Protein-metabolite Interactions

Protein-metabolite interactions are molecular associations in which small-molecule metabolites bind proteins to influence their structure, activity, stability, or localization. Binding commonly occurs through complementary shape, charge, hydrogen bonding, and hydrophobic contacts at active sites or regulatory pockets, allowing metabolites to act as substrates, cofactors, inhibitors, or allosteric effectors. In biochemistry, these interactions help regulate enzyme activity and coordinate metabolic pathways in response to changing metabolite concentrations. Characterizing them with biochemical assays, structural methods, and metabolomics supports the study of cellular regulation, disease mechanisms, biomarker discovery, and the development of compounds that modulate protein function.

Begin with a bacterial biosensor culture suspended in a medium. These engineered bacteria produce a transcription factor that, upon binding to a specific fecal metabolite, activates a promoter associated with a fluorescent reporter gene.To this culture, add a filtered human fecal sample containing metabolites, such as lactate, that are involved in host-microbiome interactions.Dilute this mixture with a fresh medium to maintain an optimal reaction volume.Incubate without shaking to mimic the...

Protein interactions are at the heart of a cell's function. Calorimetric and spectroscopic techniques are commonly used to characterize them. Here we describe fluorescence anisotropy as a tool to study the interaction between the protein mutated in the Shwachman-Diamond Syndrome (SBDS) and the Elongation factor-like 1 GTPase (EFL1).
