Dynamic Flow Bioreactor

A dynamic flow bioreactor is a controlled culture system that continuously or periodically moves fluid around biological materials, creating conditions that better reproduce the physical environment of living tissues. Pumps or other flow mechanisms regulate medium circulation, while parameters such as flow rate, nutrient delivery, oxygenation, and shear stress influence cell behavior and tissue development. In bioengineering, these systems support three-dimensional cell culture, scaffold maturation, organ and tissue models, and bioprocess development. By improving mass transport and applying physiologically relevant mechanical cues, dynamic flow bioreactors can enhance tissue organization, functional performance, and the reproducibility of engineered biological products.

Operation of a Benchtop Bioreactor

Fermentors are used to increase culture yield and productivity of bioengineered cells. After screening multiple microbial or animal cell culture candidates in shake flasks, the next logical step is to increase the selected culture’s biomass with the fermentor. This video demonstrates the setup and operation of a typical benchtop bioreactor system.

Batch and Continuous Bioreactors

Bioreactors are used to grow organisms in large volumes, thereby enabling the production of mass quantities of the target product. These reactors can be batch reactors, which contain all of the components needed for cell growth, or continuous reactors, which have inlet and outlet ports allowing for the addition of fresh growth media and the removal of cell waste. This video presents batch and continuous reactors and demonstrates the use of bioreactors to grow bacteria in the laboratory.

A novel vocal fold bioreactor capable of delivering physiologically relevant, vibratory stimulation to cultured cells is constructed and characterized. This dynamic culture device, when combined with a fibrous poly(ε-caprolactone) scaffold, creates a vocal fold-mimetic environment that modulates the behaviors of mesenchymal stem cells.

Chondrogenesis from stem cells requires fine tuning the culture conditions. Here, we present a magnetic approach for condensing cells, an essential step to initiate chondrogenesis. In addition, we show that dynamic maturation in a bioreactor applies mechanical stimulation to the cellular constructs and enhances cartilaginous extracellular matrix production.
