Cas9-mediated Cleavage

Cas9-mediated cleavage is a programmable DNA-cutting process in which the CRISPR-associated protein Cas9 uses a guide RNA to recognize and sever a complementary genetic sequence. After the guide RNA pairs with its target next to a compatible protospacer-adjacent motif (PAM), Cas9 activates its nuclease domains to cleave both DNA strands, producing a site-specific double-strand break. In biochemistry and molecular biology, this cleavage enables targeted gene disruption, sequence insertion, and genome editing through cellular DNA-repair pathways. Studying the reaction also clarifies guide-RNA recognition, nuclease specificity, and factors that influence editing efficiency and off-target activity.

The video demonstrates a method to assess caspase-mediated protein cleavage in virus-infected cells. Post-viral infection, caspases get activated and degrade various host cellular proteins and viral nucleoproteins while specific caspase inhibitors prevent this degradation, as confirmed by western blotting.
