Double Network Hydrogel

Double network hydrogels are water-rich polymer materials formed from two interpenetrating networks, combining the hydration of conventional hydrogels with substantially improved strength and toughness for bioengineering. Typically, a rigid, highly crosslinked network dissipates mechanical energy through controlled bond rupture, while a softer, loosely crosslinked network maintains elasticity and prevents catastrophic failure under load. These properties allow double network hydrogels to better mimic the mechanical behavior of biological tissues and support applications in tissue scaffolds, cartilage substitutes, wound dressings, and controlled drug delivery. Their tunable composition also enables researchers to adjust stiffness, resilience, swelling, and degradation for specific biomedical environments.

This protocol outlines the implementation of image-guided, laser-based hydrogel degradation to fabricate vascular-derived, biomimetic microfluidic networks embedded in poly(ethylene glycol) diacrylate (PEGDA) hydrogels. These biomimetic microfluidic systems may be useful for tissue engineering applications, generation of in vitro disease models, and fabrication of advanced "on-a-chip" devices.

We present a protocol for the synthesis of RGD-functionalized hydrogels as devices for cell and drug delivery. The procedure involves copper catalyzed alkyne-azide cycloaddition (CuAAC) between alkyne-modified polyacrylic acid (PAA) and a RGD-azide derivative. The hydrogels are formed using microwave-assisted polycondensation and their physicochemical properties are investigated.
