Chemotactic Analysis

Chemotactic analysis is the quantitative study of how cells or microorganisms detect and move toward or away from chemical gradients, a behavior central to transport, signaling, and tissue organization. In a typical assay, a chemoattractant or chemorepellent forms a controlled concentration gradient, and cell movement is measured through migration distance, directionality, speed, or the number of cells crossing a membrane. Bioengineers use these measurements to evaluate immune-cell recruitment, bacterial navigation, wound healing, and engineered tissue responses. Combining imaging, microfluidic platforms, and computational modeling can reveal how molecular cues regulate migration and guide the design of biomaterials, biosensors, and cell-based therapies.

Described here is an experimental system combining traction microscopy with a microfluidic system and a demonstration of how to quantify the mechanics of collective migration under biochemical gradient.

Here we present a protocol to quantify the chemotactic activity of blood monocytes in mouse models, to assess the effects of nutritional, pharmacological and genetic interventions on blood monocyte and to characterize the blood monocytes derived macrophages in mouse models using monocyte-chemoattractant protein-1 (MCP-1)-loaded basement membrane-derived gel plugs.
