利用组织培养幼苗制备和饲养无菌昆虫以研究叶甲虫宿主-肠道微生物互作

摘要

为了获得无菌昆虫,需对其卵表面进行消毒,孵化后的幼虫随后使用无菌叶片进行饲养。该方法为无菌昆虫的制备提供了一种高效途径,无需使用抗生素或开发人工饲料,也可应用于其他食叶昆虫。

摘要

昆虫肠道被多种细菌定植,这些细菌可显著影响宿主的生理特征。将特定菌株引入无菌昆虫体内,是验证肠道微生物功能并阐明肠道微生物与宿主相互作用机制的有力方法。施用抗生素或对虫卵表面进行灭菌是去除昆虫肠道细菌常用的两种方法。然而,除了抗生素可能对昆虫产生不良影响外,先前的研究表明,饲喂抗生素并不能完全清除肠道细菌。因此,通常采用无菌人工饲料来维持无菌昆虫,但这一过程繁琐且劳动密集,并不能完全模拟天然食物中的营养成分。本文所述为一种高效、简便的制备和维持叶甲无菌幼虫的实验方案。 关键词:昆虫肠道,肠道微生物,无菌昆虫,叶甲,无菌幼虫,人工饲料 (Plagiodera versicolora)。具体而言,对甲虫卵表面进行灭菌处理,随后使用无菌的杨树叶饲养无菌幼虫。昆虫的无菌状态通过依赖培养和非依赖培养的检测方法进一步验证。综上,通过结合卵表面消毒与无菌培养技术,建立了一种高效且便捷的获取无菌昆虫的方法 P. versicolora为其他食叶昆虫提供了一种易于转化应用的工具。

引言

与哺乳动物类似,昆虫的消化道是食物消化与吸收的腔室。大多数昆虫肠道内定植着多种共生细菌,这些细菌依赖宿主提供的营养在肠道中生长繁殖1。肠道共生菌群对昆虫的多种生理过程具有深远影响,包括食物消化与解毒2,3,4、营养与发育5,6,7、抵御病原体与寄生生物8,9,10,11、化学通讯12,13以及行为调控14,15。有趣的是,某些肠道微生物在特定条件下可转变为兼性致病菌,或被入侵的病原体所利用从而加重感染,表明肠道细菌在某些情况下可能对宿主有害16,17,18。此外,肠道细菌还可作为生物技术应用与害虫防治中的微生物资源。例如,从植食性和木食性昆虫中分离出的木质纤维素降解细菌已被用于植物细胞降解,以开发生物燃料19。通过释放经基因工程改造、可表达生物活性分子的肠道共生菌,是一种新颖且有前景的策略,可用于防控农业与林业害虫以及传播传染病的蚊虫19,20,21,该策略也可用于增强有益昆虫的适应能力22。因此,阐明一种肠道细菌在活体(in vivo)中的行为模式,被视为充分挖掘其功能并拓展其多领域应用的首要任务。

动物可携带1至 >肠道中的1000种共生微生物物种1因此,很难准确验证单个细菌分类群或其群落组装在动物体内的表现,以及特定功能是由宿主还是其微生物伙伴所驱动。因此,需要制备无菌幼虫,并通过单一或多种物种定植获得已知菌系昆虫,以研究细菌与昆虫之间的功能及其相互作用。23目前,施用抗生素混合物和对昆虫卵表面进行灭菌是去除肠道细菌的常用方法。14,24,25,26然而,抗生素饮食无法完全消除肠道细菌,并且对宿主昆虫的生理产生负面影响27,28因此,使用抗生素处理的昆虫可能会掩盖某些肠道细菌的真实功能。幸运的是,对卵进行表面灭菌可消除这一问题。23,29,对实验昆虫无影响或影响极小。此外,人工饲料无法完全模拟昆虫的天然食物,且人工饲料的开发过程成本高昂且耗费大量人力30,31.

柳叶甲(Plagiodera versicolora )(Laicharting)(鞘翅目:叶甲科)是一种广泛分布的食叶害虫,主要取食杨柳科植物,如柳树(Salix)和杨树(Populus L.)32,33。本研究以柳叶甲作为典型的食叶昆虫代表,建立了一种制备和培育无菌昆虫的实验方案。我们利用植物组织培养技术获得无菌杨树叶,用于从表面消毒的卵开始培育P. versicolora的无菌幼虫。通过依赖培养和非依赖培养的方法,验证了P. versicolora幼虫的无菌状态。该方案所维持的无菌昆虫比使用人工饲料饲养的昆虫更接近其自然状态。更重要的是,该方法操作简便且成本极低,有助于提高获取无菌昆虫的可行性,尤其适用于尚无成熟人工饲料体系的非模式昆虫,为今后昆虫肠道微生物互作研究提供了有力支持。

方案

1. 昆虫饲养

  1. 维持 的 P. versicolora 在27 °C、相对湿度70 ± 5%、光周期16小时光照/8小时黑暗的培养室条件下饲养种群。将昆虫置于底部打孔的塑料盒中,盒内铺有湿润的吸水纸并提供新鲜的杨树枝条作为食物。定期向吸水纸喷洒清水以保持湿度,每两天更换一次枝条。
  2. 羽化后将成虫隔离进行产卵,饲以嫩叶以获得更多的卵。
  3. 收集24小时内新产的卵。将卵置于湿润的吸水纸上孵化60小时,以获得无菌幼虫。
    ​注意:新产的卵约72小时后孵化。对卵表面进行消毒的最佳时间为孵化前一天;否则,成功孵化的卵数量将减少。

2. 无菌杨树培养

  1. 配制 Murashige 和 Skoog (MS) 培养基以及 1 mg/mL 的 α-萘乙酸 (NAA) 储备液(参见材料表)。
  2. 在生物安全操作台中,将 50 µL 的 1 mg/mL NAA 储备液加入 500 mL MS 培养基中,充分振荡混匀,每瓶组织培养容器倒入约 50 mL,静置待其凝固。
  3. 准备好解剖刀、酒精灯和镊子;在生物安全操作台内,用酒精灯火焰对解剖刀和镊子进行灭菌处理。
  4. 从生长约 1 个月的杨树幼苗(无菌组织培养苗)上切取带有顶端芽或侧芽的 3–4 cm 茎段,每瓶培养容器插入 1 至 2 段茎段。
  5. 将这些茎段置于培养室中,在 25 °C、光照强度为 50 ± 10 cd、光周期为 16 小时光照/8 小时黑暗的条件下培养约 30 天。使用无菌叶片饲喂无菌幼虫。

3. 卵表面消毒及无菌幼虫培养

  1. 将培养皿、毛笔、滤纸、蒸馏水以及含有LB(Luria-Bertani)琼脂培养基的培养皿进行高压灭菌。
  2. 将带有附着卵的叶片放入培养皿中,用镊子小心地将卵从叶片上取下,并转移至另一个培养皿中。
    注意:此步骤应极为小心,因为卵牢固地附着在叶片上。
  3. 用75%乙醇清洗这些卵8分钟,并用无菌水重复清洗四次。
  4. 将卵转移至LB琼脂培养基上,以保持湿度促进孵化。
    注意:使用LB琼脂培养基有助于验证消毒后的卵是否无菌。
  5. 将培养皿置于培养箱中,在24小时内等待卵孵化。
  6. 在生物安全柜中,于培养皿内铺三层湿润的滤纸,将无菌杨树叶片放置于滤纸上,收集幼虫并将其置于叶片上,用封口膜密封培养皿,并在培养箱中于27 °C、70 ± 5%相对湿度、16小时光照/8小时黑暗的光周期条件下培养。
    注意:组织培养的叶片较为脆弱,容易快速失水。因此,在将叶片转移至培养皿时,使用湿润的滤纸是必要的。
  7. 每两天更换一次叶片。
  8. 对于常规饲养组,将卵从叶片转移到含有湿润滤纸的培养皿中,并用无菌杨树叶片饲喂这些幼虫。

4. 通过依赖培养的检测方法验证无菌幼虫

  1. 从无菌组和常规饲养组中随机选取三只1st、2nd和3rd龄幼虫。
  2. 在体视显微镜下使用无菌剪刀和镊子解剖3rd龄幼虫,并将其肠道收集至微量离心管中;将完整的1st和2nd龄幼虫直接收集到离心管中。
    注意:由于1st和2nd龄幼虫体型过小,无法解剖,因此需整体收集,并保持其肠道完整。
  3. 向1.5 mL离心管中加入100 µL磷酸盐缓冲液和三颗钢珠,使用珠磨式均质器将组织研磨成匀浆。
  4. 取100 µL匀浆液加入LB琼脂培养基中,用无菌玻璃涂布棒涂布。
  5. 将平板置于37 °C培养24小时,观察细菌菌落生长情况。

5. 使用非培养依赖性检测方法验证无菌个体

  1. 使用DNA提取试剂盒提取组织的总DNA(组织来源于第4节)。
  2. 使用分光光度计在260 nm处测定DNA浓度。
  3. 使用通用16S rRNA引物通过PCR扩增细菌16S rRNA基因。反应体系包含18 µL 1.1x PCR预混液(见材料表)、0.5 µL 27F引物(5'-ACGGATACCTTGTTACGAC-3')、0.5 µL 1495R引物(5'-ACGGATACCTTGTTACGAC-3')和100 ng模板DNA。PCR反应条件设置为:95 °C预变性3 min;随后进行28个循环,每个循环为95 °C变性30 s、55 °C退火1 min、72 °C延伸1 min;最后72 °C终延伸10 min。PCR产物暂存于4 °C,待后续分析。
  4. 将PCR产物与核酸染料混合,使用1%琼脂糖凝胶在1x TAE缓冲液中进行电泳分析。以10 µL DNA Marker作为分子量参照。
  5. 使用紫外透射仪观察凝胶,寻找大小约为1,500 bp的目标条带。

结果

讨论

制备无菌幼虫并通过重新引入特定菌株获得无菌定植幼虫,是阐明宿主-微生物相互作用机制的有力方法。新孵化的幼虫主要通过两种途径获得肠道微生物群:来自母体的垂直传播,或来自兄弟个体及环境的水平获得34。前者可通过亲代将微生物污染至卵表面而实现35。因此,通过对昆虫卵表面进行灭菌来获得无菌幼虫具有很高的可行性27,28,29。使用多种抗生素混合处理是获得无菌昆虫的另一种方法,但存在若干缺点28。相比之下,对卵表面灭菌后辅以无菌饲料,是更优的无菌昆虫培育方法23,28,29。
