Laser Excitation Method

Laser excitation method is a technique that uses focused, coherent light to stimulate a material or biological probe, enabling controlled optical measurements and manipulation. In bioengineering, a laser is selected to match the absorption or excitation spectrum of a fluorophore, chromophore, or light-sensitive protein. The absorbed photons raise electrons to higher energy states; when they relax, emitted fluorescence or another optical response can be detected, often with wavelength filtering and spatial focusing. This method supports fluorescence microscopy, biosensor development, cellular imaging, and engineered biological systems while improving signal localization and experimental control.

This protocol describes a simple preparation method for gold nanoparticle integrated photo-responsive liposomes with the commercially available materials. It also shows how to measure the microbubble cavitation process of the synthesized liposomes upon the treatment of pulsed laser.

Resting-state functional-connectivity MRI has identified abnormalities in patients with a wide range of neuropsychiatric disorders, including epilepsy due to malformations of cortical development. Transcranial Magnetic Stimulation in combination with EEG can demonstrate that patients with epilepsy have cortical hyperexcitability in regions with abnormal connectivity.

This method aims at locating vertical subsurface defects. Here, we couple a laser with a spatial light modulator and trigger its video input to heat a sample surface deterministically with two anti-phased modulated lines while acquiring highly resolved thermal images. The defect position is retrieved from evaluating thermal wave interference minima.
