞培养物,孵育 45 分钟。
  10. 用含 0.5% BSA 的 PBS 洗涤。
  11. 根据所选蛋白,加入一抗在室温下孵育 60 分钟。
    注意:本实验方案使用一抗 DSP(M20) 抗体(1:100 稀释)和二抗多克隆兔抗山羊免疫球蛋白/HRP(1:100 稀释)。
  12. 用含 0.5% BSA 的 PBS 洗涤五次。
  13. 加入二抗,在室温下孵育 90 分钟。
    注意:使用含 0.5% BSA 的 PBS 配制抗体稀释液。
  14. 用含 0.5% BSA 的 PBS 洗涤五次,每次洗涤 1 分钟。
  15. 加入底物与显色剂混合液(浓度为每毫升底物中含 20 µL 显色剂),孵育 25 分钟。
  16. 用含 0.5% BSA 的 PBS 洗涤两次。
  17. 用苏木精复染 15 分钟。
  18. 依次用 0.037 mol/L 氨水和蒸馏水洗涤 5 分钟,以去除多余染料。
  19. 将盖玻片封固于载玻片上,封片介质使用甘油。
  20. 室温避光过夜干燥。
  21. 在显微镜下观察,并以选定的放大倍数拍摄照片。

9. 通过茜素红S检测法评估矿化

  1. 配制浓度为 40 mM 的茜素红 S 溶液18。在避光条件下搅拌 12 小时以充分混匀。
    注意:配制 100 mL 茜素红 S 溶液时,将 1.44 g 茜素粉末(分子量:360 g/mol)溶于超纯水中,并避光保存。该溶液的 pH 值至关重要,应控制在 4.1 至 4.3 之间。
  2. 按照上述方法,用可溶性提取物孵育细胞培养物。
  3. 用 PBS 洗涤细胞培养物三次。
  4. 在室温下用 4% 多聚甲醛固定 15 分钟。
  5. 用 PBS 洗涤三次。
  6. 在 37 °C、避光条件下,用茜素红染色液染色 20 分钟。
  7. 染色后,用 PBS 洗涤培养板,以去除多余的染料。
  8. 在显微镜下观察,并以选定的放大倍数拍摄照片。
  9. 向每孔中加入提取液(由 10% (w/v) 乙酸和 20% (w/v) 甲醇组成),室温下搅拌 40 分钟。
  10. 使用分光光度计在 490 nm 波长处测定吸光度19

结果

此处的代表性结果涉及牙科生物材料的研究。该提取方法可用于获取牙科材料暴露后细胞的毒性特征及细胞功能,包括对代谢活性(图2)、细胞活力、细胞死亡特征及细胞形态(图3)以及特定蛋白表达(图4)的影响。

MTT 法可用于简便快速地评估材料的细胞毒性。可对两种或多种材料进行比较(图2);即使在低浓度(6.25%)和中等浓度(50%)下,若代谢活性显著降低,则表明材料毒性较高(图2a)。同时,细胞毒性较小的材料则仅表现出轻微或无代谢活性降低(图2b)。通过比较不同时间点的结果,可判断细胞毒性效应是即时发生还是出现在后期阶段。
