In Vitro Gut Model

An in vitro gut model is a laboratory system that recreates selected physical, chemical, and biological features of the gastrointestinal tract outside the body, enabling controlled study of digestion and intestinal function. These models typically expose food components, drugs, or biomaterials to sequential conditions that mimic stomach and intestinal environments, including changes in pH, enzymes, transit, and, in some systems, gut microorganisms. In biochemistry, they help characterize nutrient breakdown, metabolite production, compound stability, and interactions between microbial communities and intestinal contents. By reducing experimental variability and supporting mechanistic analysis, in vitro gut models inform research on nutrition, drug delivery, microbiome function, and gastrointestinal disease.

We present procedures for demonstrating that ligands bind to the surface membrane of the cellulose-digesting protozoa in the gut of Formosan subterranean termites using fluorescent microscopy and that ligands coupled with lytic peptides kill these protozoa in vitro (anaerobic protozoa culture) and in vivo (injection into the termite hindgut).

This procedure was established to be used for developing advanced 3D hepatic cultures in vitro, which can provide a more physiologically relevant assessment of the genotoxic hazards associated with nanomaterial exposures over both an acute or long-term, repeated dose regimes.
