Stem Cell Growth

Stem cell growth is the controlled maintenance and expansion of stem cells while preserving their ability to self-renew and, in many cases, differentiate into specialized cell types. In bioengineering, researchers regulate culture conditions such as nutrient availability, growth-factor signaling, oxygen levels, substrate properties, and cell density to balance proliferation with maintenance of stem cell identity. These engineered environments support the production of cells for tissue engineering, regenerative medicine, disease modeling, and drug evaluation. Understanding how physical and biochemical cues influence stem cell behavior helps researchers design more reliable cultures, organoid systems, and cell-based therapies.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
