Mdck Cell Infection

MDCK cell infection is the laboratory introduction and replication of viruses in Madin-Darby canine kidney cells, an epithelial cell line widely used to study viral biology. Researchers inoculate a confluent MDCK monolayer, allowing viral particles to attach to cell-surface receptors, enter cells, replicate their genomes, and produce progeny virions; infection may also cause observable cytopathic effects. This system supports virus isolation, propagation, titration, and analysis of host-cell responses under controlled conditions. In biological techniques, MDCK infection assays help characterize viral growth, compare infectivity, evaluate antiviral compounds, and generate experimental material for molecular and cellular studies.

Source: Chang, X., et al. Preparation of Pseudo-Typed H5 Avian Influenza Viruses with Calcium Phosphate Transfection Method and Measurement of Antibody Neutralizing Activity. J. Vis. Exp. (2021)This video demonstrates a luciferase-based pseudovirus titration assay to determine the optimal target cell density for maximum infection. By comparing luminescence across cell densities, the method ensures high assay sensitivity and reproducibility for downstream applications.

Assessing Caspase-Mediated Cleavage of Viral and Host Proteins in Virus-Infected Cells

The video demonstrates a method to assess caspase-mediated protein cleavage in virus-infected cells. Post-viral infection, caspases get activated and degrade various host cellular proteins and viral nucleoproteins while specific caspase inhibitors prevent this degradation, as confirmed by western blotting.

Influenza A viruses (IAVs) are important human respiratory pathogens. To understand the pathogenicity of IAVs and to perform preclinical testing of novel vaccine approaches, animal models mimicking human physiology are required. Here, we describe techniques to evaluate IAV pathogenesis, humoral responses and vaccine efficacy using a mouse model of infection.

Human infection by Entamoeba histolytica leads to amoebiasis, a major cause of diarrhea in tropical countries. Infection is initiated by pathogen interactions with intestinal epithelial cells, provoking the opening of cell-cell contacts and consequently diarrhea, sometimes followed by liver infection. This article provides a model to assess the early host-pathogen interactions to improve our understanding of amoebiasis pathogenesis.
