Monocyte Adhesion Assay

Monocyte adhesion assay is an in vitro method for measuring how readily monocytes attach to biological or engineered surfaces, providing a functional readout of cell-surface interactions and inflammatory activation. The assay exposes isolated or cultured monocytes to a substrate under controlled conditions, removes nonadherent cells, and quantifies those retained; adhesion reflects receptor-ligand binding, surface chemistry, extracellular-matrix cues, and incubation or flow conditions. In bioengineering, researchers apply the assay to biomaterials, vascular coatings, tissue-engineered interfaces, and microfluidic platforms to assess their capacity to promote or limit monocyte attachment. These results help evaluate biocompatibility and inflammatory potential, guiding the design of implants and cell-interactive devices.

This video demonstrates an in vitro assay to explore the immunomodulatory influence of mesenchymal stem cells (MSCs) on effector T cell proliferation. The assay includes co-culturing MSCs with PBMCs to induce the monocyte population to acquire an immunomodulatory phenotype, leading to the secretion of interleukin-10 (IL-10). Following incubation with activated CD4+ T cells, the induced monocytes effectively suppress T cell proliferation.

This flow adhesion assay provides a simple, high impact model of T cell-epithelial cell interactions. A syringe pump is used to generate shear stress, and confocal microscopy captures images for quantification. The goal of these studies is to effectively quantify T cell adhesion using flow conditions.

A method of quantitating neutrophil adhesion is reported. This method creates a dynamic flow environment similar to that encountered in a blood vessel. It allows the investigation of neutrophil adhesion to either purified adhesion molecules (ligand) or endothelial cell substrate (HUVEC) in a context similar to the in vivo environment with sheer stress.
