Adult Mosquito Collection

Adult mosquito collection is the systematic capture of mature mosquitoes to measure species presence, abundance, behavior, and seasonal activity in a defined area. Researchers use methods such as light or baited traps, resting collections, and aspirators, which exploit mosquito responses to carbon dioxide, host odors, light, or sheltered resting sites. Collected specimens can be identified by morphology or molecular tools and examined for blood-feeding status or pathogen infection. In biology, these data support population ecology, biodiversity studies, and disease surveillance, while helping evaluate mosquito-control strategies and identify changes in vector distributions.

A simplified yet accurate method to collect and stain mosquito hemocytes is described. Our method combines the simplicity of perfusion with the accuracy of high injection techniques to isolate clean preparations of hemocytes in Aedes mosquitoes. This method facilitates studies requiring knowledge of the types of hemocytes and their abundance.

Reverse genetic approaches have proven extremely useful for determining which genes underly resistance to vector pathogens in mosquitoes. This video protocol illustrates a method used by the Dimopoulos lab to inject dsRNA into Anopheles gambiae mosquitoes, which harbor the malaria parasite. The technique manipulating the injection setup and injecting dsRNA into the thorax is illustrated.

(2020)This video demonstrates the depletion of native gut microbiota in mosquitoes using an antibiotic-containing sucrose solution. This is followed by controlled recolonization with a bacterial suspension delivered through a membrane feeding system to establish a defined bacterial population in the mosquito gut.

Numerous novel virus-like sequences have been found in mosquitoes due to the extensive use of sequencing technologies. We provide an effective procedure for isolating and amplifying viruses using vertebrate and mosquito cell lines, which might serve as the basis for future studies on mosquito-associated viruses, including mosquito-borne and mosquito-specific viruses.
