Proflavin Contrast Agent

Proflavin contrast agent is a fluorescent acridine dye used to increase visual contrast in biological samples, making cellular and tissue structures easier to distinguish during imaging. In bioengineering, proflavin associates with nucleic acids, particularly by intercalating between stacked base pairs, and its fluorescence enables labeled regions to be detected against surrounding material. This contrast enhancement supports microscopy, live-cell visualization, and tissue imaging, helping researchers assess cell organization, morphology, and interactions with biomaterials. Because signal depends on dye distribution, concentration, illumination, and sample context, proflavin-based imaging provides a practical platform for evaluating engineered tissues and improving biological imaging strategies.

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.
