Immunocompromised Host

An immunocompromised host is a person or animal whose immune defenses are weakened, reducing the ability to control infections, respond to vaccines, or eliminate abnormal cells. Immunosuppression can result from diseases, inherited disorders, medications, or medical treatments that impair immune-cell development, signaling, antibody production, or inflammatory responses. In bioengineering, engineered tissues, organ-on-chip systems, biomaterials, and computational models can reproduce selected features of these altered immune environments for controlled study. Such platforms support research on host-pathogen interactions, implant compatibility, drug safety, and therapies designed to restore or strengthen immune function.

This protocol describes the isolation and dissociation of mouse medulloblastoma tissue, and subsequent allografting of the tumor cells into immunocompromised recipient mice in order to initiate secondary medulloblastoma.

Intrafemoral Injection of Human Hematopoietic Stem and Progenitor Cells into Immunocompromised Mice

2023

Intrafemoral injections allow for the engraftment of a small number of hematopoietic stem and progenitor cells (HSPCs), by placing the cells directly in the bone marrow cavity. Here we describe an experimental protocol of intrafemoral injection of human HSPCs into immunodeficient mice.

Extracting Host Cell Lysate from Infected Host Cells Cultured in a Permeable Membrane System

This video demonstrates a permeable membrane insert system to investigate the impact of bacterial toxins on host cells in a controlled and isolated manner. This approach allows for the selective exposure of the cells to the toxins while maintaining a physical separation that mimics the natural host-pathogen interaction.

Here, we present a protocol to investigate the host-tissue distribution, transmission mode, and effect on the host fitness of a densovirus within a lepidopteran species, the cotton bollworm. This protocol can also be used for studying the interaction between other orally-transmitted viruses and their insect hosts.
