Spheroid Formation

Spheroid formation is the process of assembling cells into three-dimensional, tissue-like aggregates that better reproduce aspects of native tissue than flat cell cultures. Under nonadherent or low-attachment conditions, cells cluster through cell-cell adhesion, compact into spherical structures, and establish gradients of nutrients, oxygen, and signaling molecules. In bioengineering, spheroids provide models for studying cell behavior, tissue organization, disease progression, and responses to drugs or biomaterials. They also support research in tissue engineering and regenerative medicine by enabling controlled investigation of cell interactions, viability, differentiation, and the development of more physiologically relevant engineered tissues.

The uneven loss of medium from microtiter plates affects the reproducibility of uniform multicellular tumor spheroid formation. Improving culture conditions to reduce significant medium loss will improve the reproducibility of spheroid formation and the results of spheroid-based assays using the liquid-overlay technique.
