电子晶体学是进行高分辨率结构解析的有力工具。在适宜条件下,可溶性蛋白或膜蛋白等大分子能够生长形成高度有序的二维(2D)晶体。所获得的二维晶体质量对于后续通过二维图像处理实现的最终结构重构分辨率至关重要。多年来,脂单层已被用作支持层,以促进外周膜蛋白以及可溶性蛋白的二维结晶。该方法也可用于整合膜蛋白的二维结晶,但需要更广泛的实证研究,以确定促进蛋白向脂单层分配的去垢剂和透析条件。脂单层在气-水界面处形成,其中极性的脂质头部基团保持在水相中并处于水合状态,而非极性的酰基链尾部则伸入空气中,从而打破表面张力并使水面展平。头部基团所带电荷或特有的化学基团可对溶液中的蛋白质产生亲和力,促进其结合并形成二维阵列。新形成的含有二维阵列的脂单层可直接转移至电子显微镜(EM)载网上,该载网为覆碳铜网,用于捞取并支撑晶体阵列。在本研究中,我们描述了一种用于低温电子显微镜(cryo-EM)成像的脂单层方法。

在有利情况下,通过蛋白质的二维晶体或螺旋阵列进行电子衍射可达到亚纳米级分辨率1,2,3。特别值得关注的是在接近天然环境中的重组二维膜蛋白阵列或晶体1。由于晶体可作为信号放大器,增强特定空间频率下结构因子的强度,电子晶体学能够以高分辨率探测比单颗粒冷冻电镜更小的目标,例如小分子。当电子束穿过有序的二维蛋白质阵列时会发生衍射,根据图像平面在探测器上的记录位置不同,可生成衍射图样或晶格图像4。随后可提取并处理这些衍射强度,以重构晶体的二维投影结构。电子的散射截面大于X射线,其散射主要遵循卢瑟福模型,基于电子与分子中带电原子之间的库仑相互作用5。二维膜晶体的厚度通常小于100 nm,适合电子透射,且不会在样品内部发生动力学散射6。已有研究表明,电子晶体学是研究膜蛋白高分辨率结构信息以及脂质-蛋白相互作用的有力工具7,8,9,10,11,12,13,14,15,16,17

脂单层是由磷脂在气-水界面紧密排列形成的一层单分子脂质层6,可促进可溶性蛋白或外周膜蛋白形成二维阵列18。根据脂质的密度及其侧向压力,脂分子可在气-水界面形成有序的二维阵列,其酰基链伸向空气,亲水性头部基团则暴露于水相溶液中1,6,19。脂质头部基团可通过静电相互作用与蛋白质结合,也可经修饰提供亲和标签,以结合特定的蛋白质结构域。例如,DOGS-NTA-Ni(1,2-二油酰基-sn-甘油-3-[(N-(5-氨基-1-羧基戊基)亚氨基二乙酸)琥珀酰]2- Ni2+)常用于构建脂单层,以结合带有聚组氨酸标签的蛋白质20,21,22。此外,霍乱毒素B亚基可结合脂单层中神经节苷脂GM1的特定五糖结构,用于结构研究23,24。通过将蛋白质锚定在脂质头部基团上,脂单层有助于形成适用于高分辨率电子晶体学研究的薄型二维阵列。脂单层技术已应用于多种蛋白质的电子晶体学结构研究,例如链霉亲和素2,25、V型膜联蛋白26、霍乱毒素27E. coli DNA旋转酶B亚基28E. coli RNA聚合酶25,29,30、羧酶体外壳蛋白31,以及HIV-132和莫洛尼鼠白血病病毒33的衣壳蛋白。由于脂单层具有良好的稳定性与化学特性,目前已探索其在冷冻电镜成像样品制备中的多种应用34。然而,蛋白质阵列的形成仍需进行优化。

1. 聚四氟乙烯块的制备

  1. 使用耐化学腐蚀的聚四氟乙烯(PTFE)树脂制备特氟龙块。使用普通钻头在块体上钻孔,具体尺寸见图1所示。

2. 单层脂质制备

注意:预计操作时间:30–45 分钟

  1. 脂质储备液的制备
    1. 使用8.91 mL氯仿、0.99 mL甲醇和0.01 mL浓度为10 mg/mL的脂质,配制体积比为9:1(v/v)的氯仿/甲醇溶液,得到终浓度为0.01 mg/mL的脂质混合液。
  2. 聚四氟乙烯(Teflon)板的制备
    1. 将聚四氟乙烯板置于甲醇浴中超声处理5分钟。
    2. 用热水清洗15分钟。
    3. 用蒸馏水冲洗。
    4. 将聚四氟乙烯板放入干燥器中干燥30分钟,不使用时保持真空状态(图1)。

3. 在缓冲液储液池上形成脂单层

注意:预计操作时间:1.5 小时

  1. 在实验当天,将脂质混合物新鲜配制成氯仿/甲醇(9:1)溶液,浓度为 0.01 mg/mL。
  2. 将一张 1 型滤纸放入培养皿中,并将聚四氟乙烯(Teflon)块放置在滤纸上。
  3. 用 60 µL 缓冲液填充聚四氟乙烯板的各孔。
  4. 使用 Hamilton 注射器,将脂质混合物逐滴小心地加至缓冲液表面(理想情况下每滴 1 µL)。
  5. 用蒸馏水润湿滤纸,以维持培养皿内的湿度。
  6. 在室温下孵育 60 分钟。氯仿应完全挥发,并在缓冲液表面形成一层脂质单分子层。

4. 将电镜载网应用于脂单层

注意:预计操作时间:1.5 小时

  1. 将未经辉光放电处理的带孔碳膜电镜载网(碳膜面朝下)轻轻放置在每个缓冲液储液池的顶部。
  2. 小心地将蛋白质注入侧面加样孔中。初始实验建议使用约 1 µM 的终浓度蛋白,但应在正式实验前预先优化蛋白浓度。
  3. 在室温下孵育 60 分钟。
  4. 轻轻向侧面加样口注入约 20–30 µL 缓冲液。这将使载网上升至聚四氟乙烯(Teflon)块的液面之上。
  5. 立即用镊子夹起载网,并垂直于液滴方向将其提起。
  6. 制备单层样品用于负染或冷冻透射电镜(cryo-TEM)成像(图 2)。若蛋白质形成二维阵列,则图像的功率谱或傅里叶周期图会根据晶体对称性在特定空间频率处显示出衍射强度。若功率谱中未出现衍射斑点,则表明蛋白质不太可能形成二维晶体。
  7. 使用透射电子显微镜(TEM)检查电镜载网上脂单层的覆盖情况。低倍电子图像有助于识别被覆盖单层的形貌。通常,被单层覆盖区域的图像对比度略低于未覆盖区域。此外,计算得到的二维图像功率谱将有助于识别二维晶体所在位置。

沉积在透射电镜载网上的脂质单层无需染色即可在透射电子显微镜(TEM)下观察。通过有无样品区域之间的对比度差异,可识别脂质单层的存在。在电子束路径中无样品的区域因电子束穿过空孔时未发生散射而呈现更亮的照明,对比度较高;而覆盖脂质单层的区域局部对比度较低(图3)。

为了筛选二维单层结晶的条件,可使用负染电镜或冷冻电镜成像来观察样品形貌。当载物台倾斜时,若单层覆盖了载网孔洞,则可通过增强的边缘衬度进行观察。若形成晶体,图像的功率谱或周期图将显示出傅里叶衍射斑点。尺寸较小(< 200 nm)且随机取向的晶体可能在功率谱中表现为高强度的环状信号。若未形成晶体,则功率谱中不会出现傅里叶衍射斑点。

脂质单分子层是一种强大的工具,可促进生物大分子结构研究中大型二维晶体的生长。为了成功在气-水界面制备完整且无缺陷的脂质单分子层,强烈建议在实验当天新鲜配制脂质,因为脂质酰基链的氧化可能导致单分子层中分子排列紊乱,从而对最终晶体的形成产生不利影响。购买的粉末状脂质应使用氯仿:甲醇混合溶剂溶解,这种溶剂常用于溶解磷脂35。氯仿:甲醇溶剂可预先配制并储存于密封的玻璃小瓶中。当将溶解后的脂质滴加至气-水界面时,需等待氯仿:甲醇溶剂从缓冲液表面完全蒸发。在本实验中,确保脂质单分子层形成以及二维晶体生长所需的适当等待时间至关重要。通常,脂质单分子层及二维阵列的形成大约需要30至60分钟的孵育时间18
