Extracellular Matrix Scaffold

Extracellular matrix (ECM) scaffolds are three-dimensional biomaterial frameworks that support cell attachment, organization, and tissue formation, making them central to tissue engineering and regenerative medicine. They can be produced from natural matrix or engineered from synthetic materials, and their structure, composition, stiffness, and porosity provide biochemical and physical cues that guide cell adhesion, migration, proliferation, and differentiation. As cells remodel the scaffold through enzymatic degradation and new matrix deposition, the construct can gradually develop tissue-like properties. In bioengineering, ECM scaffolds support wound repair, disease modeling, and the development of engineered tissues while helping researchers study how material design influences regeneration.

This protocol describes decellularization of Sprague Dawley rat kidneys by antegrade perfusion of detergents through the vasculature, producing acellular renal extracellular matrices that serve as templates for repopulation with human renal epithelial cells. Recellularization and use of the resazurin perfusion assay to monitor growth is performed within specially-designed perfusion bioreactors.
