Horseradish Peroxidase

Horseradish peroxidase (HRP) is a heme-containing enzyme that catalyzes oxidation reactions and serves as a widely used reporter and biocatalyst in bioengineering. In the presence of hydrogen peroxide, HRP forms reactive intermediate states that oxidize suitable electron-donor substrates, producing measurable color, fluorescence, or chemiluminescence signals. This mechanism supports enzyme-linked immunosorbent assays, immunostaining, biosensors, and nucleic acid detection, where HRP converts molecular recognition into an observable output. Researchers also use HRP-mediated oxidation to crosslink phenol-functionalized polymers and proteins, enabling the formation of hydrogels and engineered biomaterials under mild aqueous conditions.

The present protocol describes procedures used to study and characterize cell wall-related enzymes, mainly β-1,3-glucanase and peroxidase, in wheat plants. Their activity levels increase during wheat-RWA interaction and are involved in the plant defense response through cell wall reinforcement, which deters aphid feeding.

RNA CISH Signal Detection: A Technique to Detect Chromogenic Signals During RNA In Situ Hybridization in Intact Tissue Specimens

This video presents the protocol for signal detection during RNA chromogenic in situ hybridization. This technique can be used to diagnose an active oncogenic infection visually in an intact tissue specimen.

ELISA-Based Method to Analyze Interactions between Bait and Prey Proteins

2025

This video describes an ELISA-based technique that detects protein-protein interactions between two proteins by measuring the binding of the second protein to the immobilized first protein. The bound second protein is detected using specific antibodies labeled with an enzyme, such as horseradish peroxidase.
