Whole Blood Processing

Whole blood processing is the controlled collection, separation, preparation, and preservation of blood and its cellular and liquid components for laboratory or clinical use. Depending on the intended product, anticoagulated blood is mixed, centrifuged to separate plasma, the buffy coat, and red blood cells, or passed through filters and transferred into defined containers under controlled conditions. In bioengineering, these steps provide standardized samples for biomaterial testing, blood-contacting device evaluation, cell isolation, and development of diagnostic assays. Careful control of temperature, timing, contamination, and component handling helps maintain sample integrity, improve reproducibility, and support reliable analysis of cellular, biochemical, and mechanical responses.

The preparing and processing of dried blood spots (DBS) for their final analysis are still poorly standardized for most diagnostic applications. To overcome this shortcoming, a comprehensive step-by-step protocol is suggested and subsequently evaluated with regard to its effectiveness for detecting markers of viral infections.

Repeated blood sampling is necessary and important in animal research. We developed a novel, non-lethal and reliable method for repeated blood collection from adult zebrafish, and applied this method to the study of blood biochemistry, including glucose metabolism.

Establishment of human models of the blood-brain barrier (BBB) can benefit research into brain conditions associated with BBB failure. We describe here an improved technique for preparation of a contact BBB model, which permits coculturing of human astrocytes and brain endothelial cells on the opposite sides of a porous membrane.
