Mouse Model

A mouse model is a living mouse used to represent human biology, disease, or a biological process under controlled experimental conditions. Researchers study naturally occurring traits or introduce targeted genetic changes, then monitor how altered genes, cells, tissues, and physiological systems produce measurable phenotypes. These models support investigations of gene function, development, immunity, cancer, and infectious disease, while enabling controlled testing of potential treatments and experimental procedures. Because mice share many conserved genes and biological pathways with humans, mouse models can connect molecular mechanisms to organism-level outcomes, although species differences require careful interpretation when translating findings to human health.

Mouse Models of Periventricular Leukomalacia

We established mouse models of periventricular leukomalacia (PVL), the predominant brain injury in premature infants characterized by periventricular white matter lesions. Hypoxia/ischemia with/without systemic infection are the primary causes of PVL. Unilateral carotid ligation and hypoxia exposure with/without lipopolysaccharide injection creates PVL-like lesions in P6 mice.

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.

Two techniques can be used to establish this model: injection of a cancer cell suspension into the cecal wall or transplantation of a piece of subcutaneous tumor onto the cecum. This model is useful for studying the natural progression of colorectal cancer and testing new therapeutic agents against colorectal cancer.

In vivo Imaging and Therapeutic Treatments in an Orthotopic Mouse Model of Ovarian Cancer

Orthotopic animal models of ovarian cancer replicate better human disease and therefore enhance our understanding of cancer progression and tumor response to therapy. A mouse model receives an intrabursal injection of luciferase-expressing ovarian tumor cells. Treatment is administered via oral gavage. Tumor growth is monitored by in vivo imaging system.
